在使用LC/MS对分析物进行量化时，在样品和标准之间实现相同的电电化效率是一项挑战。此外，由于质子亲和力和极性10的信号分裂和异质性，代谢物多样性的定量变得更加具有挑战性。最后，样品的共解基质也会影响化合物的电电化效率。为了解决这些问题，代谢物可以化学衍生，提高LC/MS系统的分离分辨率和灵敏度，同时在某些情况下减少信号分裂10，11。化学衍生的工作原理是标记代谢物的特定功能组，以调整其物理特性，如电荷或疏水性，以提高电化效率11。各种标记剂可用于针对不同的功能组（例如，胺、羟基、磷酸盐、碳化物酸等）。Aniline，一种这样的衍生剂，一次瞄准多个功能组，并在亲水分子中加入疏水成分，从而提高其分离分辨率和信号12。为了解决共发基质电抑制效应，杨和同事开发了一种基于组特定内部标准技术 （GSIST） 标签的技术，其中标准标有13 C阴宁同位素，并与样品混合12，13.代谢物和相应的内部标准自共溶以来具有相同的电化效率，其强度比可用于定量实验样品中的浓度。
在这项研究中，我们开发了一个协议来检测和量化参与糖解的40种化合物，磷酸五氯苯星通路，三聚碳酸循环，能量代谢和CFPS反应中的副因子再生。该方法基于GSIST方法，我们使用12种C-单一素和13种C-aniline来标记、检测和量化代谢物，使用逆相LC/MS。所有化合物的线性范围跨越2-3个数量级，平均相关系数为0.988。因此，该方法是一种可靠而准确的方法，用于询问无细胞代谢，以及可能全细胞提取物。

absolute quantification of cell-free protein synthesis metabolism by reversed-phase liquid chromatography-mass spectrometry
