Vaskulære endotelet består av en monolayer av endotelceller (EC) som linje lumen av blod fartøy. I en fysiologisk tilstand, quiescent EU er ansvarlig for vedlikehold av en antikoagulerende og anti-inflammatorisk miljø. 1 dette er formidlet av uttrykk for antikoagulerende og anti-inflammatoriske proteiner på EC overflaten. For eksempel EC aktivisering skyldes iskemi/reperfusion skade eller vaskulær avvisning av (xeno-) transplantert organene resulterer i en endring av endothelial overflaten fra en antikoagulerende og anti-inflammatorisk staten Pro koagulere og pro-inflammatoriske staten. 1
For å studere fascinerende og komplekse samspillet mellom de endotel, og koagulering faktorene, i vitro modeller som etterligner som nært i vivo situasjonen er svært ettertraktet. En vanlig begrensning som preger konvensjonelle i vitro koagulering analyser er bruken av anticoagulated blod som gjør analysen av koagulering-mediert effekter krevende og selv recalcification citrerte hele blod ikke kan gjengi resultater oppnåelig med friske ikke-anticoagulated blod. 2 dessuten i tradisjonelle flat-bed celle kultur systemer er det umulig å utnytte egenskapene antikoagulerende endotelet som en tilstrekkelig endothelial celle-overflate per blodvolum ikke kan nås. Modellen presentert her overvinner disse begrensningene av dyrking EC på overflaten av sfærisk microcarrier perler, slik at en EC overflate-til-blod forholdet &#62; 16 cm2/mL kan nås, som ligner situasjonen i små arterioler eller årer, og som ble beskrevet for å være tilstrekkelig til at "naturlig" anticoagulation blod av EC overflaten. 3 , 4 fullblod kan brukes uten ekstra antikoagulanter i denne innstillingen. Et annet interessant program inkluderer testing av narkotika som skal hindre endothelial celle aktivering, og dermed koagulering. 5 men denne modellen ikke kan helt erstatte dyr eksperimentering, det tilbyr en metode for å teste spesifikke funksjonelle hypoteser ex vivo bruke celler og dermed redusere antall dyr som brukes i grunnleggende forskning på iskemi/reperfusion skade eller (xeno) transplantasjon.

Endothelial celle aktivering, supplement og koagulering systemer tett kontrollert og sammen. 6 det er derfor viktig å forstå hvordan ulike transgene cellene oppfører seg etter eksponering for menneskelig blod vedheft molekyl uttrykk og cytokin utgivelse, utgytelsen av glycocalyx og tap av antikoagulerende proteiner. 7

I vivo, endotelceller er avgjørende for den naturlige anticoagulation av sirkulerende blod. Denne analysen kan også brukes til å teste medisiner som skal hindre endothelial celle aktivering, og dermed koagulering. På dens potensial for å redusere antall dyr som brukes til slike undersøkelser, er beskrevet analysen enkel å utføre og konsekvent reproduserbar.

Etter 7-10 dager av kultur i spinner kolbe (figur 1) var cellene confluent, som dekker hele overflaten av microcarrier perler (figur 2). Bekreftelse av confluency er et viktig skritt fordi en ikke-confluent monolayer av EC på microbeads vil føre til en markant nedgang i clotting tid, gitt microcarrier perler overflate er sterkt Pro koagulere (clotting tid: 4 ± 1 min) ( Figur 3).
