혈관벽은 반응성 안정 상태로 존재하며, 이는 외부 자극(예: 압력 변화, 혈관 수축제)에 대한 반응성과 혈액 응고 및 염증성 세포 침투를 방지하는 일관된 비활성화 표면을 보장합니다1. 노화 및 생활 습관에 따른 자극과 직접적인 손상에 반응하여 혈관벽은 허혈성 뇌졸중 및 심근경색과 같은 일반적인 심혈관 질환(CVD)의 원인으로 알려진 재협착 및 죽상동맥경화증과 같은 리모델링 과정을 활성화한다2. 경피적 혈관재생술과 스텐트 삽입술과 같은 중재적 접근법은 혈관 질환의 진행된 증상을 해결하기 위해 사용할 수 있지만, 이는 추가적인 혈관 손상을 유발하여 종종 재발로 이어지는 것으로 알려져 있습니다. 또한 제한된 예방 및 초기 단계 솔루션만 사용할 수 있습니다. 혈관 벽 항상성을 유지하고 기능 장애를 유발하는 메커니즘을 이해하는 것이 새로운 치료법 개발의 핵심입니다3.
분자 생물학 및 조직 공학의 끊임없는 발전과 발전에도 불구하고 동물 연구는 혈관 생물학 연구의 중요한 구성 요소로 남아 있습니다. 생체 내 동물 연구는 혈관 항상성 및 병리학의 메커니즘에 대한 엄청난 통찰력을 제공했습니다. 그러나 이러한 절차는 비용이 많이 들고 처리량이 상대적으로 낮으며 상당한 윤리적 문제를 제기합니다. 또한, 작은 동물은 인간의 혈관 생리학을 잘 대표하지 못하며, 대규모 동물 실험은 훨씬 더 비싸고 더 많은 윤리적 고려 사항을 야기합니다 4,5. 급속한 고령화 인구를 위한 제약 및 의료 솔루션에 대한 수요가 증가함에 따라 동물 사용의 단점이 확대되어 결과의 재현성, 신뢰성 및 환자 치료로의 이전 가능성에 영향을 미치고 있습니다6.
체 외 시스템은 기본 기전을 연구할 수 있는 단순화된 플랫폼을 제공하지만, 혈관 질환 발병의 중요한 결정 요인인 전체 조직의 복잡성, 세포와 세포외 기질 간의 상호 작용, 기계적 힘을 요약하지 못한다7.
인위적으로 통제된 환경에서 유지되는 전체 조직에 대해 수행된 생체 외 연구는 in vivo 복잡성을 모방하는 동시에 상대적으로 높은 처리량의 조사를 가능하게 한다8. 배양 조건과 환경을 면밀히 제어할 수 있는 능력을 감안할 때, 체외 모델은 광범위한 복잡한 연구를 가능하게 하고 혈관 생물학에서 동물 절차의 사용을 줄이기 위한 적절한 대안을 제공합니다. 정적 혈관 고리 배양은 흥미로운 통찰력을 제공했지만 중요한 혈역학적 요소(9)를 통합하지 못했습니다. 실제로, 생체 외 혈관계 연구는 혈관 벽 내의 세포에 적용되는 많은 동적 힘과 관련된 특정 문제를 제기합니다. 발광 흐름, 난류, 전단 응력, 압력 및 벽 변형과 같은 자극은 조직 병태생리학에 상당한 영향을 미친다10,11,12.
관류 생물반응기는 혈관 항상성을 연구하고 부상 또는 혈류역학적 변화에 대응하여 리모델링하는 데 필수적이다13. 또한, 관류 배양은 조직 공학 혈관(TEBV)의 성숙 및 내구성을 개선하는 데 사용될 수 있으며, 혈관 이식편에 적합한 대안을 제공할 수 있다14.
상업적으로 이용 가능한 관류 생물 반응기는 유연성과 적응성이 제한적이며 비용이 많이 듭니다. 기존의 자체 개발 된 바이오 리액터 중 다수는 특수 제작 된 구성 요소 7,8,9,10,11,12의 제한된 설명과 사용할 수 없기 때문에 다른 실험실에서 복제하기가 어렵습니다. 이러한 한계를 극복하기 위해 우리는 최근 새로운 바이오리액터(EasyFlow)를 개발했는데, 이 바이오리액터는 생산이 경제적이고, 다양한 조직을 수용할 수 있으며, 다양한 연구 요구에 적응하기 위해 비교적 간단한 수정이 가능하다13. 인서트는 3D 프린팅되어 표준 50mL 원심분리 튜브의 뚜껑에 맞습니다. 모듈식 설계와 3D 프린팅 제조를 통해 다양한 실험실에서 접근 및 재현할 수 있을 뿐만 아니라 다양한 과학적 요구에 맞게 쉽게 수정할 수 있습니다. 이 프로토콜은 동맥 관류 설정에서 생물반응기 시스템의 조립 및 기본 작동을 설명합니다.
