기존의 2D 배양 모델에 비해 in vivo와 유사한 구조적 및 형태학적 특성을 더 많이 제공하는 3D in vitro 배양 모델은 조직 공학, 질병 모델링 및 약물 스크리닝과 같은 다양한 생물의학 응용 분야에서 유망한 시스템으로 인정받고 있습니다 1,2,3. 3D 배양 모델의 한 유형인 세포 스페로이드는 일반적으로 세포 응집을 의미하며, 향상된 세포-세포 및 세포-기질 상호 작용을 특징으로 하는 3D 구상체 구조를 생성합니다 4,5,6. 따라서 세포 스페로이드 제조는 다양한 생물학적 연구를 가능하게 하는 강력한 도구가 되었습니다.
스페로이드를 얻기 위해 행잉 드롭(hanging drop)7, 비접착 플레이트(non-adhesive plate)8 또는 마이크로웰 장치(microwell device)9를 포함한 다양한 기술들이 개발되었다. 원칙적으로, 이러한 방법은 일반적으로 세포와 기판 사이의 상호 작용을 최소화하면서 중력과 같은 물리적 힘을 이용하여 세포 조립을 용이하게 합니다. 그러나 종종 노동 집약적인 공정이 포함되고 생산성이 낮으며 스페로이드 크기10,11을 제어하는 데 어려움이 있습니다. 중요한 것은 원하는 크기와 균일성을 가진 스페로이드를 충분한 양으로 생산하는 것이 특정 생물학적 응용 분야를 충족하는 데 가장 중요하다는 것입니다. 상술한 방법들과는 대조적으로, 음파는, 외력-구동 기술(12,13,14)의 한 유형으로서, 외력(15,16,17,18)을 통해 세포 응집을 강화하는 원리에 기초하여, 높은 품질과 처리량을 갖는 세포 스페로이드의 대량 제조에 대한 가능성을 보여주었다 . 전자기 또는 자기력과 달리 음향 기반 세포 조작 기술은 비침습적이고 표지가 없어 우수한 생체 적합성으로 스페로이드 형성을 가능하게 합니다19,20.
일반적으로, 스탠딩 표면 음향파(SAW) 및 벌크 음파(BOW) 기반 장치는 대응하는 스탠딩 음향장(21,22,23)에 의해 생성된 음향 노드(AN)를 활용하여 스페로이드를 생성하기 위해 개발되었다. 특히, BAW를 기반으로 하는 음향 조립 장치는 제조가 편리하고, 조작이 용이하며, 확장성이 우수하다는 장점이 있어 셀 스페로이드24,25 제조에 주목받고 있다. 우리는 최근에 높은 처리량을 가진 스페로이드를 생성할 수 있는 손쉬운 BAW 기반 음향 조립 장치를 개발했습니다26. 제안된 장치는 X/Y/Z 평면에 각각 배열된 3개의 지르콘산 납 티타네이트(PZT) 변환기가 있는 정사각형 폴리메틸 메타크릴레이트(PMMA) 챔버로 구성됩니다. 이러한 배열은 구동 셀 조립을 위한 공중 부양 음향 노드(LAN)의 3D 도트 어레이 패턴을 생성할 수 있습니다.
