この方法は、マウスモデルにおけるヒト乳癌の進行と治療を模倣する方法に関する乳癌分野の重要な質問に答えるのに役立ちます。このマウス乳癌モデルの主な利点は、整形移植後の腫瘍形成率が高く、乳房切除後の局所再発率が低い点である。まず、麻酔を受けた雌マウスのつまみつまみへの応答の欠如を確認し、動物の手足を外科用ボードにテーピングすることから始めます。滅菌綿棒を使用して、クロルヘキシジン、ヨウ素および75%エタノールで外科領域を殺菌する。そして、胸壁の真ん中に5ミリメートルの皮膚切開を行います。次に、切開に隣接する皮膚の右側を持ち上げ、滅菌マイクロディションハサミを使用して胸壁から皮膚を取り外します。皮膚を反転させて右2番目の乳脂肪パッドを露出させ、28.5ゲージの針を備えた1ミリリットルのインスリン注射器を使用して、脂肪パッドに創傷を通して20マイクロリットルの癌細胞懸濁液を慎重に注入する。脂肪パッド内で針を5秒間保持し、ゼラチン性癌細胞タンパク質混合物が針を取り除く前に固化し、滅菌5-0非吸収性縫合糸で切開を閉じる。その後、完全な債務まで監視ときれいなケージに動物を置きます。癌接種後8日後に乳房切除術を行うために、ちょうど実証したように、手術のためにマウスを準備し、5ミリメートルの皮膚切開を癌細胞接種から外科的瘢痕の左に2ミリメートルにする。前肢の根に向かって切開を伸ばして腫瘍を除去し、外科用瘢痕を含む皮膚と、腫瘍に接触する病変と、乳房切除術時に通常目に見えるリンパ節が存在しない腋窩リンパ節流域を引き出す。その後、Y.の形をした無菌5-0の非吸収性縫合糸で皮膚欠損を閉じ、完全な現職まで監視してケージに動物を戻します。肺転移定量術のために 21 接種後 21 日 D-ルシフェリン IP 15 分前に 1 キログラムあたりミリグラムを注入します。.そして、湾曲したマヨハサミを使用して、中腹部の皮膚と腹骨を開きます。横隔膜が視覚化されるまで肝臓を押し下げ、横隔膜と両側肋骨をカウダードからセファラードに切る。前胸郭壁を反転して肺を露出させ、肺を持ち上げて胸部食道を識別します。食道をマイクロディセクサで切り、鉗子を使って両側の肺と心臓をカウダードに向かって引っ張る。その後、気管と主要な血管をセファルドの肺の頂点に切断し、肺を生物発光イメージングのために10センチメートルのペトリ皿に入れます。はさみを使って肺組織から心臓を取り除きます。乳腺脂肪パッド癌細胞接種後約8日後に始まり、肺の生物発光は生きたマウスの原発性腫瘍負担を反映した原発性病変よりも深く、小さい。カリパーを用いた腫瘍サイズの測定は、その中での腫瘍体積の推定を可能にする。この乳房切除術モデルは、動物を安楽死させることなく連続転移性腫瘍負担モニタリングも可能にする。肺転移の腫瘍の負担は、ex vivoイメージングによっても定量することができるが、マウスは安楽死させなければならないという制限がある。外科的処置後のマウスの生存は、翻訳可能な臨床エンドポイントとしても監視することができる。その開発後、この技術は、乳がんの分野の研究者が新薬開発だけでなく、一般的に乳がん研究を探求する道を開きました。
