De synthetische benadering die in dit artikel wordt beschreven, biedt een methode voor het synthetiseren van cyclische peptiden met behulp van Cys en Met in de peptidesequentie, waarbij de elementaire lineaire peptiden worden geconstrueerd door gemeenschappelijke vaste-fase peptidesynthesetechnieken. Voor de bisalkylering van cyclische peptiden tussen Cys en Met kan de hele synthetische route worden onderverdeeld in drie belangrijke processen: de bescherming van Cys op de hars, de koppeling van de linker en de cycliserin...

Eiwit-eiwit interacties (PPI's)1 spelen een cruciale rol in onderzoek en ontwikkeling van geneesmiddelen. Het construeren van gestabiliseerde peptiden met een vaste conformatie met chemische middelen is een van de belangrijkste methoden voor het ontwikkelen van mimetische motieven van PPI's2. Tot op heden zijn verschillende cyclische peptiden die zich richten op PPI's ontwikkeld voor klinisch gebruik3. De meeste peptiden zijn beperkt tot een α-helix conformatie om de conformationele entropie te verminderen en de metabole stabiliteit, doelbindingsaffiniteit en celpermeabiliteit te verbeteren 4,5. In de afgelopen 2 decennia zijn de zijketens van Cys 6,7, lysine 8,9, tryptofaan10, arginine11 en Met 12,13 in onnatuurlijke aminozuren ingebracht om het peptide in een cyclische conformatie te fixeren. Dergelijke cyclische peptiden kunnen zich richten op een unieke chemische ruimte of speciale plaatsen, waardoor een covalente reactie wordt geactiveerd om eiwit-peptide covalente binding 14,15,16,17 te vormen. In een recent rapport van Yu et al. werd een chloroacetamide verankerd in het domein van peptideliganden, waardoor een covalente conjugatiereactie met een uitstekende eiwitspecificiteitwerd gewaarborgd 18. Bovendien werden elektrofiele kernkoppen, zoals acrylamide en arylsulfonylfluoride (ArSO2F), verder opgenomen in peptiden door Walensky et al.19 om gestabiliseerde peptide covalente remmers te vormen en het antitumoreffect van peptideremmers te verbeteren. Daarom is het erg belangrijk om een extra functionele groep te introduceren om eiwit-peptide liganden covalent te modificeren20. Deze groepen reageren niet alleen met eiwitten aan de zijketen, maar stabiliseren ook de secundaire structuur van het peptide21. De toepassing van covalent gemodificeerde eiwitten geïnduceerd door peptideliganden is echter beperkt vanwege de gecompliceerde synthetische route en de niet-specifieke binding van de chemische groepen22,23. Effectieve strategieën voor de synthese van cyclische peptiden zijn daarom dringend nodig.
Geïnspireerd door de veelzijdige strategieën van cyclische peptiden 2,24,25,26, probeert dit protocol een eenvoudige en efficiënte methode te ontwikkelen voor het stabiliseren van peptiden. Bovendien merkten we op dat de zijketengroep van een stabiel peptide covalent kon reageren met een doeleiwit wanneer het ruimtelijk dicht bij de peptideliganden lag. Het gebrek aan chemisch gemodificeerde Met werd in 2013 door de Deming-groep opgevuld door een nieuwe methode te ontwikkelen voor het produceren van selectief gemodificeerd peptide methionine27. Op basis van deze achtergrond richtten de Shi et al. zich op de ontwikkeling van de ringsluiting van zijketens om een sulfoniumzoutcentrum te vormen. Wanneer het peptide ligand zich combineert met het doeleiwit, reageert de sulfoniumzoutgroep covalent met het ruimtelijk nauwe Cys-eiwit. In de afgelopen jaren hebben de Shi et al. een nieuwe methode ontworpen voor het stabiliseren van cyclisch peptide28. Het sulfoniumzout op het cyclische peptide werd gereduceerd door een reductiemiddel met een sulfhydrylgroep die reversibel werd gereduceerd tot Met. De reactie had echter een lage efficiëntie, wat schadelijk was voor latere biologische toepassingsstudies. In de huidige studie werd een Met-Cys en propargylbromide-Cys ringsluitingsreactie ontworpen, waarbij een enkel sulfoniumzout achterbleef aan de zijketen van het cyclische peptide. Het sulfoniumzout fungeerde als een nieuwe kernkop die covalent reageerde met het eiwit Cys onder ruimtelijke nabijheid. Kortom, een Cys en Met gemuteerd peptide werd gecyclizeerd door intramoleculaire alkylering, wat resulteerde in het genereren van een on-tether sulfoniumcentrum. In dit proces was de vorming van een zijketenbrug van cruciaal belang voor cyclische peptiden. Over het algemeen beschrijft dit protocol een gedetailleerde op sulfonium gebaseerde peptidecyclisatie die wordt bereikt met behulp van eenvoudige reactieomstandigheden en -bewerkingen. Het doel is om een mogelijke methode te ontwikkelen voor verdere brede biologische toepassingen.

In de afgelopen jaren hebben cyclische peptiden steeds meer aandacht getrokken op het gebied van medicijnontdekking vanwege hun uitstekende biologische activiteiten, en als gevolg daarvan worden ze nu klinisch gebruikt. Het is daarom van cruciaal belang om effectieve strategieën te zoeken voor het synthetiseren van cyclische peptiden om hun toepassing op het gebied van medicijnontdekking te bevorderen. Dit artikel rapporteert een gedetailleerd protocol voor de efficiënte synthese van cyclische peptiden met behulp van on-resin of intramoleculaire (intermoleculaire) bisalkylering. Met behulp van dit protocol werden lineaire peptiden gesynthetiseerd door gebruik te maken van vaste-fase peptidesynthese met cysteïne (Cys) en methionine (Met) die tegelijkertijd aan de hars werden gekoppeld. Verder werden cyclische peptiden gesynthetiseerd via bisalkylering tussen Met en Cys met behulp van een instelbare tether en een on-tether sulfoniumcentrum. De hele synthetische route kan worden onderverdeeld in drie belangrijke processen: de bescherming van Cys op de hars, de koppeling van de linker en de cyclisering tussen Cys en Met in een trifluorazijnzuur (TFA) splitsingsoplossing. Bovendien, geïnspireerd door de reactiviteit van het sulfoniumcentrum, werd een propargylgroep aan de Met gehecht om thiol-yne-toevoeging te activeren en een cyclisch peptide te vormen. Daarna werden de ruwe peptiden gedroogd en opgelost in acetonitril, gescheiden en vervolgens gezuiverd door high-performance vloeistofchromatografie (HPLC). Het molecuulgewicht van het cyclische peptide werd bevestigd door vloeistofchromatografie-massaspectrometrie (LC-MS), en de stabiliteit van de cyclische peptidecombinatie met het reductant werd verder bevestigd met behulp van HPLC. Bovendien werd de chemische verschuiving in het cyclische peptide geanalyseerd door 1H nucleaire magnetische resonantie (1H NMR) spectra. Over het algemeen was dit protocol gericht op het vaststellen van een effectieve strategie voor het synthetiseren van cyclische peptiden.

Alle lineaire peptiden werden gesynthetiseerd op Rink-amide MBHA-hars door standaard handmatige Fmoc vaste-fasesynthese. Een model cyclisch hexapeptide (Ac (cyclo-I)-WMAAAC-NH2) werd geconstrueerd zoals beschreven in figuur 5A. In dit protocol werd het sulfoniumzout geconstrueerd dat kon fungeren als een nieuwe hand die reageerde met het eiwit cysteïne op de speciale nabijheid. Het voordeel van deze techniek was dat deze reactie snel en efficiënt was, en Met-katalysator vrij en ontwikkelde een eenvoudige en efficiënte methode voor het stabiliseren van peptiden. Bereid om te beginnen de N-terminale 9-fluorinyl-methyloxycarbonyl-deprotectie-oplossing van 20% piperidine of 50% morfoline en DMF in een bekerglas of kolf van 500 milliliter.Voeg vervolgens 10 milliliter van de deprotectieoplossing toe aan de kolom en bel stikstof in de oplossing gedurende 30 minuten of twee keer gedurende vijf minuten. De HPLC-sporen van de epimeren en het geconjugeerde product toonden de tijdsafhankelijke conversie tussen het cyclische peptide en zijn product. Het molecuulgewicht van cyclische peptiden gesynthetiseerd met behulp van een thiol-yne-type reactie werd bepaald door LCMS en het peptide werd geïsoleerd en gezuiverd door HPLC.De peptiden werden verder gekenmerkt door proton NMR en heteronucleaire single quantum coherente spectra, die de chemische verschuiving onthulden. Proton NMR spectra van de tijdsafhankelijke conversie tussen cyclisch peptide en dithiothreitol en deuteriumoxide werd verkregen om de stabiliteit van peptide te onderzoeken als gevolg van het openen van de sulfoniumring. De resultaten toonden aan dat er na 24 uur geen toevoegings- of ringopeningsproducten werden gevormd.Deze procedure heeft een effectieve strategie vastgesteld voor het synthetiseren van cyclische peptiden. De reactie zegt dat de selectiviteit ook is verbeterd door een bevestiging te vormen die het cyclisatiepeptide stabiliseert, wat aangeeft dat cyclische peptiden veelbelovende medicijnkatalysator waren.
