Sehnen erleichtern die Bewegung, indem sie Kräfte vom Muskel auf den Knochen übertragen. Obwohl Sehnenverletzungen häufig sind, sind diese ziemlich schwer zu behandeln und das Ergebnis für die Patienten ist oft schlecht. Derzeit beinhalten alle Behandlungen von Sehnenverletzungen eine Art Physiotherapie, und dies spiegelt die Tatsache wider, dass mechanische Kräfte eine so zentrale Rolle in der Sehnenbiologie spielen.
Gute experimentelle Modelle zur Untersuchung von Sehnenschäden und -reparaturen gibt es nicht wirklich, daher entwickelt mein Labor aktiv neue Modelle, die wichtige Merkmale der Sehnenphysiologie und Pathophysiologie besser erfassen können. In früheren Studien konnten wir zeigen, dass der Sehnenkern, der den tragenden Teil der Sehne darstellt, an sich eine sehr begrenzte Reparaturkapazität hat. In Kombination mit anderen Forschungen auf diesem Gebiet stellten wir die Hypothese auf, dass ein verletzter Kern Zellen aus dem extrinsischen Sehnenkompartiment rekrutieren würde, um ihm bei der Heilung zu helfen.
Tissue-Engineering-Sehnenmodellsysteme bieten zwar eine belastbare 3D-Umgebung, entsprechen aber nicht den Feinheiten einer exozellulären In-vivo-Matrix. Explantat-Modellsysteme tun dies, aber sie sind oft schwer am Leben zu erhalten und über längere Zeiträume mechanisch zu belasten oder es fehlt ihnen das extrinsische Kompartiment, das für die Reparaturprozesse zentral ist. Unser einzigartiges Modellsystem kombiniert die Vorteile von aus marinen Schwanzsehnen gewonnenen Kernexplantaten mit denen von 3D-Hydrogel-Basissystemen.
Es bietet eine belastbare, in vivo-ähnliche Kernmatrix sowie ein künstliches extrinsisches Kompartiment. Seine Zusammensetzung kann auf die Forschungshypothese und die biomimetische Kreuzkompartimentbarriere zwischen den beiden abgestimmt werden. Unsere Hybrid-Hydrogel-Explantat-Assembloide sind in einer erstklassigen Position, um die Biologie des Sehnenkerns, die Wechselwirkungen zwischen den Funktionen der Matrixstruktur und die kompartimentären Wechselwirkungen zwischen bestimmten Zellpopulationen in einer fein abstimmbaren Mikroumgebung zu untersuchen.
Die Entdeckungen aus den mit diesem System durchgeführten Studien werden die In-vivo-Forschung und Behandlungsentwicklung leiten.
