Betydning IR er en av de viktigste årsakene til hud trykksår. De tidlige stadier (I og II) av trykksår beskrive tilstanden av den menneskelige hud (i forhold til de underliggende subkutane vev og muskler). Imidlertid er en forståelse av den immunologiske etiologi fortsatt mangler. Her presenterer vi en enkel og robust IR-modellen på mus øret huden for å løse dette gapet. Iskemi-reperfusjonsskade (IRI) oppstår når det er en forbigående hypoksi på grunn av obstruksjon av blodstrømmen (iskemi), etterfulgt av en etterfølgende ny oksygenering av vev (reperfusjon). I huden, er iskemi-reperfusjon (IR) antas å være en av de medvirkende faktorer til patofysiologien av trykksår, hvor langvarig sengeleie predisponerer langtidssykehuspasienter til skade. Hos disse pasientene er både hud og underliggende muskler stadig utsatt for vekt press over områder av bendel, som resulterer i lokaliserte skader som, hvis venstre ubehandlet, kan bli nekrotisk en. De som er involvert i en IRI skader er todelt. Under ischemi, okklusjon av blodkar fører til en drastisk nedgang av oksygentilførsel til vev. Dette resulterer i en reduksjon av ATP og pH, som inaktiverer ATPaser som er involvert i cellulær metabolisme. I sin tur, cellular kalsium nivåer pigg, og stresset eller skadet calen gjennomgå apoptose eller nekrose to. Utgivelsen av intracellulære innhold eller skader forbundet molekylære mønstre (damp), som HMGB1, bidrar til den inflammatoriske respons tre. Den andre fornærmelse oppstår under reperfusjon. Selv om oksygen og pH-verdier er gjenopprettet i løpet av reperfusjon, dette resulterer i dannelsen av reaktive oksygenarter (ROS), som fører til oksidasjon av intracellulære lipider, DNA og proteiner. Følgelig er pro-inflammatoriske mediatorer aktivert, noe som utløser en sekundær betennelsesreaksjon som innebærer rekruttering av immunceller til provoserende for 2.. Mens kaskade av biokjemiske hendelser som fører opp til den inflammatoriske responsen har vært godt beskrevet er den romlige og tidsmessige regulering av immuncelle aktiviteter ikke godt forstått. Her beskriver vi en robust IR-modellen på mus øret huden ved hjelp av enkel magnet klem. Sammen med multiphoton intra imaging (MP-IVM), vietablert en modell for å studere in vivo inflammatoriske responser som oppstår etter reperfusjon finner sted. Bakgrunnen for utvikling og bruk av denne teknikken er å prøve å forstå hvordan både interstitiell og infiltrere celler svare på IR i sanntid. Eksisterende modeller av IR hjelp av klem teknikk på huden flanke er meget invasiv, da de krever kirurgisk implantering av stålplater i huden flanke, noe som gjør dem mindre enn ideelle for immunologiske studier 4. En tilsvarende ikke-invasiv klem teknikken har blitt beskrevet i musehud flanke 5,6. Men på grunn av inkorporering av den intravital avbildning komponent i denne metoden, vi i stedet valgte øret huden som det målrettede IR-området, da den omgår bevegelser på grunn av pusting og gir stabilitet under avbilding 7,8. Videre leukocytter undergrupper som strekker interstitium er identiske mellom øret hud og hud flanke, selv omantall og andelen kan variere noe 9. Dermed representerer øret huden en ideell avbildning nettstedet. I tillegg har de fleste data som hentes fra disse IRI modellene er begrenset til makroskopiske evalueringer (gradering av magesår) og mikroskopiske analyser av endepunkt inflammatoriske indikatorer 10. Ved å bruke denne modellen, er sanntids visualisering av den cellulære responsen av neutrofiler etter reperfusjon i huden av et fluorescerende reporter mus aktivert. En tidligere utgitt intra øre bildeinformasjonen blir benyttet til 8 med ytterligere modifikasjoner (figurene 1, 2).

inducing ischemia-reperfusion injury in the mouse ear skin for intravital multiphoton imaging of immune responses

Ischemia-reperfusion injury (IRI) occurs when there is transient hypoxia due to the obstruction of blood flow (ischemia) followed by a subsequent re-oxygenation of the tissues (reperfusion). In the skin, ischemia-reperfusion (IR) is the main contributing factor to the pathophysiology of pressure ulcers. While the cascade of events leading up to the inflammatory response has been well studied, the spatial and temporal responses of the different subsets of immune cells to an IR injury are not well understood. Existing models of IR using the clamping technique on the skin flank are highly invasive and unsuitable for studying immune responses to injury, while similar non-invasive magnet clamping studies in the skin flank are less-than-ideal for intravital imaging studies. In this protocol, we describe a robust model of non-invasive IR developed on mouse ear skin, where we aim to visualize in real-time the cellular response of immune cells after reperfusion via multiphoton intravital imaging (MP-IVM).
