췌장은 소화관에 있는 외분 염 기능 및 혈 류로 호르몬을 분비의 내 분 비 기능을 가진 복 기관. 췌장의 고해상도 세포 화상 진찰은 췌장염, 췌장암 및 당뇨병 mellitus1을포함하여 췌장을 관련시키는 각종 질병의 병리생리학을 드러낼 수 있었습니다. 컴퓨터 단층 촬영, 자기 분해영상 및 초음파와 같은 기존의 진단 영상 도구는 임상 분야1,2에서널리 이용된다. 그러나, 이러한 이미징 양식은 구조적 또는 해부학적 변화만을 시각화하는 것으로 제한되며, 세포 또는 분자 수준에서의 변화는 결정될 수 없다. 인간내 당뇨병 또는 췌장암의 분자 변화가 진단3,4전에 10년 이상 시작할 수 있다는 점을 감안할 때, 잠기 기간 동안 분자 전환에서 췌장질환의 검출은 조기 진단과 적시개입을 제공할 가능성이 있다. 따라서, 해상도의 한계를 극복하고 기능에 대한 귀중한 통찰력을 제공하는 이미징은 당뇨병의 진행 중에 췌장암의 조기 진단 또는 섬 변경의 고급 식별을 제공함으로써 현저하게 주의를 얻을 것이다5.

살아있는 동물의 현미경 해상도 수준에서 병리학을 드러내는 인트라바이탈 이미징은 최근 큰 관심을 받고 있다13. 생체 내 이미징 방법 중, 마우스의 복부에 창을 이식하는 복부 이미징창(14)의개발은 새로운 발견(즉, 초기 간 전이의 사전 미세메타증 단계및 장 상피16)에서줄기 세포 유지 장치의 메커니즘을 발견할 수 있게 하였다. 복부 화상 진찰 창은 귀중한 결과를 제공하지만, 췌장에 대한이 창의 응용 프로그램과 췌장을 포함하는 질병에 기초한 그 결과 중요한 화상 진찰 연구는 광범위하게 조사되지 않았습니다.
인간 췌장의 잘 정의된 고체 장기 특성과는 달리, 마우스의 췌장은 확산분포된 연조직형구조(17)이다. 따라서 연동 및 호흡을 포함한 생리적 움직임에 끊임없이 영향을 받습니다. 췌장에 대한 복부 이미징 창의 적용에 대한 이전 연구는 배회가 배변(18)에의해 유도된 운동 유물로 인해 발생한다는 것을 입증했다. 심한 흐림은 마이크로 스케일 구조의 시각화 및 식별을 방해하는 결과 평균 이미지에서 관찰되었다.
본명, 췌장과 관련된 질병의 종방향 세포 수준 사건을 조사하기 위해19,20의 인포탈 현미경 검사법과 결합된 새로운 지지 기체 통합 췌장 내 인트라바이탈 이미징 창의 사용을 설명한다. 이전 연구18의방법론에 대한 상세한 설명 외에도 췌장과 관련된 다양한 질병에 대한 췌장 이미징 창의 확장된 적용이 이 논문에서 해결될 것이다. 이 프로토콜에서는 맞춤형 비디오 속도 레이저 스캐닝 공초점 현미경 시스템을 중요한 현미경 시스템으로 활용했습니다. 4개의 레이저 모듈(405, 488, 561, 640 nm의 파장)이 여기원으로 활용되었고, 밴드패스 필터(BPF1: FF01:FF01-442/46)를 통해 광증관(PMT)에 의해 4개의 방출 신호 채널이 검출되었다.
