전기영동은 생명 과학 실험실에서 생체 분자의 분리 및 분석을 위해 선호되는 방법이다. TGGE는 크기가 같지만 서열이 다른 이중 가닥 DNA 단편의 분리를 가능하게 합니다. 이 기술은 DNA 단편의 용융 온도에서의 서열 관련 차이 및 선형 온도 구배 9,10을 갖는 다공성 겔에서의 이동성의 후속 변화에 의존합니다. DNA 단편의 용융은 특정 용융 프로파일을 생성한다. 일단 가장 낮은 용융 온도를 갖는 도메인이 겔 내의 특정 위치에서 상응하는 온도에 도달하면, 나선형 구조로부터 부분적으로 용융된 구조로의 전이가 발생하고, 분자의 이동이 실질적으로 중단될 것이다. 따라서 TGGE는 DNA 단편의 이동성 (크기 정보)과 온도 유도 구조적 전이 (서열 의존 정보)를 모두 활용하여 DNA 단편의 특성화에 대한 강력한 접근 방식을 제공합니다. DNA 분자의 세 가지 구조적 전이에 해당하는 TGGE 용융 패턴의 특징점은 가닥 초기-해리점, 가닥 중간-해리점, 및 가닥 말단-해리점이다(그림 1). 도메인 내의 서열 변이, 심지어 단일 염기 차이조차도 용융 온도에 영향을 미치므로, 상이한 서열을 갖는 분자는 TGGE 분석에서 이산 용융 (변성) 패턴을 보여줄 것이다. 따라서 TGGE는 게놈 RNA에서 SNV를 분석하는 데 사용할 수 있으며 RNA 편집을 탐지하는 데 매우 중요한 고처리량 방법이 될 수 있습니다. 이러한 높은 처리량 이득은 전통적인 겔 전기영동 기반 TGGE가 사용될 때 손실된다. 그러나, microTGGE(μTGGE)로 명명된 TGGE의 소형화된 버전은 겔 전기영동 시간을 단축하고 생산성(11)의 100배 증가로 분석을 가속화하는데 사용될 수 있다. μTGGE 방법의 단순성과 소형화는 현장에서 적용 가능하고 핸드헬드이며 저렴한 겔 전기영동 시스템인 PalmPAGE12의 도입으로 개선되었습니다.

A-to-I RNA 편집 타입의 경우, 변형된 G 염기를 갖는 편집된 샘플은 원래의 A 염기를 갖는 편집되지 않은 샘플보다 가닥 말단 융점에서 더 긴 용융 패턴을 표시하였다. 역방향 U-to-C RNA 편집 유형의 경우, 변형된 C 염기를 갖는 첫 번째 유전자의 편집된 샘플은 원래의 U 염기를 갖는 편집되지 않은 샘플보다 가닥의 초기 및 말단 융점 사이에 더 긴 용융 패턴을 보였다.그러나, 다른 유전자에 대해서도 유사한 패턴이 관찰되지 않았다.
