The following experimental protocol followed the guidelines of the Institutional Review Board of the Pennsylvania State University College of Medicine, and the human subject gave written informed consent before participating in the study.
Note: For the purpose of demonstration, a simple odor stimulation paradigm using a commercially available, MRI-compatible olfactometer is presented. This paradigm has proven effective in reducing the habituation effect and has produced reliable olfactory fMRI data15. Certain steps outlined in this protocol may be specific to the type of olfactometer used. However, any type of equipment&#8212;home-made or commercially available with similar capabilities&#8212;may be used in an analogous fashion. The olfactometer must be capable of monitoring respiration as well as presenting a sequence of odorants with precise timing. Make sure that the whole odor delivery system (including the olfactometer) is built with materials inert to odorant chemicals (e.g., glass and polytetrafluoroethylene), and the odor pathway is smooth and air-tight with minimal dead space.
1. Paradigm Design
<ol>
	<li>Create a new paradigm by specifying the airflow valve sequence on a programmable olfactometer. 
	NOTE: The valve sequence is the order and timing of the opening and closing of specific air channels that hold different concentrations or types of odorants. In this demonstration, each of the valves for the six channels was opened twice for a total of twelve odor deliveries. Whenever one valve was open, all the other valves were closed, and each valve was opened again only after all the other valves had already opened once.
	<ol>
		<li>Assign duration for the stimulus (the opening of a specific channel) as well as duration for the channel to be closed. 
		NOTE: In this demonstration, the duration for the odor presentation was 6 s, while the duration for the channels to be closed varied from 22 s to 38 s.</li>
		<li>Set the number of repetitions for the sequence of valve opens and closes. Here, the number of repetitions is 1.</li>
		<li>Interleave each odorant presentation with a presentation of odorless air at the same flow rate. For example, deliver the airflow to the subject with or without odor at a flow rate of 6 L/min in 50% relative humidity and room temperature at 22 &#176;C. 
		NOTE: This is important, as variations in airflow may cause tactile sensation.</li>
	</ol>

2. Odorant Preparation
<ol>
	<li>Choose a proper odorant for the odor stimulation paradigm by considering the odor valence, pleasantness, intensity, familiarity, and trigeminal component (see Table 1). 
	Note: Table 1 lists some commonly used odorants. Lavender odor was chosen for this demonstration because it has minimal trigeminal stimulation at low to moderate concentrations and is generally perceived as pleasant and familiar.</li>
	<li>Choose a proper solvent (e.g., water, mineral oil, 1,2-propanediol, ethanol) to prepare the odorant solutions. 
	NOTE: Here, 1,2-propanediol was used as the solvent for odorant solution preparation.</li>
	<li>Choose a proper odorant concentration for the odor stimulation paradigm. For example, dilute lavender oil in 1,2-propanediol at 0.10% (volume/volume) concentration for the olfactory stimulation19. 
	NOTE: This can be done by a psychophysical evaluation of a series of different concentrations by a group of normal subjects.</li>
	<li>Put the proper odorant solutions in the odorant containers. Ensure that all containers have the same amount of space, same amount of solution, and same surface area for the solution. For example, use six 300-mL size glass bottles as the odorant containers with each bottle holding 50 mL of 0.10% lavender oil solution.</li>
	<li>Connect all the odorant containers to the proper channels for odor delivery.</li>
</ol>
3. Olfactometer Set-up
<ol>
	<li>Check the connections to ensure that all odorant containers are properly attached to the odorant carrier. Do not overtighten, as this can damage the seal. Proper tightness is ensured in a later step by checking the airflow through each odorant container.</li>
	<li>Place the odorant carrier in the magnet room and connect each tube to the olfactometer outside of the room, as the main unit is not MR-compatible. Visually check for any kinks in the tubing, as this will affect airflow. The airflow of each channel will be checked in a later step.</li>
	<li>Securely connect all tubes from the olfactometer to the odorant carrier by matching the numbers to the correct ports. For accuracy, color-code the tubes, such that pink for channel 1, blue for channel 2, etc.</li>
	<li>Ensure the air flow through all the channels is consistent by attaching a flowmeter to the output end of the tubing. Manually open each channel on the control panel of the olfactometer, adjust the total air flow as well as flow rates of each channel and the flush line until the flow rate of each channel is consistent.</li>
	<li>Connect the face mask or nose piece to the odorant carrier with polytetrafluoroethylene (PTFE) tubing. Make sure that the air flow (e.g., 6 L/min) delivered to the subject is consistent when the channels are switched.</li>
	<li>Connect the radiofrequency trigger from the MRI system to the &#8220;trigger in&#8221; port on the olfactometer to synchronize the odor stimulation paradigm and fMRI image acquisition. An optical-electrical signal converter may be needed.</li>
	<li>Adjust the total air flow and flow rates for each channel and the flush line to the designed amounts. For example, a total air flow of 6 L/min and the flow rates for each channel and the flush line to be 3 L/min.</li>
	<li>Connect the pneumatic respiratory sensor belt to the response port of the olfactometer via the pneumatic-electrical signal converter box.</li>
	<li>If a subjective response is needed, connect the pneumatic response pad to the response port of the olfactometer via the pneumatic-electrical signal converter box.</li>
</ol>
4. Experimental Procedure
<ol>
	<li>Conduct a pre-screening to ensure that the MRI procedure is safe for the subject.
	<ol>
		<li>Ask the subject about medical history, including potential implants, claustrophobia, or other preexisting conditions that may interfere with the subject&#8217;s ability to safely participate in the fMRI study. Additionally, perform a smell threshold test of the odorants to ensure that the subject can smell the odorants during the experiment.</li>
	</ol>
	</li>
	<li>Have the subject lie supine on the MRI examination bed. Place the face mask or nose piece properly on the subject to ensure air blows into the nostrils. Place the respiratory sensor on either the chest or abdomen. Ask the subject to breathe normally. Manually adjust the tightness and placement of the belt holding the respiratory sensor according to the respiration pattern seen on the olfactometer display.</li>
	<li>Create a data folder to record the respiratory data in the olfactometer. Click &#34;file manager&#34;, enter the subject ID assigned to the current subject, then &#34;confirm&#34; the input.</li>
	<li>Use the &#34;paradigm check&#34; option to test the synchronization of odor delivery and inhalation without involving stimulus delivery, and if necessary, manually adjust the &#34;valve delay&#34; time to ensure the onset of odor delivery is synchronized with the subject&#39;s inhalation phase.</li>
	<li>Set the synchronization between odor stimulation and fMRI image acquisition by selecting &#8220;trigg-in&#8221; mode on the control unit of the olfactometer. 
	Note: This allows the odor stimulation paradigm to be started with an external trigger over the &#8220;trigger in&#8221; port originated from the MRI system. Thus, the paradigm will not run until the external trigger from the scanner is received. Please note what kind of trigger pulse (electrical or light) that the MRI scanner sends out. A signal converter may be needed to link the two systems.</li>
	<li>Activate the respiratory trigger by selecting &#8220;resp trigger start&#8221; on the control unit of the olfactometer. 
	Note: When activated, the start of each paradigm sequence element is synchronized with inhalation. This can be achieved empirically by delaying the odor delivery approximately half of a cycle of respiration from the beginning of the exhalation phase.</li>
	<li>Start the fMRI image acquisition on the MRI console; the odor stimulation paradigm will start as soon as image acquisition starts. Monitor the respiration pattern for any irregular respiratory activity. 
	Note: Irregular respiratory activity may be in the form of plateaus, wider and longer cycles, or erratic waves. Here, a BOLD signal-sensitive T2*-weighted echo-planar-imaging sequence was used for fMRI image acquisition with 2,000 ms repetition time, 30 ms echo time, 90&#176; flip angle, 220 mm &#215; 220 mm field of view, 80 &#215; 80 acquisition matrix, 30 4-mm-thick axial slices, and acceleration factor of 2 for integrated parallel imaging techniques.&#160;</li>
	<li>At the completion of the imaging protocol, move the subject out of the magnet and remove the face mask/nose piece.</li>
</ol>
5. Olfactometer Clean-up
<ol>
	<li>Power off the air pump. Detach the odorant containers from the odorant carrier and replace with clean, empty ones.</li>
	<li>Power on the air pump. Flush each channel with odorless air for 5 min to remove residual odorants in the air line.</li>
	<li>Shut down the olfactometer.</li>
	<li>Disinfect the nose piece or face mask with alcohol wipes. Rinse the face mask or nose piece with warm water and then air dry.</li>
</ol>
6. Data Analysis
<ol>
	<li>To process the data, load the respiration data file to the open-source software Olfactory Network Stimulation Editing Tool (ONSET) (www.pennstatehershey.org/web/nmrlab/resources/software/onset)15 
	Note: The ONSET software was developed by Xiaoyu Sun. Onsets of odor stimulation based on the timing of the paradigm and respiration trace will be automatically detected. The actual stimulation vector is defined as the start time of each effective inhalation during odor delivery.
	<ol>
		<li>Measure and compare the respiration rate and volume (the area under each inhalation and exhalation phase pair) between odor and odorless periods15. 
		NOTE: There should be no significant difference in these respiratory parameters between the odor and odorless periods.</li>
		<li>Process the fMRI data with the actual onset and duration vectors from ONSET for the activation of the central olfactory system15.</li>

The human olfactory system is understood to be much more than a sensory system because olfaction also plays an important role in homeostatic regulation and emotions. Clinically, the human olfactory system is known to be vulnerable to attacks of many prevalent neurological diseases and psychiatric disorders, such as Alzheimer&#39;s disease, Parkinson&#39;s disease, post-traumatic stress disorder, and depression1,2,3,4,5. Currently, functional magnetic resonance imaging (fMRI) with blood-oxygen-level-dependent (BOLD) contrast is the most valuable technique for mapping functions of the human brain. A significant amount of knowledge about specific functions of central olfactory structures (e.g., piriform cortex, orbitofrontal cortex, amygdala, and insular cortex) has been acquired with this technique6,7,8,9,10.
The application of fMRI to studies of the human central olfactory system and associated diseases, however, has been hindered by two major obstacles: rapid habituation of BOLD signal and variable modulation by respiration. In everyday life, when exposed to an odorant for a period of time, we quickly habituate to the scent. In fact, when studied using olfactory fMRI, the odor-induced fMRI signal is rapidly attenuated by habituation, which poses a challenge on stimulation paradigm designs8,10,11,12,13,14. The initial&#160;significant BOLD signal in the primary olfactory cortex only persists for several seconds after odorant onset. Therefore, olfactory fMRI paradigms should avoid prolonged or frequent odor stimulations in a short period of time. To reduce the habituation effect, some studies have attempted to present alternating odors in an fMRI paradigm. However, this approach may complicate data analysis since each odorant can be treated as an independent stimulation event.
Another technical issue arises with variability in subjects&#39; respiration patterns; inhalation does not always synchronize with odorant administration during a fixed-timing paradigm. The onset and duration of olfactory stimulation are modulated by each individual&#39;s respiration, which confounds fMRI data quality and analysis. Some studies have attempted to mitigate this problem with visual or auditory cues to synchronize breathing and odorant onset, but the compliance of subjects is variable, especially in the clinical population. The brain activations associated with these cues could also complicate data analysis in certain applications. Thus, synchronizing inhalation with odorant delivery can be crucial for olfactory fMRI studies15.
An additional consideration vital to olfactory fMRI, especially in the data analysis process, is odorant selection. Finding an appropriate odorant concentration with respect to perceived intensity is important for quantification and comparison of activation levels in the brain under various experimental conditions or diseases. Odorant selection must also take into consideration odor valence, or pleasantness. This is known to cause divergent temporal profiles in olfactory learning16,17. Lavender odor was chosen for this demonstration partially for this reason. Depending on the purpose of a specific study, different odorants may be better choices. In addition, trigeminal stimulation must be minimized to reduce activation not directly related to olfaction18.
In this report, we demonstrate an fMRI technique to set up and run a respiration-triggered paradigm using an olfactometer in the magnetic resonance environment. We also present a post-processing tool that can diminish some timing errors that may have occurred during data acquisition in an attempt to further improve data analysis.

The study of human olfaction is a highly complex and valuable field with applications ranging from biomedical research to clinical evaluation. Currently, evaluation of the functions of the human central olfactory system with functional magnetic resonance imaging (fMRI) is still a challenge because of several technical difficulties. There are some significant variables to take into account when considering an effective method for mapping the function of the central olfactory system using fMRI, including proper odorant selection, the interaction between odor presentation and respiration, and potential anticipation of or habituation to odorants. An event-related, respiration-triggered olfactory fMRI technique can accurately administer odorants to stimulate the olfactory system while minimizing potential interference. It can effectively capture the precise onsets of fMRI signals in the primary olfactory cortex using our data post-processing method. The technique presented here provides an efficient and practical means for generating reliable olfactory fMRI results. Such a technique can ultimately be applied in the clinical realm as a diagnostic tool for diseases associated with olfactory degeneration, including Alzheimer&#39;s and Parkinson&#39;s disease, as we begin to further understand the complexities of the human olfactory system.

Figure 1 demonstrates the set-up of olfactory fMRI inside and outside of the magnet room, taking into account MR-compatibility. Figure 2a demonstrates a standard fixed-timing paradigm, while Figure 2b demonstrates a paradigm where the &#34;respiration trigger&#34; allows for the synchronization of odor delivery and inhalation.

Combining Transcranial Magnetic Stimulation and fMRI to Examine the Default Mode Network

The default mode network is a group of brain regions that are active when an individual is not focused on the outside world and the brain is at "wakeful rest."1,2,3 It is thought the default mode network corresponds to self-referential or "internal mentation".2,3
It has been hypothesized that, in humans, activity within the default mode network is correlated with certain pathologies (for instance, hyper-activation has been linked to schizophrenia 4,5,6 and autism spectrum disorders 7 whilst hypo-activation of the network has been linked to Alzheimer's and other neurodegenerative diseases 8). As such, noninvasive modulation of this network may represent a potential therapeutic intervention for a number of neurological and psychiatric pathologies linked to abnormal network activation. One possible tool to effect this modulation is Transcranial Magnetic Stimulation: a non-invasive neurostimulatory and neuromodulatory technique that can transiently or lastingly modulate cortical excitability (either increasing or decreasing it) via the application of localized magnetic field pulses.9
In order to explore the default mode network's propensity towards and tolerance of modulation, we will be combining TMS (to the left inferior parietal lobe) with functional magnetic resonance imaging (fMRI). Through this article, we will examine the protocol and considerations necessary to successfully combine these two neuroscientific tools.

Simple and Computer-assisted Olfactory Testing for Mice

Olfaction is highly conserved among species and is required for reproduction and survival.
In humans, olfaction is also one of the senses that is affected with aging and is a strong predictor of neurodegenerative diseases. Thus, olfaction testing is used as a non-invasive diagnostic method to detect neurological deficits early on. In order to understand the mechanisms underlying olfactory network susceptibility, olfactory research in rodents has gained momentum in the past decade.
Here, we present a very simple, time efficient and reproducible olfactory testing method of innate odor perception and sensitivity in mice without the need of any prior food or water restriction. The tests are performed in a familiar environment to the mice, require only the scents and a 2 min session of odorant exposure. The analysis is performed, post-hoc, using computer-assisted commands on ImageJ and can be, therefore, carried out from start to end by one researcher.
This protocol does not require any special hardware or setup and is indicated for any laboratory interested in testing olfactory perception and sensitivity.

We present a simple and unbiased olfactory test in mice. With this protocol olfactory discrimination, preference, avoidance and sensitivity to a novel odor as compared to water can be assessed in single behavioral sessions. This method is indicated for a single experimenter and analysis is based on computer-assisted video processing.

Lentivirus-mediated Genetic Manipulation and Visualization of Olfactory Sensory Neurons in vivo

Development of a precise olfactory circuit relies on accurate projection of olfactory sensory neuron (OSN) axons to their synaptic 
targets in the olfactory bulb (OB). The molecular mechanisms of OSN axon growth and targeting are not well understood. Manipulating gene expression and subsequent 
visualizing of single OSN axons and their terminal arbor morphology have thus far been challenging. To study gene function at the single cell level within a specified 
time frame, we developed a lentiviral based technique to manipulate gene expression in OSNs in vivo. Lentiviral particles are delivered to OSNs by microinjection 
into the olfactory epithelium (OE). Expression cassettes are then permanently integrated into the genome of transduced OSNs. Green fluorescent protein expression 
identifies infected OSNs and outlines their entire morphology, including the axon terminal arbor. Due to the short turnaround time between microinjection and 
reporter detection, gene function studies can be focused within a very narrow period of development. With this method, we have detected GFP expression within as 
few as three days and as long as three months following injection. We have achieved both over-expression and shRNA mediated knock-down by lentiviral microinjection. 
This method provides detailed morphologies of OSN cell bodies and axons at the single cell level in vivo, and thus allows characterization of candidate gene function 
during olfactory development.

Correlating Behavioral Responses to fMRI Signals from Human Prefrontal Cortex: Examining Cognitive Processes Using Task Analysis

The aim of this methods paper is to describe how to implement a neuroimaging technique to examine complementary brain processes engaged by two similar tasks. Participants' behavior during task performance in an fMRI scanner can then be correlated to the brain activity using the blood-oxygen-level-dependent signal. We measure behavior to be able to sort correct trials, where the subject performed the task correctly and then be able to examine the brain signals related to correct performance. Conversely, if subjects do not perform the task correctly, and these trials are included in the same analysis with the correct trials we would introduce trials that were not only for correct performance. Thus, in many cases these errors can be used themselves to then correlate brain activity to them. We describe two complementary tasks that are used in our lab to examine the brain during suppression of an automatic responses: the stroop1 and anti-saccade tasks. The emotional stroop paradigm instructs participants to either report the superimposed emotional 'word' across the affective faces or the facial 'expressions' of the face stimuli1,2. When the word and the facial expression refer to different emotions, a conflict between what must be said and what is automatically read occurs. The participant has to resolve the conflict between two simultaneously competing processes of word reading and facial expression. Our urge to read out a word leads to strong 'stimulus-response (SR)' associations; hence inhibiting these strong SR's is difficult and participants are prone to making errors. Overcoming this conflict and directing attention away from the face or the word requires the subject to inhibit bottom up processes which typically directs attention to the more salient stimulus. Similarly, in the anti-saccade task3,4,5,6, where an instruction cue is used to direct only attention to a peripheral stimulus location but then the eye movement is made to the mirror opposite position. Yet again we measure behavior by recording the eye movements of participants which allows for the sorting of the behavioral responses into correct and error trials7 which then can be correlated to brain activity. Neuroimaging now allows researchers to measure different behaviors of correct and error trials that are indicative of different cognitive processes and pinpoint the different neural networks involved.

The goal of our research is to correlate behavior to brain activity. Accurate behavioral measures and imaging techniques allow us to elucidate brain-behavior relationships.

Imaging Odor-Evoked Activities in the Mouse Olfactory Bulb using Optical Reflectance and Autofluorescence Signals

In the brain, sensory stimulation activates distributed populations of neurons among functional modules which participate to the coding of the stimulus. Functional optical imaging techniques are advantageous to visualize the activation of these modules in sensory cortices with high spatial resolution. In this context, endogenous optical signals that arise from molecular mechanisms linked to neuroenergetics are valuable sources of contrast to record spatial maps of sensory stimuli over wide fields in the rodent brain.
Here, we present two techniques based on changes of endogenous optical properties of the brain tissue during activation. First the intrinsic optical signals (IOS) are produced by a local alteration in red light reflectance due to: (i) absorption by changes in blood oxygenation level and blood volume (ii) photon scattering. The use of in vivo IOS to record spatial maps started in the mid 1980's with the observation of optical maps of whisker barrels in the rat and the orientation columns in the cat visual cortex1. IOS imaging of the surface of the rodent main olfactory bulb (OB) in response to odorants was later demonstrated by Larry Katz's group2. The second approach relies on flavoprotein autofluorescence signals (FAS) due to changes in the redox state of these mitochondrial metabolic intermediates. More precisely, the technique is based on the green fluorescence due to oxidized state of flavoproteins when the tissue is excited with blue light. Although such signals were probably among the first fluorescent molecules recorded for the study of brain activity by the pioneer studies of Britton Chances and colleagues3, it was not until recently that they have been used for mapping of brain activation in vivo. FAS imaging was first applied to the somatosensory cortex in rodents in response to hindpaw stimulation by Katsuei Shibuki's group4.
The olfactory system is of central importance for the survival of the vast majority of living species because it allows efficient detection and identification of chemical substances in the environment (food, predators). The OB is the first relay of olfactory information processing in the brain. It receives afferent projections from the olfactory primary sensory neurons that detect volatile odorant molecules. Each sensory neuron expresses only one type of odorant receptor and neurons carrying the same type of receptor send their nerve processes to the same well-defined microregions of &tilde;100&mu;m3 constituted of discrete neuropil, the olfactory glomerulus (Fig. 1). In the last decade, IOS imaging has fostered the functional exploration of the OB5, 6, 7 which has become one of the most studied sensory structures. The mapping of OB activity with FAS imaging has not been performed yet.
Here, we show the successive steps of an efficient protocol for IOS and FAS imaging to map odor-evoked activities in the mouse OB.

Olfactory Context Dependent Memory: Direct Presentation of Odorants

Information is retrieved more effectively when the retrieval occurs in the same context as that in which the information was first encoded. This is termed context dependent memory (CDM). One category of cues that have been shown to effectively produce CDM effects are odors. However, it is unclear what the boundary conditions of these CDM effects are. In particular, given that olfaction has been called an implicit sense, it is possible that odors are only effective mnemonic cues when they are presented in the background. This assertion seems even more likely given that previous research has shown odors to be poor cues during paired associate memory tests, where odors are in the focus of attention as mnemonic cues for other information. In order to determine whether odors are only effective contextual mnemonic cues when presented outside the central focus of an observer, an olfactory CDM experiment was performed in which odorants were presented directly, rather than ambiently. Direct presentation was accomplished with the aid of an olfactometer. The olfactometer not only allows for direct presentation of odorants, but provides other methodological benefits, including the allowance of trial by trial manipulations of odorant presentations and, relatedly, time-specific releases of odorants. The presence of the same odor during both encoding and retrieval enhanced memory performance, regardless of whether the odor was presented ambiently or directly. This finding can serve as a basis for future olfactory CDM research which can utilize the benefits of direct presentation.

The use of an olfactometer for directly presenting odorants opens exciting opportunities for researchers of olfactory memory. The current paper discusses issues related to this methodology as related to a previously published experiment on olfactory context dependent memory.

Training Dogs for Awake, Unrestrained Functional Magnetic Resonance Imaging

We present a canine functional Magnetic Resonance Imaging (fMRI) training protocol that can be done in a cost-effective manner, with high-energy dogs, for acquisition of functional and structural data. This method of training dogs for awake, unrestrained fMRI employs a generalization procedure of stationing in several dissimilar locations to facilitate transfer of the stationing behavior to the real MRI scan environment; it does so without the need for extensive training time in the MRI scan environment, which can be expensive. Further, this method splits the training of a stationing (i.e., chin rest) behavior from desensitization to the MRI environment (i.e., 100+ decibel scan audio), the latter accomplished during dedicated Auditory Exposure conditioning sessions. The complete training and testing protocol required 14 hours and resulted in immediate transfer to novel locations. We also present examples of canine fMRI data that have been acquired from visual face processing and olfactory discrimination paradigms.

Magnetic resonance imaging (MRI) on unrestrained awake dogs is a new method with several advantages over imaging with physical or chemical restraint. Effect of Odor on Brain Excitability Assessed by a Breath-Synchronized Olfactometer

Combining a Breath-Synchronized Olfactometer with Brain Simulation to Study the Impact of Odors on Corticospinal Excitability and Effective Connectivity

It is widely accepted that olfactory stimulation elicits motor behaviors, such as approaching pleasant odorants and avoiding unpleasant ones, in animals and humans. Recently, studies using electroencephalography and transcranial magnetic stimulation (TMS) have demonstrated a strong link between processing in the olfactory system and activity in the motor cortex in humans. To better understand the interactions between the olfactory and the motor systems and to overcome some of the previous methodological limitations, we developed a new method combining an olfactometer that synchronizes the random order presentation of odorants with different hedonic values and the TMS (single- and dual-coil) triggering with nasal breathing phases. This method allows probing the modulations of corticospinal excitability and effective ipsilateral connectivity between the dorsolateral prefrontal cortex and the primary motor cortex that could occur during pleasant and unpleasant odor perception. The application of this method will allow for objectively discriminating the pleasantness value of an odorant in a given participant, indicating the biological impact of the odorant on brain effective connectivity and excitability. In addition, this could pave the way for clinical investigations in patients with neurological or neuropsychiatric disorders who may exhibit&#160;odor hedonic alterations and maladaptive approach-avoidance behaviors.

This paper describes using a breath-synchronized olfactometer to trigger single- and dual-coil transcranial magnetic stimulation (TMS) during odorant presentation synchronized to human nasal breathing. fMRI: Functional Magnetic Resonance Imaging

Functional magnetic resonance imaging (fMRI) is a non-invasive neuroimaging technique used to investigate human brain function and cognition in both healthy individuals and populations with abnormal brain states. Functional MRI utilizes a magnetic resonance signal to detect changes in blood flow that are coupled to neuronal activation when a specific task is performed. This is possible because hemoglobin within the blood has different magnetic properties depending on whether or not it is bound to oxygen. When a certain task is performed, there is an influx of oxygenated blood to brain regions responsible for that function, and this influx can then be detected with specific MRI scan&#160;parameters. This phenomenon is termed the blood oxygen level ependent (BOLD) effect, and can be used to create maps of brain activity.
This video begins with a brief overview of how MRI and fMRI signal is obtained. Then, basic experimental design is reviewed, which involves first setting up a stimulus presentation that is specifically designed to test the function that will be mapped. Next, key steps involved in performing the fMRI scan are introduced, including subject safety and setting up at the scanner. Commonly used steps for data processing are then presented, including pre-processing and statistical analysis with the general linear model. Finally, some specific applications of fMRI are reviewed, such as investigations into abnormal function in psychological disorders, and combining fMRI with complimentary imaging modalities, such as diffusion tensor imaging (DTI).
