Het nucleaire porecomplex (NPC) controleert de invoer en uitvoer van grote moleculen in en uit de celkern. In tegenstelling tot kleine moleculen, die de nucleus zonder regulatie 1 kunnen binnengaan, wordt het transport van grotere moleculen sterk gecontroleerd door de NPC. Deze grote moleculen, zoals eiwitten en RNA, associëren met transportfactoren, waaronder importins en exportins, worden geïmporteerd en geëxporteerd vanuit de kern 2 . Om geïmporteerd te worden, moeten eiwitten een klein peptidemotief bevatten, in het algemeen een nucleair lokalisatiesignaal (NLS) genoemd, dat gebonden is door importins 2 . Deze sequenties van aminozuren fungeren als een label en zijn verschillend in samenstelling 3 , 4 . Proteïnen kunnen worden geëxporteerd van de kern naar het cytoplasma door hun associatie met exportinS, die een signaalsequentie binden, in het algemeen genaamd een nucleair uitvoer signaal (NES) 2 . Zowel importins als exportins zijn in staat om hun lading te transporteren door de regulering van het kleine Ras-gerelateerde nucleaire eiwit GTPase (Ran) 2 . Ran is gebleken te bestaan ​​in verschillende conformationalstaten, afhankelijk van of het aan GTP of het BBP is gebonden. Wanneer gebonden aan GTP, kan Ran binden aan importins of exportins. De meeste gevallen van ALS zijn geclassificeerd als sporadisch (SALS), zonder enige schijnbare genetische koppeling, maar 10% is overheersend geërfd (familiale ALS; FALS). Onlangs werden hexanucleotide herhalingsuitbreidingen (HREs) in het chromosoom 9 open leesframe 72 (C9orf72) gen geïdentificeerd 14 , 15 als een genetische oorzaak van ALS en FTD. Als gevolg hiervan is alleen ernstige nucleocytoplasmische transporttekorten geëvalueerd 11 , 17 , 18 . Dit protocol beschrijft een nieuw en veRij gevoelige test om de nucleocytoplasmatische transportdysfunctie in realtime te evalueren en te kwantificeren. De invoerhoeveelheid van een NLS-NES-GFP-eiwit (shuttle-GFP) kan in real-time worden gekwantificeerd met behulp van fluorescerende microscopie. Dit wordt gedaan met behulp van een exportinhibitor, zoals eerder beschreven 18 , waardoor de shuttle-GFP alleen in de kern komt. Met behulp van florescerende microscopie en een software die in staat is fluorescerende veranderingen in real-time te kwantificeren, is het mogelijk om geleidelijke veranderingen in de fluorescentie-intensiteit van de shuttle-GFP, die zich in de kern bevindt, te kwantificeren. Als gevolg hiervan kan een Michaelis-Menten-achtige verzadigingscurve van de fluorescentie-tot-tijd-as, die de hoeveelheid nucleaire shuttle-GFP op elk gewenst moment vertegenwoordigt, worden gemaakt. Door gebruik te maken van de initiële lineaire snelheid van de krommen, is het mogelijk om een ​​helling te genereren, die de snelheid van toegang tot de kern voor fluorescentie verzadiging vertegenwoordigt.
