Sener letter bevægelse ved at overføre kræfter fra muskel til knogle. Selvom seneskade er almindelig, er disse ret vanskelige at behandle, og resultatet for patienterne er ofte dårligt. I øjeblikket involverer alle behandlinger af seneskade en form for fysioterapi, og det afspejler det faktum, at mekaniske kræfter spiller en så central rolle i senebiologien.
Gode eksperimentelle modeller til undersøgelse af seneskader og reparation findes ikke rigtig, så mit laboratorium udvikler aktivt nye modeller, der bedre kan fange vigtige træk ved senefysiologi og patofysiologi. I tidligere undersøgelser kunne vi vise, at senekernen, som udgør den bærende del af senen, i sig selv har en meget begrænset reparationskapacitet. Kombineret med anden forskning på området antog vi, at en skadet kerne ville rekruttere celler fra det ydre senerum for at hjælpe det med at helbrede.
Vævsmanipuleret senemodelsystem kan give et belastbart 3D-miljø, men matcher ikke forviklingerne i en in vivo exocellulær matrix. Det gør Explant-modelsystemer, men de er ofte vanskelige at holde i live og mekanisk belaste over længere perioder eller mangler det ydre rum, der er centralt for reparationsprocesserne. Vores unikke modelsystem kombinerer fordelene ved marine haleseneafledte kerneeksplanter med fordelene ved 3D-hydrogelbasesystemer.
Det giver en lastbar, in vivo-lignende kernematrix sammen med et kunstigt ydre rum. Dens sammensætning kan indstilles til forskningshypotesen og den biomimetiske tværfaglige barriere mellem de to. Vores hybridhydrogel explant assembloids er i en førsteklasses position til at studere senekernebiologi, matrixstrukturfunktionsinteraktioner og tværfaglige interaktioner mellem specifikke cellepopulationer i et fint justerbart mikromiljø.
Opdagelser fra de undersøgelser, der udføres med dette system, vil guide in vivo-forskning og behandlingsudvikling.
