タンパク質間相互作用(PPI)1は、医薬品の研究開発において極めて重要な役割を果たします。化学的手段によって立体構造が固定された安定化ペプチドを構築することは、PPIの模倣モチーフを開発するための最も重要な方法の1つです2。現在までに、PPIを標的とするいくつかの環状ペプチドが臨床使用のために開発されています3。ほとんどのペプチドは、コンフォメーションエントロピーを低下させ、代謝安定性、標的結合親和性、および細胞透過性を改善するためにαらせんコンフォメーションに拘束されています4,5。過去20年間で、Cys 6,7、リジン8,9、トリプトファン10、アルギニン11、およびMet 12,13の側鎖が非天然アミノ酸に挿入され、ペプチドを環状立体配座に固定してきました。このような環状ペプチドは、固有の化学空間または特別な部位を標的とすることができ、それによって共有反応を誘発してタンパク質-ペプチド共有結合を形成する14、15、16、17。Yuらによる最近の報告では、クロロアセトアミドがペプチドリガンドのドメインに固定され、優れたタンパク質特異性を有する共有結合反応を保証した18。さらに、アクリルアミドやアリールスルホニルフルオリド(ArSO2F)などの求電子性弾頭は、Walenskyらによってさらにペプチドに組み込まれ、安定化ペプチド共有結合阻害剤を形成し、ペプチド阻害剤の抗腫瘍効果を改善しました。したがって、タンパク質−ペプチドリガンド20を共有結合的に修飾するために追加の官能基を導入することが非常に重要である。これらの基は、側鎖上のタンパク質と反応するだけでなく、ペプチド21の二次構造を安定化させる。しかしながら、ペプチドリガンドによって誘導される共有結合修飾タンパク質の適用は、複雑な合成経路および化学基の非特異的結合のために制限される22,23。したがって、環状ペプチドの合成のための効果的な戦略が緊急に必要とされている。
