Bakteriociner er antimikrobielle peptider produsert av et bredt spekter av bakteriearter 1 , 2 . Disse forbindelsene og deres produsenter, spesielt LAB, har blitt utforsket og utnyttet verden over i flere tiår for deres potensielle bruksområder i matvern og medisin 3 . Flere bakteriociner er kjent for å drepe viktige patogener, inkludert arter av Listeria, Enterococcus, Staphylococcus og Bacillus . Noen bakteriociner har selv muligheten til å modulere immunresponsen 4 . Mange bakteriociner har relativt smale spektra, en egenskap som er mye verdsatt i enkelte applikasjoner. For eksempel kan noen smalspektrede bakteriociner brukes til å lede spesifikk aktivitet mot utvalgte grupper av problematiske bakterier, uten mye forstyrrelse på commensal eller gunstig flora som deler samme nisje; Dette er spesielt viktig i tarmenOnment, hvor mange gunstige mikrober trives på en interaktiv og dynamisk måte 5 . Bakteriociner er også svært attraktive for profylaktisk eller probiotisk bruk, da de kan undertrykke (ut) vekst av patogener, patobioner eller opportunistiske bakterier som kan ubalanse tarmhemostasen 6 , 7 . Når det gjelder deres natur og fysisk-kjemiske egenskaper, er bakteriociner svært forskjellige, da de har forskjellige strukturer, målspesifikasjoner, virkemåter etc. De fleste bakteriociner er studert i stor detalj i in vitro- innstillinger, men svært få er testet i mat Matriser 8 , 9 eller in vivo, som for eksempel i en dyregutt 6 , 10 . In vitro- egenskapene kan variere i stor grad når de vurderes in vivo på grunn av kompleksiteten oF tarmmiljøet og også til påståelige utilsiktede effekter på gunstige bakterier. De fleste probiotika er LAB. De produserer en rekke andre metabolitter, inkludert kortkjedede fettsyrer, som er kjent for å påvirke vertenes fysiologi, samt å demonstrere antimikrobielle egenskaper mot visse bakterier. Derfor, når det gjelder probiotiske stammer som produserer bakteriociner, er det best å etablere realistiske analyser, for eksempel sunne dyr med normal mikrobiota. I den foreliggende studien gir vi en strategi som muliggjør vurdering av effekten av forskjellige bakteriocin-produserende stammer, hvis bakteriociner har forskjellige hemmende spektra på friske mus. Vår strategi inkluderer fôring av mus med isogene ikke-bakteriocinmutanter, som muliggjør differensiering av bakteriocin-medierte effekter fra ikke-bakteriocin-medierte effekter. Sequencing 16S rDNA muliggjør å følge de dynamiske endringene av bakteriepopulationen i tarmen. SubsEkvivalent statistisk analyse avgir korrelasjoner mellom bakteriearter og også mellom bakteriearter og målte fysiologiske parametere ( f.eks. Kroppsvekt, serumbiokjemiske parametere, etc. ). Vi tror at protokollen som presenteres i denne studien, også gjelder for andre probiotiske eller prebiotiske anvendelser utover studiet av bakteriociner hos levende dyr.

Svært spennende spørsmål oppstår med vår fremvoksende kunnskap om tarmmikrobiotasammensetningen og forholdet til helse, spesielt knyttet til forhold som bidrar til å opprettholde befolkningsbalansen. Det er imidlertid begrenset tilgjengelige metoder for å evaluere disse faktorene. Bakteriociner er antimikrobielle peptider produsert av mange bakterier som kan gi en konkurransefortrinn for matinnkjøp og / eller nisjestifting. Mange probiotiske melkesyrebakterier (LAB) -stammer har stort potensiale for å fremme menneskers og dyrs helse ved å hindre veksten av patogener. De kan også brukes til immunmodulasjon, da de produserer bakteriociner. Den antagonistiske aktiviteten til bakteriociner bestemmes imidlertid normalt av laboratorie bioassays under veldefinerte, men overforenklede forhold sammenlignet med det komplekse tarmmiljøet i mennesker og dyr, hvor bakterier står overfor multifaktoriale påvirkninger fra verten og hundrevis av mikrobielle arter sHaring samme nisje. Dette arbeidet beskriver en komplett og effektiv prosedyre for å vurdere effekten Av en rekke bakteriociner med forskjellige mål-spesifisiteter i et murint system. Endringer i mikrobiotasammensetningen under bakteriocinbehandlingen overvåkes ved anvendelse av komposisjonell 16S rDNA-sekvensering. Vår tilnærming bruker både bakteriocinprodusentene og deres isogene ikke-bakteriocin-produserende mutanter, sistnevnte gir muligheten til å skille bakteriocin-relatert fra ikke-bakteriocin-relaterte modifikasjoner av mikrobiota. Fekal DNA-ekstraksjonen og 16S rDNA-sekvenseringsmetodene er konsistente og sammen med bioinformatikk utgjør en kraftig prosedyre for å finne svake forandringer i bakterieprofilene og å etablere korrelasjoner, når det gjelder kolesterol og triglyseridkonsentrasjon, mellom bakteriepopulasjoner og helsemarkører. Produksjonen av bakteriociner har vært ansett som en positiv probiotisk egenskap i LAB, da det antas å forhindre vekst av opportunistiske bakterier og patogener.
