imaging cleared embryonic and postnatal hearts at single-cell resolution

Einzel klonalen Verfolgung und Analyse auf Vollherzhöhe kann Herzvorläuferzellverhalten und Differenzierung während der Herzentwicklung, bestimmen und für die Untersuchung der zellulären und molekularen Grundlagen von normalen und abnormalen Herzmorphogenese ermöglichen. Neue aufkommende Technologien der retrospektiven Einzel klonalen Analysen machen die Untersuchung von Herz-Morphogenese auf Einzelzellauflösung möglich. Allerdings Gewebe Opazität und Lichtstreuung des Herzens als Abbildungstiefe von ganzem Herzen Bildgebung auf Einzelzellauflösung hinder erhöht. Um diese Hindernisse zu überwinden, eine ganze Embryo Clearing-System, das das Herz hoch transparent für beide Beleuchtung und Detektion machen können, müssen entwickelt werden. Zum Glück, in den letzten Jahren, viele Methoden für die ganze Organismus Clearingsystemen wie CLARITY, Sca le, SeeDB, ClearT, 3DISCO, kubisch, DBE, BABB und PACT berichtet. Dieses Labor ist daran interessiert, die zellulären und molekularen Mechanismen der KarteIAC Morphogenese. Vor kurzem haben wir einzelne Zelllinie über die ERT2 ROSA26-Cre Tracing; ROSA26-Confetti System zu dünn Label - Zellen während der Herzentwicklung. Wir passten mehrere ganze Embryo-Clearing - Methoden einschließlich Sca le und CUBIC (klar, ungehinderte Bildgebung des Gehirns Cocktails und Computeranalyse) mit whole mount Färbung Bild einzelne Klone im Inneren des Herzens , den Embryo in Kombination zu löschen. Das Herz wurde erfolgreich auf Einzelzell Auflösung abgebildet. Wir fanden , dass Sca le die embryonale Herz beseitigen können, aber die postnatale Herz nicht effektiv beseitigen können, während CUBIC die postnatale Herz beseitigen können, sondern schädigt die embryonale Herz durch das Gewebe auflöst. Die hier beschriebenen Methoden wird die Untersuchung der Genfunktion in einem einzigen Klon Auflösung während kardialen Morphogenese ermöglichen, was seinerseits die zellulären und molekularen Basis angeborener Herzfehler aufdecken kann.
