Zoals eerder vermeld, PC12 en de subclones worden op grote schaal gebruikt voor het bestuderen van neurite uitbreiding omdat ze uitstekende transfectie efficiëntie2,3. In tegenstelling, primaire neuronen hebben een lage transfectie tarief, dat is een groot obstakel voor het bestuderen van neuriet uitgroei regulators door transfectie7. Hier beschrijven we een handig protocol voor het kwantificeren van neuriet uitgroei in primaire neuronen....

Neurites, met inbegrip van zowel axonen en dendrites, zijn de projecties van neuronen die betrokken zijn bij de oprichting van de neurale netwerken. De dynamische uitgroei van neurites is essentieel voor neuro ontwikkeling. De onderliggende regelgevings mechanismen eronder blijven echter onduidelijk. In het bijzonder, neuriet schade wordt vaak waargenomen in verschillende neurodegeneratieve ziekten en na hersenletsel1. Daarom zou onderzoek naar de rollen van putatieve moleculen in verschillende neuriet uitgroei regelgevende trajecten ons begrip van het proces verbeteren. Bovendien, het kan onthullen nieuwe therapeutische doelen voor verschillende neurologische aandoeningen. Neuronale cellijnen zijn waardevolle modellen voor het bestuderen van neuronale processen met inbegrip van neuriet uitgroei als ze zijn gemakkelijk te manipuleren en transfect2,3. Echter, genetische drift is gemeld te voorkomen in sommige veelgebruikte cellijnen, die kunnen leiden tot variaties in hun fysiologische reacties4. Bovendien is differentiële eiwit expressie aangetoond tussen neuronale cellijnen en primaire neuronen. Bijvoorbeeld, PC12, een neuronale cellijn afgeleid van Rat bijnier die op grote schaal wordt gebruikt voor het bestuderen van neuriet uitgroei2,3, doet niet Express NMDA receptoren5. Bovendien is voorgesteld dat de verminderde responsiviteit van de muis neuroblastoom lijn neuro-2a naar neurotoxines in vergelijking met primaire neuronen is te wijten aan het gebrek aan expressie van bepaalde membraan receptoren en ionenkanalen6. Daarom, primaire neuronen zijn een meer wenselijk en representatief model voor het onderzoek van neuriet uitgroei. Echter, het gebruik van primaire neuronen wordt belemmerd door hun lage transfectie-efficiëntie7.
Hier beschrijven we een methode waarbij de co-transfectie van het eiwit van belang (POI) en EGFP aan primaire rat corticale neuronen. De EGFP fungeert als een morfologische marker voor de identificatie van succesvol getransfunde neuronen en maakt de meting van neurites mogelijk. We gevalideerd deze methode met behulp van verbindingen/moleculen die zijn gemeld aan het moduleren van neuriet uitgroei. Bovendien, FE65, een neuronale adapter eiwit dat is aangetoond dat het stimuleren van neuriet uitgroei, werd gebruikt ter illustratie van deze aanpak8,9. Dit protocol omvat (1) de isolatie van primaire corticale neuronen van embryonale dag 18 (E18) Rat embryo's, (2) de co-transfectie van neuronen met EGFP en de POI (FE65 in deze studie) en (3) de beeldvorming en analyse van de neuronen met behulp van de beeldverwerking software ImageJ met de NeuronJ plugin10,11.

Neuriet uitgroei is een fundamenteel voorval in de vorming van de neurale circuits tijdens de ontwikkeling van het zenuwstelsel. Ernstige neuriet schade en synaptische dysfunctie optreden in verschillende neurodegeneratieve ziekten en leeftijdsgebonden degeneratie. Onderzoek van de mechanismen die neuriet uitgroei reguleren zou niet alleen waardevolle licht werpen op de ontwikkeling van de hersenen processen, maar ook op dergelijke neurologische aandoeningen. Vanwege de lage transfectie-efficiëntie is het momenteel uitdagend om het effect van een specifiek eiwit op neuriet uitgroei in primaire zoogdieren neuronen te bestuderen. Hier beschrijven we een eenvoudige methode voor het onderzoek naar neuriet uitgroei door de co-transfectie van primaire rat corticale neuronen met EGFP en een eiwit van belang (POI). Deze methode maakt de identificatie mogelijk van POI-getransfunde neuronen via het EGFP-signaal, en dus kan het effect van de POI op neuriet-uitgroei nauwkeurig worden bepaald. Deze EGFP-gebaseerde assay biedt een gemakkelijke aanpak voor het onderzoek van trajecten reguleren van neuriet uitgroei.

Om deze methodologie te testen, we gebruikten cyto D en zenuw groeifactor NGF, die is aangetoond dat remt en stimuleren van neuriet uitgroei respectievelijk14,15,16. Lage transfectie-efficiëntie is een belangrijk obstakel om te bepalen hoe eiwit neurietuitgroei in primaire neuronen reactiveert. Ons protocol heeft dit probleem opgelost door mede-transfectie van EGFP om met succes transfected cellen te identificeren. De hoge co-transfectie snelheid van EGFP en de eiwitten van belang zorgt voor de nauwkeurigheid van de test.En daarom is nu immunofluorescentievlekken nodig. Aangezien neuriet ontgroeiing optreedt tijdens neurale regeneratie, kan deze methode worden toegepast op de identificatie van nieuwe doelen voor het herstel van het beschadigde neuronale netwerk bij hersentrauma of neurodegeneratieve ziekten. Voordat u met de procedure begint, plaatst u een steriele 18 millimeter cirkelvormige coverslip in elke put van een 12 goed weefselkweekplaat en coat elke coverslip met vijf microgram per millimeter Poly-D-Lysine-oplossing in een bevochtigde 37 graden Celsius incubator gedurende ten minste een uur.Aan het einde van de incubatie, aspirate de Poly-D-Lysine oplossing en spoel de coverslips met steriel water. Plaats op embryonale dag 18 de baarmoeder die is geoogst van een getimede zwangere Sprague Dawley-rat in een petrischaaltje van 10 centimeter en gebruik een kleine ontleedschaar om de baarmoeder en de vruchtzak te openen. Gebruik de schaar om de placenta te verwijderen en gebruik tangen om een heel embryo over te brengen in een nieuwe petrischaal van 10 centimeter met gekoelde PBS-glucose.Gebruik de schaar te snijden langs de sagittale hechting van de schedel en gebruik een kleine platte spatel om de embryonale hersenen over te brengen in een nieuwe 10 centimeter petrischaal met verse ijskoude PBS glucose. Met behulp van twee nummer vijf pincet en een dissectie microscoop, scheiden de twee hersenhelften van het cerebellum en hersenstam en verwijder de hersenvliezen. Isoleer vervolgens de cortex van de hersenhelften en plaats de cortex in een 15 milliliter buis met verse ijskoude PBS glucose.Voor primaire corticale neuron cultuur, in een klasse II bioveiligheid kabinet, laat de cortex te regelen door de zwaartekracht gedurende vijf minuten op vier graden Celsius voor de vervanging van de PBS glucose met 0,05% trypsine EDTA. Meng met zachte tikken en uitbroed het weefsel in een 37 graden Celsius waterbad gedurende 10 minuten met extra zachte tikken om de twee minuten. Aan het einde van de incubatie, stop de spijsvertering met vier milliliter onderhoudsmedium en gebruik een 1 milliliter pipet om het weefsel te scheiden met zachte trituratie.Sediment de cellen door centrifugatie en resuspend de pellet in een milliliter onderhoudsmedium voor een tweede centrifugatie. Resuspend de pellet gevormd na de tweede centrifugatie in een milliliter van vers onderhoud medium voor het tellen en plaat van de geïsoleerde neuronen op een 6,5 keer 10 tot de vierde levensvatbare cellen per centimeter kwadraat concentratie in een milliliter van onderhoud medium per goed in een 12 put plaat. Na twee dagen bij 37 graden Celsius en 5% kooldioxide, verdun EGFP en doel eiwit plasmid DNAs en transfectie reagens in twee afzonderlijke 1,5 milliliter buizen en meng ze samen.Transfecteer de cellen met EGFP construct door het toevoegen van de transfectie mix aan de putten. Na 24 uur was je de cellen met 37 graden Celsius PBS en bevestig ze met 4% paraformaldehyde in PBS gedurende 10 minuten in het donker bij kamertemperatuur. Aan het einde van de incubatie, was de vaste cellen drie keer met verse PBS per wasbeurt en voeg een minimaal volume van fluorescentie montage medium op een microscoop glas dia per monster.Breng vervolgens de gecoate covers voorzichtig over op de montagemedia met de monsters naar de glazen glijbanen. Om de neurietgroei in beeld te brengen, selecteert u de 40X-doelstelling op een epifluorescente microscoop en klikt u op het vastleggen van beelden van ten minste 40 intacte EGFP-positieve neuronen per transfectie. Wanneer alle neurites zijn afgebeeld, open de vastgelegde beelden in ImageJ met de NeuronJ plugin en meet de lengte van de langste neurite van elk neuron van het cellichaam naar het puntje van de groeikegel.Analyseer vervolgens de gegevens verkregen met de software om het effect van de beoogde eiwitten op de groei van neuriet te bepalen. Cytochalasine D onderdrukt neurite extensie in een dosis-afhankelijke manier, terwijl neurite uitgroei wordt verbeterd in neuronen behandeld met zenuwgroei factor. Bovendien stimuleert het neuronale groeiadaptereiwit FE65 de tweevoudige onverzette neurotische uitgroei aanzienlijk in vergelijking met onverzetteerde neuronen.Ondanks een lage transfectie-efficiëntie, immunofluorescentie analyse blijkt dat meer dan 80% van een twee-daagse celcultuur kan met succes worden gecotransfected met EGFP en FE65. EGFP-expressie wordt gedetecteerd op zes uur na de transfectie, terwijl FE65 voor het eerst wordt gedetecteerd op uur 12 en beide signalen worden gedetecteerd voor maximaal zeven dagen na de transfectie. De transfectie van primaire rattencorticale neuronen met verschillende hoeveelheden FE65 plasmide DNA onthult een dosisafhankelijke toename van neurietextensie met 0 tot 0,5 microgram FE65 plasmide.Er wordt echter geen significant verschil in groei waargenomen bij neuronen die worden doorfecteel met 0,5 of een microgram van het plasmide. Bij het proberen van het protocol is het belangrijk om het juiste gebied te isoleren van de embryonale hersenen. Deze techniek kan ook worden toegepast op de studie van neurale groei in menselijke iPSC-afgeleide neuronen die waardevolle instrumenten voor regeneratieve geneeskunde hebben.
