Pezen vergemakkelijken beweging door krachten over te brengen van spieren naar botten. Hoewel peesblessures vaak voorkomen, zijn deze vrij moeilijk te behandelen en is de uitkomst voor patiënten vaak slecht. Momenteel omvatten alle behandelingen van peesblessures een vorm van fysiotherapie, en dit weerspiegelt het feit dat mechanische krachten zo'n centrale rol spelen in de peesbiologie.
Goede experimentele modellen voor het bestuderen van peesschade en -herstel bestaan niet echt, dus mijn lab is actief bezig met het ontwikkelen van nieuwe modellen die belangrijke kenmerken van peesfysiologie en pathofysiologie beter kunnen vastleggen. In eerdere studies konden we aantonen dat de peeskern, die het dragende deel van de pees vertegenwoordigt, van zichzelf een zeer beperkte herstelcapaciteit heeft. In combinatie met ander onderzoek in het veld, veronderstelden we dat een gewonde kern cellen uit het extrinsieke peescompartiment zou rekruteren om het te helpen genezen.
Weefsel-gemanipuleerd peesmodelsysteem kan een belastbare 3D-omgeving bieden, maar komt niet overeen met de fijne kneepjes van een in vivo exocellulaire matrix. Explant-modelsystemen doen dat wel, maar ze zijn vaak moeilijk in leven te houden en mechanisch te belasten over langere perioden of missen het extrinsieke compartiment dat centraal staat voor de reparatieprocessen. Ons unieke modelsysteem combineert de voordelen van explantaten van mariene staartpees met die van 3D-hydrogelbasissystemen.
Het biedt een belastbare, in vivo-achtige kernmatrix, naast een kunstmatig extrinsiek compartiment. De samenstelling ervan kan worden afgestemd op de onderzoekshypothese en de biomimetische dwarscompartimentele barrière tussen de twee. Onze hybride hydrogel explantatie-assembloids bevinden zich in een uitstekende positie om de biologie van de peeskern, interacties tussen matrixstructuurfuncties en cross-compartimentele interacties tussen specifieke celpopulaties in een fijn afstembare micro-omgeving te bestuderen.
Ontdekkingen uit de studies die met dit systeem zijn uitgevoerd, zullen als leidraad dienen voor in vivo onderzoek en ontwikkeling van behandelingen.
