Le développement de ligands pour moduler les interactions protéine-protéine (IPP) offre une approche intéressante pour la découverte de médicaments modernes. Ainsi, beaucoup d’efforts ont été investi en étudiant les modalités chimiques roman qui pourraient moduler efficacement IPP1,2,3. IPP se composent généralement des surfaces qui interagissent peu profonds, grands ou abandonnées, et les petites molécules sont généralement considérés comme des ligands inadaptés pour la modulation de l’IPP4,5. Avec une approprié interaction surface exposée, petits peptides qui imitent les caractéristiques structurales des interfaces de protéines représentent des candidats idéaux pour régler ce problème6,7. Cependant, les petits peptides sont généralement non structurées dans une solution aqueuse. Cela est dû au fait que les molécules d’eau qui concurrencent le réseau de liaison hydrogène intramoléculaire du squelette peptidique et conformations bien définies sont entropiquement défavorables dans l’eau8. En outre, les peptides' intrinsèquement faible stabilité et propriétés de perméabilité cellulaire largement limitent leur utilisation dans des applications biologiques9,10. Selon l’analyse de protein data bank (PDB), > 50 % des IPP impliquent hélice α court interactions11. Ainsi, les méthodes chimiques différentes ont été développées en ce qui concerne la stabilisation de l’hélice. Ceux-ci incluent disulfure/thioéther bond formation12,13,14, métathèse de fermeture de cycle15, lactam ring formation16, « cliquez » chimie17, ajout de perfluoroarenes18,19et vinyle-sulfure formation20.
Peptides hélicoïdales stabilisés sont largement utilisés pour différentes cibles intracellulaires, dont p53, oestrogène récepteurs, Ras, BCL-2 protéines de la famille et d’autres21,22,23,24. ALRN-6924, un tout-hydrocarbure agrafé peptide inhibiteur double de MDM2 et MDMX, est actuellement utilisé pour l’investigation clinique25. En quelques années, notre groupe a mis l’accent sur le développement de méthodes de stabilisation nouveau peptide utilisant thiol-ene et thiol-yne réactions26,27,28. En général, nous avons démontré que ces réactions amorcées sont efficaces dans des conditions douces lorsqu’il est naturellement abondante cystéine est utilisé. En outre, nous avons montré que ces réactions ont une tolérance de l’excellent groupe fonctionnel, sont bio-orthogonaux et sont est avérées pour être applicable pour les peptides et les protéines modifications29. Les peptides de sulfure tethered thioéther/vinyle qui en résulte largement améliorer l’espace chimique des peptides de contrainte, fournissent un centre de labile sur sangles modification et s’avère pour être applicable pour des utilisations dans nombreuses applications biologiques30 ,31,32. A ce jour, seuls les rapports limités ont été décrits au sujet de la cyclisation de peptide de thiol-ene/thiol-yne. Dans une étude publiée par Anseth et coll. en 2009, une réaction sur la résine intramoléculaire thiol-ene de cyclisation de peptide entre alcènes activés avec la cystéine a été démontrée33. En 2015, Chou et al. décrit une réaction bi-composants radical initiés thiol-ene peptide agrafage34 et une réaction de couplage thiol-yne/ene ultérieurs, séquentiel35. Récemment, nous avons décrit une série de travaux basés sur thioéther/vinyle sulfure tethered peptides20,26,27. Ce protocole décrit une synthèse détaillée des peptides susmentionnés de thioether/vinyle sulfure attaché dans l’espoir qu’il sera utile pour la collectivité de la recherche.

constructing thioether/vinyl sulfide-tethered helical peptides via photo-induced thiol-ene/yne hydrothiolation

Nous décrivons ici un protocole détaillé pour la préparation de peptides thioéther-attaché à l’aide de résine sur intramoléculaire/intermoléculaire thiol-ene hydrothiolation. En outre, ce protocole décrit la préparation de peptides vinyle-sulfure-attaché à l’aide de dans-solution intramoléculaire thiol-yne hydrothiolation entre amino acids qui possèdent des chaînes latérales alcène/alcyne et résidus de cystéine à i, i + 4 positions. Peptides linéaires ont été synthétisés à l’aide d’une synthèse de peptide de phase solide axée sur le Fmoc standard (SPPS). Thiol-ene hydrothiolation est réalisée en utilisant une réaction intramoléculaire thio-ène ou une réaction intermoléculaire thio-ene, selon la longueur de peptide. Dans cette recherche, une réaction intramoléculaire thio-ène est effectuée dans le cas des peptides plus courts utilisant sur résine déprotection des groupes trityle des résidus de cystéine après la synthèse complète du peptide linéaire. La résine est ensuite définie à une irradiation UV à l’aide de photo-initiateur 4-méthoxyacétophénone (carte) et 2-hydroxy-1-[4-(2-hydroxyethoxy)-phenyl]-2-methyl-1-propanone (MMP). La réaction intermoléculaire thiol-ene est réalisée en dissolvant Fmoc-Cys-OH dans un solvant de N, N- diméthylformamide (DMF). Cela réagit ensuite avec le peptide en utilisant les résidus de l’alcène portant sur résine. Après cela, la macrolactamization est effectuée en utilisant hexafluorophosphate de benzotriazole-1-yl-oxytripyrrolidinophosphonium (PyBop), 1-hydroxybenzotriazole (HoBt) et 4-méthylmorpholine (NMM) comme réactifs d’activation sur la résine. Suite à la macrolactamization, la synthèse de peptide est maintenue à l’aide de SPPS standard. Dans le cas de la hydrothiolation thio-yne, le peptide linéaire est clivé de la résine séché et ensuite dissous dans le DMF dégazé. C’est ensuite irradié à l’aide de rayons UV avec photo-initiateur 2,2-dimethoxy-2-phenylacetophenone (DMPA). Suite à la réaction, DMF est évaporée et le résidu brut est précipité et purifié à l’aide de la chromatographie en phase liquide à haute performance (HPLC). Ces méthodes pourraient fonctionner afin de simplifier la génération de peptides cycliques thioéther-attachés en raison de l’utilisation de la chimie de clic de thio-ene/yne qui possède la tolérance supérieure groupe fonctionnel et avec un bon rendement. L’introduction des liaisons thioéther peptides profite du caractère nucléophile des résidus cystéine et oxydo-réduction-inerte par rapport aux liaisons disulfide.

Les spectres HPLC et MS du peptide Ac-YmS5AAAC-NH2 et son produit cyclisés Ac - Y-(cyclo-1,5)-[mS5AAAC] - NH2 qui ont été générés à l’aide de la photoréaction sur résine intramoléculaire thiol-ene sont représentés en Figure 6 b. le peptide cyclique s’est avéré pour avoir un poids moléculaire identique par rapport à son précurseur linéaire.
