1. 凍結保存乳腺腫瘍の移植のための調製

動物間の腫瘍増殖の変動を減らすためには、移植のために腫瘍組織を1mm3 断片に切断することが重要である。軟組織を成長させるモデルは作業が難しく、腫瘍断片を少し大きく切る必要がある(1-2 mm3)。乳脂肪パッドポケットに組織を配置する場合は、これは複数の小さな腫瘍または奇妙な形の腫瘍をもたらすので、複数の部分に組織を分割しないように注意してください。...

ヒト乳癌の不均一性と薬物反応を忠実に再現する前臨床モデルは、腫瘍生物学を研究し、標的治療の潜在的なバイオマーカーを同定するために重要である。不死化細胞株は、これらの細胞株が成長しやすく、分子機構を研究するために遺伝的に改変することが容易であるため、乳癌研究に広く使用されています。しかし、インビトロでの長期細胞培養からの選択的な圧力のために、遺伝的ドリフトが経つにつれて起こり、乳癌細胞株は原発性乳房腫瘍7、8、9における収差とは異なる細胞株特異的ゲノム変化を運ぶ可能性がある。
患者由来異種移植片(PDX)腫瘍塊は、ヒト疾患の異質性を再現することができ、起源の腫瘍10、11、12、13、14、15、16、17、18、19、20、21、22に組織学的および免疫組織学的に類似している 、23,24,25,26,27,28,29.重要なことに、PDXモデルは、10、11、12、13、14、15、16、17、18、19、20、21、22によって証明されるように、複数の移植にわたって典型的に安定である、23,24,25,26,27,28,29.PDXモデルは、臨床的に観察されたものに匹敵する治療応答を示す10,11,12,13,14,15,16,17,18,19,20,21,22,23,24, 25、26、27 、28、29。エストロゲン受容体陽性(ER+)、プロゲステロン受容体陽性(PR+)、上皮成長因子2陽性(ERBB2+、HER2+)およびトリプルネガティブ乳癌(TNBC)PDXモデルが確立されており、内分泌、化学、標的療法を試験するための優れたプラットフォームを提供する。しかし、現在のPDXモデルの主な注意点の1つは、マウスの機能的免疫系の欠如である。
Trp53ホモ接合性ヌル、cMyc、Wnt1、PyMT、またはHer2過剰発現モデルなどの遺伝子組み換えマウスモデル(GEMM)は、無傷の免疫系の文脈における自発的な腫瘍開始、進行および転移の研究を可能にする。しかし、腫瘍の遅延は長く、複数の腕30、31で前臨床試験を行うことは困難です。しかし、GEMMをシンジニック宿主体に移植して、ヒト腫瘍32、33、34、35、36、37、38、39、40、41、42、43、44、45、42、44、45を密接に再現する腫瘍を十分な数の腫瘍を生成することができる。 46,47,48,49,50,51,52 ,53,54,55.例えば、p53-null BALB/cマウスからの乳腺上皮を、同種性野生型レシピエントマウスのクリアされた脂肪パッドに移植して原発性腫瘍を形成し、これはさらに同系宿主56,57に移植することができる。p53-ヌル腫瘍は、ヒト腫瘍の異なるサブタイプを再現した。
PDXモデルと移植可能なGEMMの組み合わせは、乳房腫瘍生物学、薬物応答および抗腫瘍免疫を調査するための貴重な前臨床ツールを提供する。現在のプロトコルでは、マウス乳腺脂肪パッドへのPDXおよびGEMM腫瘍断片の異所移植の方法が記載されている。これらのモデルは、シリアル通路に適しており、通常は安定した表現型を保持します。経時経過を越えて遺伝的ドリフトまたは異種性の喪失のリスクを軽減するために、生物学的または形態学的変化が経時29、58に観察される場合には、その後の移植のために、複数の組織断片が各通路で凍結保存される。
