Bakteriociner är antimikrobiella peptider framställda av ett brett spektrum av bakteriearter 1 , 2 . Dessa föreningar och deras producenter, särskilt LAB, har utforskats och utnyttjats över hela världen i årtionden för deras potentiella tillämpningar i livsmedelsförvaring och medicin 3 . Flera bakteriociner är kända att döda viktiga patogener, inklusive arter av Listeria, Enterococcus, Staphylococcus och Bacillus . Vissa bakteriociner har även förmågan att modulera immunsvaret 4 . Många bakteriociner har relativt snäva spektra, en egenskap som är mycket uppskattad i vissa applikationer. Till exempel kan vissa smala spektrum bakteriociner användas för att rikta specifik aktivitet mot utvalda grupper av problematiska bakterier utan stor störning på kommensal eller fördelaktig flora som delar samma nisch. Detta är särskilt viktigt i tarmkanalenOnment, där många positiva mikrober trivs på ett interaktivt och dynamiskt sätt 5 . Bakteriociner är också mycket attraktiva för profylaktisk eller probiotisk användning, eftersom de kan undertrycka (ut) tillväxten av patogener, patobioner eller opportunistiska bakterier som kan obalansera tarmhemostasen 6 , 7 . Med avseende på deras natur och fysikalisk-kemiska egenskaper är bakteriociner mycket olika, eftersom de har olika strukturer, målspecificiteter, verkningsmetoder etc. De flesta bakteriociner har studerats i stor detalj i in vitro- inställningar, men väldigt få har testats i mat Matriser 8 , 9 eller in vivo, såsom i ett djurkärl 6 , 10 . In vitro- egenskaperna kan i stor utsträckning skilja sig in vivo på grund av komplexiteten oF tarmmiljön och även på förmodade oavsiktliga effekter på fördelaktiga bakterier. De flesta probiotika är LAB. De producerar en rad andra metaboliter, inklusive kortkedjiga fettsyror, vilka är kända för att påverka värdens fysiologi, liksom att visa antimikrobiella egenskaper mot vissa bakterier. Därför är det bäst att fastställa realistiska analyser, såsom friska djur med normal mikrobiota, när det gäller probiotiska stammar som producerar bakteriociner. I den föreliggande studien tillhandahåller vi en strategi som möjliggör bedömning av effekten av olika bakteriocinproducerande stammar, vars bakteriociner har olika hämmande spektra på friska möss. Vår strategi innefattar att mata möss med isogena icke-bakteriocinmutanter, vilket möjliggör differentiering av bakteriocinmedierade effekter från icke-bakteriocinmedierade effekter. Sekvensering 16S rDNA möjliggör att följa de dynamiska förändringarna av bakteriepopulationen i tarmen. SubsEkvivalent statistisk analys avkänner korrelationer mellan bakteriearter och även mellan bakteriearter och uppmätta fysiologiska parametrar ( t.ex. kroppsvikt, serumbiokemiska parametrar etc. ). Vi anser att protokollet som presenteras i denna studie också är tillämpligt på andra probiotiska eller prebiotiska tillämpningar bortom studier av bakteriociner hos levande djur.

Mycket spännande frågor uppstår med vår framväxande kunskap om gutmikrobiotasammansättning och förhållandet till hälsa, särskilt vad gäller de faktorer som bidrar till att bibehålla befolkningsbalansen. Det finns dock begränsade tillgängliga metoder för att utvärdera dessa faktorer. Bakteriociner är antimikrobiella peptider som produceras av många bakterier som kan ge en konkurrensfördel för matförvärv och / eller nischinrättning. Många probiotiska mjölksyrabakterier (LAB) -stammar har stor potential för att främja människors och djurs hälsa genom att förhindra tillväxten av patogener. De kan också användas för immunmodulering, eftersom de producerar bakteriociner. Den antagonistiska aktiviteten hos bakteriociner bestäms emellertid normalt av laboratorie bioassays under väldefinierade men överförenkladda betingelser jämfört med den komplexa tarmmiljön hos människor och djur, där bakterier står inför multifaktoriella influenser från värden och hundratals mikrobiella arter sHar samma nisch. I detta arbete beskrivs ett komplett och effektivt förfarande för att utvärdera effekten Av en mängd olika bakteriociner med olika målspecificiteter i ett murinsystem. Förändringar i mikrobiotasammansättningen under bakteriocinbehandlingen övervakas med användning av kompositionell 16S-rDNA-sekvensering. Vårt tillvägagångssätt använder både bakteriocinproducenterna och deras isogena icke-bakteriocinproducerande mutanter, vilka senare ger möjlighet att skilja bakteriocinrelaterade från icke-bakteriocinrelaterade modifieringar av mikrobiotan. Fekal DNA-extraktion och 16S rDNA-sekvenseringsmetoder är konsekventa och tillsammans med bioinformatiken utgör ett kraftfullt förfarande för att finna svaga förändringar i bakterieprofilerna och för att fastställa korrelationer i termer av kolesterol och triglyceridkoncentration mellan bakteriepopulationer och hälsomarkörer. Vårt protokoll är generiskt och kan därför användas för att studera andra föreningar eller näringsämnen med potential att ändra värdmikrofonenRobiota-kompositionen, antingen vid studier av toxicitet eller fördelaktiga effekter.

Produktionen av bakteriociner har ansetts vara en positiv probiotisk egenskap hos LAB, eftersom det antogs hindra tillväxten av opportunistiska bakterier och patogener. Syftet med detta arbete var att visa bakteriocins förmåga att modulera tarmmikrobiotapopulationer i en musmodell.
