Bakteriocine sind antimikrobielle Peptide, die durch einen breiten Bereich von Bakterienarten 1 , 2 hergestellt werden . Diese Verbindungen und ihre Produzenten, insbesondere LAB, wurden seit Jahrzehnten weltweit für ihre Einsatzmöglichkeiten in der Lebensmittelkonservierung und Medizin erforscht und ausgenutzt 3 . Mehrere Bakteriocine sind bekannt, um wichtige Pathogene, einschließlich Arten von Listeria, Enterococcus, Staphylococcus und Bacillus zu töten. Einige Bakteriocine haben sogar die Fähigkeit, die Immunantwort zu modulieren 4 . Viele Bakteriocine haben relativ schmale Spektren, eine Eigenschaft, die in einigen Anwendungen sehr geschätzt wird. Bakteriocine sind auch für den prophylaktischen oder probiotischen Gebrauch sehr attraktiv, da sie das (Aus-) Wachstum von Pathogenen, Pathobionten oder opportunistischen Bakterien, die die Darmhomöostase 6 , 7 ausgleichen können, unterdrücken können. In ihrer Natur und ihren physikochemischen Eigenschaften sind Bakteriocine sehr unterschiedlich, da sie unterschiedliche Strukturen, Zielspezifitäten, Wirkungsweisen usw. aufweisen. Die meisten Bakteriocine wurden in vitro- Einstellungen sehr genau untersucht, aber nur wenige wurden in Lebensmitteln getestet Matrizen 8 , 9 oder in vivo, wie in einem Tierdarm 6 , 10 . Die in vitro Eigenschaften können sich in hohem Maße unterscheiden, wenn sie in vivo aufgrund der Komplexität o bewertet werdenF die Darmumgebung und auch zu mutmaßlichen unbeabsichtigten Wirkungen auf nützliche Bakterien. Die meisten Probiotika sind LAB. Sie produzieren eine Reihe von anderen Metaboliten, einschließlich kurzkettiger Fettsäuren, von denen bekannt ist, dass sie die Physiologie des Wirts beeinflussen, sowie antimikrobielle Eigenschaften gegenüber bestimmten Bakterien zu demonstrieren. Daher ist es im Falle von probiotischen Stämmen, die Bakteriocine produzieren, am besten, um realistische Assays, wie gesunde Tiere mit normaler Mikrobiota, zu etablieren. In der vorliegenden Studie stellen wir eine Strategie zur Verfügung, die die Beurteilung der Wirkung verschiedener Bakteriocin-produzierender Stämme ermöglicht, deren Bakteriocine bei gesunden Mäusen unterschiedliche Hemmungsspektren aufweisen. Unsere Strategie umfasst die Fütterung von Mäusen mit isogenen Nicht-Bakteriocin-Mutanten, die die Differenzierung von Bakteriocin-vermittelten Effekten aus Nicht-Bakteriocin-vermittelten Effekten ermöglichen. Sequenzierung 16S rDNA erlaubt es, die dynamischen Veränderungen der Bakterienpopulation im Darm zu verfolgen. SubsGleiche statistische Analyse entschlüsselt Korrelationen zwischen Bakterienarten und auch zwischen Bakterienarten und gemessenen physiologischen Parametern ( zB Körpergewicht, serumbiochemische Parameter usw. ). Wir glauben, dass das in dieser Studie vorgestellte Protokoll auch auf andere probiotische oder präbiotische Anwendungen anwendbar ist, die über die Untersuchung von Bakteriocinen in lebenden Tieren hinausgehen.

Änderungen der Mikrobiota-Zusammensetzung während der Bakteriocin-Behandlung werden unter Verwendung einer kompositorischen 16S-rDNA-Sequenzierung überwacht. Unser Ansatz nutzt sowohl die Bakteriocin-Produzenten als auch ihre isogenen Nicht-Bakteriocin-produzierenden Mutanten, wobei letztere die Fähigkeit gibt, Bakteriocin-bezogene von nicht-bakteriocinbezogenen Modifikationen der Mikrobiota zu unterscheiden. Die Fäkal-DNA-Extraktion und die 16S-rDNA-Sequenzierungsmethoden sind konsistent und bilden zusammen mit der Bioinformatik ein mächtiges Verfahren, um schwache Veränderungen in den Bakterienprofilen zu finden und Korrelationen hinsichtlich der Cholesterin- und Triglyceridkonzentration zwischen Bakterienpopulationen und Gesundheitsmarkern herzustellen. Unser Protokoll ist generisch und kann so verwendet werden, um andere Verbindungen oder Nährstoffe mit dem Potential zu untersuchen, das Wirtsmikro zu verändernRobiota Zusammensetzung, entweder beim Studium der Toxizität oder nützliche Auswirkungen.
