このプロトコルは、ネイティブニッチでヒトアストロサイトの分子機能を研究することに興味がある科学者を助けることができます。この技術の主な利点は、蛍光活性化核選別を用いてバルクヒト脳組織標本から特定の細胞型、成体アストロサイトの単離および濃縮を可能にすることである。この方法論は、他の適切な蛍光結合抗体を用いてニューロンおよび希突起膠細胞前駆細胞などのヒト新皮質から他の細胞型を単離するために適用することができる。新鮮な凍結ヒト成人皮質組織サンプルから約200〜400ミリグラムの組織の解剖を開始する。氷上の氷冷溶解バッファーを4ミリリットル含む7ミリリットルのガラス組織ドバウサーに組織を置き、組織を約50回粉砕する。ホモジネートを12ミリリットルの超遠心ポリプロピレンチューブに移します。5ミリリットルのピペットを使用して、スクロースと組織ホモジネートの間の界面を乱すことなく、6.5ミリリットルの氷冷スクロース緩衝液を超遠心管の底部に加えます。核を回収するには、溶解液を101、814倍Gで1時間超遠心分離し、ペレットを乱すことなく上清と破片を慎重に吸引します。PBS中の0.1%BSAを超遠心チューブに加え、核ペレットを再懸濁する前に氷上で10分間インキュベーションする。次に、トリパンブルーを使用して、無傷の核を光学顕微鏡下で可視化し、濃度が1ミリリットルあたり10〜第5核であることを確認する 単離された核の蛍光活性化ソートのために、Alexa fluor 555結合マウス抗NeuN抗体を含む抗体溶液に約20マイクロリットルの再懸濁サンプルを加える。約20マイクロリットルの再懸濁サンプルを、APC結合マウス抗PAX6抗体を含む抗体溶液に加える。光から保護した振盪を伴う摂氏4度の1時間のインキュベーションの後、DAPIをすべてのサンプルおよび対照に1:1000の比率で加える。適切なサイズの核を含み、赤血球および破片を排除するには、コントロールチューブを使用して、前方および側面散乱領域によって細胞をゲートする。一重項核集団を選択するには、前方散乱領域対前方散乱幅または高さおよび側面散乱領域対側面散乱幅または高さによってゲートする。DAPIによるゲートは、無傷の原子核一重項のみを含み、任意のデブリおよびダブレットを除外する。任意の追加の蛍光対照に従ってゲーティングした後、DAPIのみの対照試料を実行する。NeuN Alexa 蛍光 555 のみのコントロールを実行して、Alexa 蛍光 555 チャンネルでの NeuN 陽性染色のカットオフを決定します。PAX6 APC のみのコントロールを実行して、APC チャネルでの PAX6 陽性染色のカットオフを決定します。すべてのコントロールを実行した後、NeuN陽性細胞をゲートしてニューロンを収集し、PAX6陽性細胞をNeuN陰性集団とゲートしてアストロサイトを収集します。NeuN陰性PAX6陰性集団から関心のある追加のグリア集団をゲートして収集する。バルク単核RNAシーケンシングの場合、0.04%BSA添加PBSで50,000~500,000個の核を採取した後、2ミリリットルのスクロース溶液、50マイクロリットルの1モル塩化カルシウム、および30マイクロリットルの1モルの酢酸マグネシウムをサンプルに加える。懸濁液の最終容量をPBSで10ミリリットルまで持っていきます。チューブ内容物を反転させて混合し、氷上で反応物を15分間インキュベートする。遠心分離により核をペレット化し、核を1ミリリットルのRNA抽出試薬に再懸濁した。次いで、チューブをボルテックスし、ドライアイス上で試料を凍結し、核をマイナス80°Cで保存した。バルク単核トランスポザーゼ許容クロマチンシーケンシングでは、50,000~75,000個の核を0.04%BSA添加PBSに集め、5%BSAでコーティングした微量遠心チューブに入れ、下流分析まで核を凍結します。ニューロン、アストロサイト、および希突起膠細胞前駆核集団は、実証されるように新鮮な凍結死後ヒト側頭新皮質サンプルから選別することができる。単一核RNAシーケンシング研究はさらに、PAX6を原形質および線維性成体アストロサイト亜集団の両方にわたって発現差のある核転写因子のトップとして優先順位付けした。選別後、原発性病理学的てんかん新皮質における細胞型特異的トランスクリプトーム変化を特徴付けることができる。この解析では、トランスクリプトーム解析により、PAX6陽性NeuN陰性集団が汎神球マーカーで強固に濃縮され、ニューロンマーカーで枯渇していることが確認された。免疫蛍光染色は、ヒト皮質アストロサイトにおけるPAX6とグリア線維性酸性タンパク質との共局在を確認するために使用することができる。死後の間隔が短いほど、新鮮な組織サンプルからの無傷の核回収率が高くなります。無傷の核の高収率は、死後24時間まで凍結組織から回収することができるが、30時間で、非常に少数の無傷の核を回収することができる。覚えておくべき最も重要なことは、PAX6も発現するニューロンが存在するため、NeuN陰性集団からPAX6陽性集団を得ることであり、それらを除外したい。この技術はまた、てんかん手術用脳標本中のアストロサイトの研究を可能にし、特定の病理学的ニッチの文脈におけるヒトアストロサイトの分子調節不全をさらに解明した。
