Menneskelige iPS og ES celler kan ikke være dyrket på konvensjonelle vev kultur polystyren (TCP) retter33,34 fordi human ES og iPS celler krever bestemte binding til ECMs, for eksempel vitronectin eller laminin å opprettholde deres pluripotency under langsiktige kultur. human pluripotent stem cell culture on polyvinyl alcohol-co-itaconic acid hydrogels with varying stiffness under xeno-free conditions

Effekten av fysisk stikkord, for eksempel stivhet av biologisk materiale om spredning og differensiering av stamceller, har blitt undersøkt av flere forskere. Men har de fleste av disse etterforskere brukt polyakrylamid hydrogels for stamcelleforskningen kultur i sine studier. Derfor er resultatene kontroversielle fordi disse resultatene kan stamme fra Karakteristika av polyakrylamid og ikke fysisk køen (stivhet) av de biologisk materiale. Her beskriver vi en protokoll for å forberede hydrogels, som ikke er basert på polyakrylamid, hvor ulike stilk, cellene menneskelig embryonic stem (ES) celler og menneskelige indusert pluripotent-stamceller (iPS), kan kultivert. Hydrogels med varierende stivhet var forberedt fra bioinert polyvinylacetat alkohol-co-itaconic syre (P-IA), med stivhet kontrollert av crosslinking grad ved å endre crosslinking tid. P-IA hydrogels podet med og uten oligopeptides fra ekstracellulær matrix ble undersøkt som fremtidige plattform for stamcelleforskningen kultur og differensiering. Kulturen og tidens amniotic væske stamceller, liggende under adipose-avledet stilk celler, human ES celler og menneskelige iPS celler er beskrevet i detalj her. Oligopeptide P-IA hydrogels viste overordnede forestillinger, som ble indusert etter stivhet egenskaper. Denne protokollen rapporter syntese av biomateriale, deres overflate manipulasjon, samt kontrollere egenskapene stivhet og til slutt deres innvirkning på stamcelleforskningen skjebne xeno-fri kultur betingelser.
