膵臓は、消化管の外分泌機能と血流中にホルモンを分泌する内分泌機能を有する腹部器官である。膵臓の高解像細胞イメージングは、膵炎、膵臓癌、および糖尿病を含む膵臓を含む様々な疾患の病態生理を明らかにすることができる1。コンピュータ断層撮影、磁気分解能画像、および超音波検査などの従来の画像診断ツールは、臨床分野1,2で広く利用可能である。しかし、これらのイメージングモダリティは構造的または解剖学的変化のみを視覚化することに制限されているが、細胞レベルまたは分子レベルでの変化は決定できない。ヒトにおける糖尿病や膵臓癌の分子変化が診断3、4の10年以上前に開始できることを考えると、潜伏期間中の分子転移から膵臓疾患を検出することは、早期診断とタイムリーな介入を提供する可能性を秘めている。したがって、分解能の限界を克服し、機能に関する貴重な洞察を提供するイメージングは、膵臓癌の早期診断または、糖尿病の進行期における膵島の改変の高度な同定を提供することによって顕著に注目を集める5。
特に島と共に、核イメージング、生物発光イメージング、および光コメレンストモグラフィーは、非侵襲的な島頭撮影技術6として示唆されている。しかし、これらの方法の分解能は、数十~数百マイクロメートルの典型的な値で、小島の細胞レベルでの変化を検出する限られた機能を提供する、実質的に低い。一方、以前の高解像研究では、ex vivo7,8(例えば、 膵臓のスライスまたは消化、非生理学的9(例えば、膵臓の外在化)および異所性状態10、11、12(例えば、腎臓カプセルの下、肝臓内、および眼の前房に移植)、解釈および臨床的意味を制限する。もし、高解像イメージングのin vivo、生理学、および異形性体モデルを確立できれば、膵島の調査に不可欠なプラットフォームとなるでしょう。
生体動物の微視的分解能レベルで病態生理を明らかにする生体内イメージングは、最近、大きな注目を集めています13.in vivo イメージング法のうち、マウスの腹部に窓を埋め込む腹部イメージングウィンドウ14の開発により、新しい知見(すなわち、早期肝転移15 の前微小転移段階および腸上皮16における幹細胞維持のメカニズム)の発見が可能になった。腹部イメージングウィンドウは貴重な結果をもたらすが、膵臓に対するこの窓の用途および膵臓を含む疾患に基づく結果として生じる生体内イメージング研究は、広範囲に調査されていない。
ヒト膵臓の固形器官特性とは異なり、マウスの膵臓は、拡散分散した軟組織様構造17である。したがって、蠕動や呼吸などの生理学的な動きによって絶え間なく影響を受けます。膵臓の腹部イメージングウィンドウの適用に関する以前の研究は、腸の動きによって誘発された動きアーティファクトのためにさまようことを実証した18.結果として得られた平均画像では重度のぼかしが観察され、マイクロスケール構造の視覚化と同定が妨げられていた。
ここでは、膵臓を含む疾患における縦方向細胞レベル事象を調べるため、生体内顕微鏡19,20と組み合わせた新規支持基盤統合膵臓生体内イメージングウィンドウの使用について説明する。前の研究18の方法論の詳細な説明に加えて、膵臓を含む様々な疾患に対する膵画像化窓の拡張応用について本論文で説明する。このプロトコルでは、カスタム構築されたビデオレートレーザースキャン共焦点顕微鏡システムを、生体内顕微鏡システムとして利用した。4つのレーザーモジュール(405、488、561、および640 nmの波長)が励起源として利用され、4つの発光信号がバンドパスフィルタ(BPF1:FF01-442/46)を介して光増倍管(PMT)によって検出されました。BPF2: FF02-525/50;BPF3: FF01-600/37;BPF4: FF01-685/40)。レーザースキャンは、回転する多角形ミラー(X軸)と、ビデオレートスキャン(30フレーム/秒)を可能にするガルバノの走査ミラー(Y軸)で構成されていました。生体内顕微鏡に関する詳細は、前の研究10、18、19、20、21、22、23に記載されている。
以前の島の調査18では、遺伝子導入マウスモデル(MIP-GFP)24を用いて、生きたマウスの中の小島をGFPでタグ付けした状態で安定した画像を作成することに成功しました。この方法により、1週間の間に小島の変化を高解像度で可視化することができました。また、最大3週間の同じ膵島のイメージングを促進し、これは、糖尿病の病因の病因の間に機能的追跡またはモニタリングのための膵島の長期研究の実現可能性を示唆する。さらに、蛍光膵臓癌細胞(PANC-1 NucLightRed)25をマウスの膵臓に直接移植する、正腸膵臓癌モデルを開発した。膵臓内イメージングウィンドウの適用により、このモデルは、膵臓癌の腫瘍微小環境における細胞および分子病態生理学を調査し、新しい薬剤候補の治療的モニタリングのためのプラットフォームとして利用することができる。

このプロトコルは、新しい膵臓のインビタルイメージングウィンドウを用いた膵臓の細胞レベルの安定化および反復的な細胞レベルを記述する。この技術の主な利点は、生きたマウスの膵臓で3週間にわたって細胞レベルまでの解像度で動かない3次元イメージングを行うことが可能である。手術プラットフォームを準備し、70%エタノールで表面を消毒することから始めます。麻酔後、体温をモニタリングするために、ホーム熱制御加熱パッド付き直腸プローブを使用します。マウスの左脇腹から毛髪を取り除き、アルコールとヨウ素を代わりに塗布し、切開部位から外表面に回転し、切開部位に戻ることはなく、ヨウ素で終わる。その後、マウスの左脇腹に1.5センチメートルの切開を行い、皮膚と筋肉を解剖する。黒またはナイロンの4-0縫合糸を使用して、切開マージンで財布ひも縫合を行います。リング鉗子で慎重に脾臓を引っ張り、膵臓を識別します。マウスの脇腹に窓を置き、窓の空きスペースを通して脾臓と膵臓を渡します。次に、窓の空きスペースに脾臓を置きながら、画像窓のプレートに膵臓をそっと置きます。膵臓に直接PANC-1ナクリテレッドを注入します。31ゲージのカテーテル針を使用して、n-ブチルシアノアクリル酸接着剤の滴をイメージングウィンドウのマージンに塗布し、最小限の接着剤を塗布します。12ミリメートルの丸いカバーガラスを撮像窓の余白にそっと塗布します。次に、縫合ループを保持して窓の側面溝に収まり、3回結びます。最後に、マウスが起きているときにこれらのタイトなステッチの中断を防ぐために、ネクタイの最大近位部位を切断する。レーザーパワーを含む生体内顕微鏡のスイッチをオンにすることで開始します。血管カテーテルを挿入するには、人差し指と第3指で尾部の近位側に圧力をかける。必要に応じて、ランプで尾を加熱します。70%エタノールスプレーで尾静脈を消毒し、30ゲージのカテーテルを側面尾静脈に挿入し、PE-10チューブ内の血液の逆流を可視化します。カテーテルに絹テープを貼って安定させる。蛍光プローブの組み合わせに従って、FITCデキストランおよびTMRデキストラン、または他の蛍光プローブを適宜噴射します。直腸プローブを挿入して、家庭温熱パッドシステムで体温を自動的に制御します。次に、インビタル顕微鏡のセットアップ時に準備した膵臓イメージングウィンドウを窓ホルダーに挿入します。手術用プラットフォームからイメージング段階にマウスを移します。生体内イメージングを行うために、膵臓の画像化を4回などの低倍率でイメージングし、膵臓イメージングウィンドウ内の膵臓の全景をスキャンする。対象領域が決定されたら、20倍または40倍のような高倍率対比レンズに切り替え、それぞれ約0.5マイクロメートルおよび3マイクロメートルの横方向および軸解像度で細胞レベルのイメージングを行います。Zスタックまたはタイムラプスイメージングを行い、細胞移動などの3D構造または細胞レベルのダイナミクスを観察します。膵臓内イメージングウィンドウと組み合わせた生体内顕微鏡検査は、高解像イメージングの取得を可能にする長期組織安定性を提供し、個々の膵島を最大3週間追跡します。C57黒6Nマウスに静脈内注射抗CD31抗体を埋め込んだ窓は、Alexa 647フルオロフォアと共役し、広域イメージングを促進し、膵臓の3Dイメージングを拡大した。Acinar細胞は、隣接する血管系における膵臓組織の平均画像で同定され、それぞれ自己蛍光および抗CD31抗体を用いて可視化された。広視野と高解像度の画像を組み合わせたモザイクイメージング法を用いて、隣接する脈管構造を有する約40~50個の小島をMIP-GFPマウスで可視化した。以前の研究では、この安定したイメージング法を使用して、小島を最大3週間追跡できることが示されました。がん細胞を可視化するために、PANC-1ヌクリテライト赤細胞を手術中にマウス膵臓に直接移植し、近くの血管をAlexa 647と共役した抗CD31で染色した。このプロトコルを用いて、膵臓癌の広視野画像が生成された。これは、腫瘍のマージンを引き出すだけでなく、単一の細胞レベルで高解像度の3D画像を達成するのに役立ちました。この方法で最も重要なステップは、マウスの膵臓画像化窓の巧みな移植です。蛍光マウス細胞と抗体プローブの他の組み合わせを用いて、近くの細胞と小島またはキャンセル細胞の動的相互作用を同定することができた。この方法は膵臓癌の様々な病態や微小環境における膵島の変化をその場で探求する者によって広く応用できる。
