適切な抗体に対する望ましい抗原の効率的かつ確実な結合は、DC標的化における重要なステップであり、他の要因の中でも抗原の形式に依存する。精製抗体に対する全長タンパク質の化学的結合は、考えられる1つの戦略である。これまで、マウスでの インビボ DC標的化研究に対して、モデル抗原オボアルブミン(OVA)とDEC-205特異的IgG2a抗体(αDEC-205)の架橋の確立に成功しました。プロトコルの最初のステップは、アフィニティークロマトグラフィーによるNLDC(非リンパ系樹状細胞)-145ハイブリドーマの上清からの抗体の精製である。精製された抗体は、スルホ-SMCC(スルホフチニミジル4-[N-マレイミドメチル]シクロヘキサン-1-カルボキシル酸)によって化学的共役のために活性化されると同時に、OVAタンパク質のスルフヒドリル基はTCEP-HCl(トリス(トリス(トリス(トリス(2-カルボシテヒリル)ホドロフェでインビクテーションを介して露出する。過剰なTCEP-HClおよびスルホ-SMCCは除去され、抗原は一晩の結合のために活性化された抗体と混合される。得られたαDEC-205/OVAコンジュゲートは濃縮され、非結合OVAから解放される。αDEC-205へのOVAの結合に成功した結合は、ウェスタンブロット分析および酵素結合免疫吸着測定法(ELISA)によって検証される。

このプロトコルは、実験条件のすべてが慎重に最適化されており、OVA抗原に対するDEC-205抗体の正常かつ効率的な化学的結合を促進するために使用することができます。この技術の主な利点は、タンパク質抗原と抗体の柔軟な選択を可能にし、これらの成分の遺伝的融合に依存しないことである。また、このプロトコルを使用して、生体内DCターゲティング用のC型肝炎ウイルスタンパク質および抗DEC-205のコンジュゲートを生成しました。これは同様に、抗腫瘍ワクチン接種アプローチにとって価値がある。反DEC-205生産を開始するには、 1%ペニシリンとストレプトマイシンを添加した37°Cの9ミリリットルのISF-1培地で1〜500万個の解凍された凍結保存NLDC 145細胞の1ミリリットルの体積を再懸濁し、細胞を37°Cで25平方センチメートルの細胞培養フラスコ培養用培養液に播種する。細胞が70%合流に達したら、細胞懸濁液全体を15ミリリットルの円錐管に移し、遠心分離によって細胞を採取する。ペレットを新鮮な完全な37度ISF-1培地の12ミリリットルに再懸濁し、75平方センチメートルのフラスコで細胞を再プレートします。48〜72時間の培養後、2つの新しい75平方センチメートルのフラスコの間で70%コンフルエント培養を分割し、各フラスコに新鮮な完全なISF-1培地の6ミリリットルを加えます。培養物が70%合流に達したら、10ミリリットルのピペットを使用して細胞懸濁液で細胞培養フラスコの底部および培養表面を洗い流し、すべての細胞を採取し、各拡張NLDC145細胞懸濁液の10ミリリットルを個々のPETGローラーボトルに移します。その後、各ローラーボトル培養に完全なISF-1培地の140ミリリットルを追加し、37°C、5%の二酸化炭素と3日間毎分25ラウンドでローラーボトルを培養します。NLDC 145上清からの抗体精製の場合、シリコンチューブの端部を上清で満たされたビーカーに入れ、800ミリリットルの上清がプロテインGセファローズカラムを通って滴下するようにします。溶出の場合は、1.5モルTris-HCLの100マイクロリットルを20個の1.5ミリリットルチューブのそれぞれに加え、ゴムプラグをカラムから取り外します。0.1モルグリシンの1ミリリットルをカラムの上部チャンバーに移し、溶出物を含む抗体を調製された1.5ミリリットルチューブの1つに集め、各チューブについて繰り返します。抗体を全て採取し、抗体含有溶出液をプールしたら、20センチメートル長の透析管の底を適切な透析チューブ閉鎖で閉じ、抗体溶出をチューブに慎重にピペットします。2つ目のクランプで透析管の上部を閉じ、透析管の上部クランプをフローティングスタンドに固定します。PBSのビーカーに磁気攪拌棒を追加し、磁気スターラーの上にビーカーを置きます。摂氏4度で一晩透析した後、1つのクランプを開いて、10キロダルトン分子量カットオフで遠心濃縮器に完全な透析液をローディングし、濃縮器を693倍Gと摂氏4度で30分間遠心分離します。スピンの終わりに、抗体溶液の最後の1〜1.5ミリリットルの体積が得られるまで、2番目の遠心分離のためにPBSの10ミリリットルで遠心濃縮器をロードします。OVAを抗DEC-205抗体にコンジュゲートするには、500マイクログラムのOVAタンパク質、240マイクロリットルの新たに調製されたTCEP-HCL溶液、560マイクロリットルの無菌超純水を1.5ミリリットルの室温で1.5ミリリットルのインキュベートに添加し、PBSの200マイクロリットルを加えます。同時に、900マイクロリットルのPBSに2.5ミリグラムの抗DEC-205を加え、100マイクロリットルの新しく調製したスルフォ-SMCCを2番目の1.5ミリリットルチューブに加え、加熱ブロックで毎分550回転で37°Cで30分のインキュベーションを行います。インキュベーションの最後に、緩めたキャップで脱塩カラムを置き、底をねじって個々の15ミリリットルの円錐管に入れ、遠心分離機でカラムを液体を取り除きます。遠心分離後、キャップを取り外した新しいチューブにカラムを入れ、溶液1列1カラムのコンパクト樹脂ベッドの中心に、スルフォ-SMCCおよびOVA TCEP-HCL溶液をゆっくりとロードします。遠心分離後、カラムを捨て、すぐに穏やかなピペットで抗体とOVA溶液を混合します。抗DEC-205/OVA溶液を遠心タンパク質濃縮器に積み込み、濃縮器にPBSを15ミリリットルの体積に充填します。2,000倍Gと室温で5分間コンセントレータを遠心分離し、さらに10ミリリットルのPBSで濃縮器を充填します。同じ遠心分離機条件下で少なくとも8分間の濃度を遠心した後、上部チャンバーから濃縮サンプルを静かに集める。ウェスタンブロット分析による結合の成功を確認するには、各サンプルからタンパク質標準のアリコートをSDSゲルにロードし、標準SDS-PAGE分析プロトコルに従ってゲルを実行します。ゲルブロッティング後、西洋ワサビペルオキシダーゼコンジュゲート抗体で膜を染色し、標準的なプロトコルに従って抗DEC-205またはOVAを検出します。その後、各サンプルに各タンパク質が存在する場合に、膜を分析することができます。ELISAによる共役の成功を確認するには、1ウェルあたり100マイクロリットルの抗OVA抗体を適切な96ウェルELISAプレートにコーティングし、抗DEC-205/OVAを1〜2の比率で連続して希釈し、ブロッキングバッファーを1ミリリットル当たり6マイクログラムから93.8ナノグラムに希釈します。各希釈液の100マイクロリットルを抗体コーティングプレートの適切なウェルに加え、室温で1時間のインキュベーションを行います。インキュベーションの最後に、室温で1時間インキュベートするためにプレートに抗DEC-205/OVAコンジュゲート抗体に対して100マイクロリットルの西洋ワサビペルオキシダーゼを加え、その後50マイクロリットルの西洋ワサビペルオキシダーゼ基質を各ウェルに加えて、分光光測定による色反応の分析を可能にする。並列ウェスタンブロット分析は共役OVAと共役抗DEC-205の両方を検出するために使用することができる。OVAの染色は、存在する場合は過剰な遊離OVAの検出を可能にし、抗DEC-205の染色は、裸の反DEC-205と共役との間の分子量の増加を通じて結合の成功を確認する。ELISAは、検出されたシグナルと、抗DEC-205/OVA共役の生成が成功したことを示すタンパク質の分析量との間の正の関連を持つ結合の成功を評価するためにも使用できます。フローサイトメトリーおよび免疫蛍光分析は、抗DEC-205コアが骨髄由来のCD11c陽性細胞に効率的に結合することを明確に示している。アジュバントと組み合わせた抗DEC-205/OVAによるワクチン接種は、OVAおよびアジュバント単独でのワクチン接種と比較して、マウスレシピエントにおけるOVA特異的IgGタイターの増加を誘発する。さらに、抗DEC-205/OVAは、抗DEC-205/OVA誘導CD8陽性T細胞応答を用いて、過特異的CD4およびCD8陽性T細胞応答を効率よく誘導し、OVA単独で誘導されたT細胞応答を大きく上回る。抗DEC-205抗体に対するOVA抗原の結合を成功させるためには、実証された材料を調製し、一貫した条件下で結合ステップを実行することが重要です。抗原と抗体の結合に成功した後、結合解析やインビボでの病原体または腫瘍関連免疫の誘導など、数多くの後続のアプローチが可能です。
