Nanotechnologie verwijst naar de toepassing van nano-sized materialen die worden gebruikt in alle wetenschappelijke gebieden, met inbegrip van de geneeskunde, materialen wetenschap, en biochemie. Bijvoorbeeld, ZnO NPs die bekend staan om hun ultraviolet verstrooiing, chemische sensing, en anti-microbiële eigenschappen, evenals hoge elektrische geleidbaarheid, worden gebruikt in de productie van verschillende consumentenproducten zoals voedsel verpakking, cosmetica, textiel, rubbers, accu's, katalysator voor de behandeling van auto-staart gassen, en biomedische-gerelateerde toepassingen1,2,3.
De ontluikende toepassingen van ZnO-producten op basis van NP, die leiden tot een toegenomen menselijke blootstelling aan ZnO NPs, hebben echter bezorgdheid geuit over hun potentiële nadelige gevolgen voor de menselijke gezondheid. Een aantal in vitro cellulaire studies hebben aangetoond dat ZnO NPs kan induceren oxidatieve stress, autophagy-gerelateerde cytotoxiciteit, ontsteking, en genotoxiciteit4,5,6,7,8 . Met name, de toxiciteit van ZnO NPs wordt verondersteld te worden veroorzaakt door de ontbinding van Zn tot vrije Zn2 + ionen, evenals de oppervlakte reactiviteit van ZnO, resulterend in de cellulaire Ionische en metabole onevenwichtigheden die zijn gekoppeld aan verminderde Ionische homeostase en een remming van Ion transport4,7,9,10. Belangrijk, studies hebben aangetoond dat de generatie van reactieve zuurstof soorten (ROS) is een van de primaire mechanismen onderliggende ZnO NPs-geassocieerde toxiciteit. Onvoldoende anti-oxidatieve activiteit na ROS-belediging is aangetoond dat zij verantwoordelijk zijn voor het opwekken van de cytotoxiciteit en DNA-schade9. De toxische effecten van ZnO NPs zijn ook gerapporteerd in diermodellen, waaronder knaagdieren1, zebravis11,12, evenals de ongewerveld Drosophila13.
Drosophila fungeert als een goed opgezet alternatief diermodel voor toxiciteits screening van chemische entiteiten en nanomaterialen (nm's)14,15. Belangrijk is dat er een hoge mate van genetische en fysiologische gelijkenis bestaat tussen humaan en Drosophila die het gebruik van Drosophila rechtvaardigt als een in vivo model voor het evalueren van biologische reacties op milieuverontreinigingen zoals nm's 16. Bovendien zijn er veel voordelen van het gebruik van Drosophila vanwege zijn geringe omvang, korte levensduur, genetische geschiktheid en eenvoudig en kosteneffectief onderhoud. Bovendien is Drosophila op grote schaal goedgekeurd voor de studie van genetica, moleculaire en ontwikkelingsbiologie, sinds zijn volledige genoom jaren geleden volledig is gesequenced terug in 2000, waardoor het geschikt is voor een verscheidenheid aan hoge doorvoer screening en voor het aanpakken van onopgeloste biologische vragen17,18,19,20,21. In de afgelopen jaren zijn een aantal studies gerelateerd aan immunotoxiciteit met behulp van verschillende soorten NPs in Drosophila gemeld15,22,23,24. Deze fundamentele nieuwe kennis verkregen uit de studies met Drosophila heeft geholpen om meer inzicht te geven in ons begrip van nano toxicologie.
ROS is een bekende boosdoener voor cytotoxiciteit en genotoxiciteit veroorzaakt door NPs, in het bijzonder, op metaal gebaseerde NPs25. ROS zijn zuurstof bevattende chemische soorten met hogere reactieve eigenschappen dan moleculaire zuurstof. Vrije radicalen zoals superoxide radicaal (O2-) en zelfs, niet-radicale moleculen zoals waterstofperoxide (H2O2) kunnen fungeren als Ros. Onder normale fysiologische aandoening, ze zijn vereist voor het handhaven van cellulaire homeostase26, echter, buitensporige Ros als gevolg van overproductie of disregulatie van de antioxidant afweersysteem kan oxidatieve stress veroorzaken, wat leidt tot schade aan eiwitten, lipiden en deoxyribonucleïnezuur (DNA)27. Hier bieden we protocollen voor het uitvoeren van cellulaire en genetische analyses met behulp van gekweekte zoogdieren cellen en Drosophila om te bepalen van de mogelijke nadelige effecten van ZnO NPs. Zinkoxide nanodeeltjes (ZnO NPs) hebben een breed scala aan toepassingen, maar het aantal rapporten over ZnO NP-geassocieerde toxiciteit is de afgelopen jaren snel gegroeid. Echter, studies die de onderliggende mechanismen voor ZnO NP-geïnduceerde toxiciteit verhelderden zijn scanty. We bepaalden de toxiciteits profielen van ZnO NPs met behulp van zowel in vitro als in vivo experimentele modellen. Een significante afname van de levensvatbaarheid van de cel werd waargenomen in ZnO NP-blootgestelde MRC5 Long fibroblasten, waaruit blijkt dat ZnO NPs cytotoxische effecten uitoefent. Evenzo, interessant, gut blootgesteld aan ZnO-NPs tentoongesteld een dramatische toename van reactieve zuurstof soorten niveaus (ROS) in de fruitvlieg Drosophila. Meer diepgaande studies zijn nodig om een risicobeoordeling voor het toegenomen gebruik van ZnO NPs door consumenten vast te stellen.

Onze methoden schetsen een eenvoudige en herhaalde manier voor het bestuderen van nanotoxiciteit, zoals het onderzoeken van de soorten celdood en het onderzoeken van de productie van reactieve zuurstofsoorten. Fluorescentie geactiveerd cel sorteren is een strikt formaat dat ons in staat stelt om de subpopulatie van cellen en de groeiende verschillende soorten celdood te bestuderen. De DHE sonde studiemethode stelt ons in staat om reactieve zuurstofsoorten te detecteren en te kwantificeren, gevolgd door flow cytometrie, microscopische beeldvorming met hoge inhoud en door een op elkaar gebaseerde fluorescentieanalyse.Om te beginnen, plaats een Eppendorf buis met zinkoxide nanodeeltjes in zinkoxide nanodeeltjes voorraad oplossing in een sonicator gedurende 15 minuten. Bereid zinkoxide nanodeeltjes in verschillende concentraties met behulp van een-milligram-per-milliliter zinkoxide nanodeeltjes voorraad. Dan, in een bioveiligheidskast, zaad MRC5 menselijke longfibroblasten op drie zes-put kweekplaten bij de concentratie van één keer 10 tot de vijfde cellen per put.Broed de cellen uit op 37 graden Celsius. Behandel de cellen na één dag met twee milliliter zinkoxidenanodeeltjes gedurende acht uur, 16 uur of 24 uur. Verzamel de cellen op elk moment in een centrifugebuis van 15 milliliter en vercentrifugeer vijf minuten op 300 keer g.Was de celpellets twee keer met fosfaatgebufferde zoutoplossing en zet de cellen opnieuw op met 1X bindingsbuffer. Voeg vervolgens vijf microliters van FITC Annexin V-vlek toe in vijf microliter van propidium jodide DNA-vlek. Broed de gekleurde cellen gedurende 15 minuten bij kamertemperatuur in het donker.Voordat u de cellen sorteert op stroomcytometrie, moet u de monsters opnieuw opvullen met een extra 400 microliter 1X-bindingsbuffer. Tabuleren van de staafdiagram met behulp van de mediane intensiteit verkregen. Gebruik nu een pipet om een milliliter nanodeeltjes toe te voegen in verschillende concentraties in flacons, gevolgd door negen milliliter vliegvoedsel om een laatste concentratie van 0,1 milligram per milliliter, 0,25 milligram per milliliter of 0,5 milligram per milliliter zinkoxide nanodeeltjes te maken.Pipet op en neer om grondig te mengen. Verdoven dan volwassen mannelijke en vrouwelijke vliegen in een flesje met kooldioxide, en introduceer vijf mannelijke vliegen en vijf vrouwelijke vliegen in elke flacon gedurende vijf dagen.Laat ze paren en leg eieren op het oppervlak van het voedsel. Na verdovende houding, verwijder de ouderlijke vliegen en laat de eieren om verdere ontwikkeling te ondergaan bij kamertemperatuur, die bestaat uit vier verschillende ontwikkelingsstadia. Embryonale, larve, pupal, en volwassen stadium.Na 72 tot 120 uur ontwikkelen vers gelegde eieren zich tot de late derde instarlarven. Verzamel met behulp van tangen de larven van de wand van de flesjes voor analyses. In een put van een dissectieschotel met dissectiemedium of PBS plaatst u de larven in de oplossing.Gebruik onder de stereomicroscoop het puntje van de tangen om een klein gaatje te maken en breek de nagelriemlaag van de larven open. Plaats de darm in een 1,5 milliliter microcentrifugebuis met Schneiders Drosophila Medium.Voeg een microliter van de DHE-sonde toe en broed gedurende 10 minuten in het donker in het donker. Was de darm drie keer met Schneider's Medium om de vijf minuten. Monteer de darm op glazen platen en lever 10 microliter antifademontagemedium met DAPI.Leg beelden vast onder een confocale microscoop. Selecteer een gebied zonder fluorescentie om de achtergrond in te stellen. Klik op meten.Exporteer de gegevens naar de spreadsheet en bepaal de gecorrigeerde totale celfluorescentie. Een staafdiagram maken en statistische analyses uitvoeren. In deze studie vertonen met zinkoxide nanodeeltjes behandelde cellen een hoger percentage vroege R3- of late apoptotische R6-cellen dan controlecellen R5. Necrotische celdood werd aangeduid door R4. De darm uit de drosophila larve was verdeeld in drie afzonderlijke domeinen van verschillende ontwikkelingsoorsprong.Namelijk, de voorgedei, midgut, en achterduit. DHE voorraad heeft de neiging om vrij snel verlopen, dus wees voorzichtig met de voorraad oplossing die u gebruikt. U ook andere sondes proberen, bijvoorbeeld cel Roche, die is ontworpen om de intracellulaire ROS in groene signalen te detecteren, en dat is fototabler dan de DHE.Ons protocol biedt een algemeen overzicht voor het bestuderen van de intracellulaire ROS-productie en kwantificering van het ROS-niveau voor nanotoxiciteitsstudie. DMSO staat bekend als een beter oplosmiddel voor DHE dan de PBS. DMSO is echter giftig.Daarom wil ik de gebruiker adviseren om het hele proces voor het bestuderen te beperken tot maximaal 30 minuten. Dit komt omdat u een vals-positieve uitkomst waarnemen als DMSO celdood kan veroorzaken.
