Den vaskulære endotel består af en éncellelag i endothelial celler (EF) som linje lumen af blodkar. I en fysiologisk tilstand, inaktiv EF er ansvarlig for opretholdelsen af en antikoagulans og anti-inflammatoriske miljø. 1 dette er medieret af udtryk af antikoagulans og anti-inflammatoriske proteiner på EF-overflade. For eksempel, EF aktivering skyldes iskæmi/reperfusion skader eller vaskulær afvisning af (xeno-) transplanteres organer resulterer i en ændring i endothelial overfladen fra en antikoagulans og anti-inflammatorisk tilstand til en Pro koagulant og pro-inflammatoriske stat. 1
For at studere den fascinerende og komplekse samspil mellem endotel og koagulation faktorer, in vitro- modeller, som efterligner som muligt i vivo situation er særdeles ønskværdige. En fælles begrænsning, der præger konventionelle in vitro- koagulation assays er brugen af antikoaguleret blod, hvilket gør analysen af koagulation-medierede effekter besværlig og endda recalcification værdier fuldblod kan ikke reproducere resultater fås med frisk ikke-antikoaguleret blod. 2 desuden i traditionelle flat-bed celle kultur systemer er det umuligt at udnytte endotelet antikoagulerende egenskaber som en tilstrækkelig endotel celleoverfladen pr. blodvolumen ikke kan nås. Modellen præsenteres her overvinder disse begrænsninger ved dyrkning af EF på overfladen af sfæriske microcarrier perler, så en EF overflade-til-blod forholdet mellem &#62; 16 cm2/mL kan nås, som ligner situationen i små arterioler eller vener, og som blev beskrevet for at være tilstrækkelige til, at "naturlige" antikoagulation af blodet af EF overflade. 3 , 4 fuldblod kan bruges uden tilsat antikoagulantia i denne indstilling. Blodprøver kan indsamles under eksperimentet og cytokiner, koagulationsfaktorer og opløselige komplement aktivering markører kan påvises og kvantificeres. Desuden kan EF-belagt microcarrier perler analyseres for supplement og immunoglobulin aflejring samt udtryk for EF aktivering markører af Konfokal mikroskopi. En anden interessant program omfatter test af lægemidler, som formodes for at forhindre endotel celle aktivering og dermed koagulation. 5 selv om denne model ikke kan helt erstatte dyreforsøg, det tilbyder en metode til at teste specifikke funktionelle hypoteser ex vivo ved hjælp af celler og dermed mindske antallet af dyr, der bruges i grundforskning på iskæmi/reperfusion skade eller (xeno) transplantation.
Den beskrevne model blev brugt til at efterligne en xenotransplantation indstilling i hvilke svin aorta endothelial celler (PAEC) er vokset på microcarrier perler og inkuberes med hele, ikke-antikoaguleret blod. Forskellige transgene PAEC, bærer flere menneskelige gener såsom CD46 til regulering af komplementsystemet og/eller thrombomodulin (hTBM) for regulering af koagulation system, blev analyseret for deres antikoagulerende egenskaber. Endotel celle aktivering, supplement og koagulation systemer er stramt kontrolleret og indbyrdes forbundne. 6 det er derfor vigtigt at forstå, hvordan de forskellige transgene celler opfører sig efter udsættelse for humant blod vedhæftning molekyle udtryk og cytokin udgivelse, afgivelse af glycocalyx og tab af antikoagulerende proteiner. 7

In vivo, endotelceller er afgørende for den naturlige antikoagulation af cirkulerende blod. Derfor fører endotel celle aktivering til blodets koagulation. Dette fænomen er observeret i mange kliniske situationer, som organtransplantation i overværelse af præformerede anti-donor antistoffer, herunder xenotransplantation, samt i iskæmi/reperfusion skade. For at reducere dyreforsøg ifølge 3R standarder (reduktion, udskiftning og raffinement), in vitro- modellerne til at studere effekten i endothelial celle ville aktivering på blodets koagulation være ønskeligt. Dog giver fælles flatbed systemer i endothelial cellekultur et overflade-til-volumen forhold på 1-5 cm2 af endotel pr. mL blod, der ikke er tilstrækkelig for naturlige, endotel-medieret antikoagulation. Dyrkning endotelceller på microcarrier perler kan forhøje overflade-til-volumen forhold til 40-160 cm2/mL. Denne øgede ratio er tilstrækkelig til at sikre den "naturlige" antikoagulation af fuldblod, således at brugen af antikoagulantia kan undgås. Her er en in vitro- microcarrier-baseret system beskrevet at studere virkningerne af genetisk modifikation af svin endotelceller på koagulation af hele, ikke-antikoaguleret blod. I den beskrevne assay, primære svin aorta endotelceller, enten vildtype (WT) eller transgene for menneskelige CD46 og thrombomodulin, var vokset på microcarrier perler og derefter udsat for frisk tegnede ikke-antikoaguleret blod. Denne model giver mulighed for måling og kvantificering af cytokin frigivelse samt aktivering markører for supplement og koagulation i blodplasma. Derudover blev imaging aktiveret endotel celle og deposition af immunoglobuliner, supplement- og koagulation proteiner på de endothelialized perler udført af Konfokal mikroskopi. Denne analyse kan også bruges til at teste medicin, der er meningen for at forebygge endotel celle aktivering og dermed koagulation. Oven på dens potentiale til at nedbringe antallet af dyr, der anvendes for sådanne undersøgelser, er beskrevet analysen let at udføre og konsekvent reproducerbare.

Efter 7-10 dage af kultur i spinner kolbe (figur 1) var cellerne sammenflydende, der dækker hele overfladen af microcarrier perler (figur 2).
