Epileptic seizures cause severe and long-lasting events on the architecture of the brain, including neuronal cell death, accompanied neurogenesis, reactive gliosis, and mossy fiber sprouting. However, it remains uncertain whether these functional and anatomical alterations are associated with the development of hyperexcitability, or as inhibitory processes. Neurotrophic factors are probable mediators of these pathophysiological events. The present study was designed to clarify the role of various neurotrophic factors on the pilocarpine model of seizures. At 4 h following pilocarpine-induced seizures, expression of NGF, BDNF, HB-EGF, and FGF-2 increased only in the mice manifesting tonic-clonic convulsions and not in mice without seizures. NT-3 expression decreased in pilocarpine-treated mice experiencing seizures, tonic-clonic or not, compared to mice with no seizures. Neuronal cell damage, which was evident by Fluoro-Jade B staining, was observed within 24 h in the mice exhibiting tonic-clonic seizures, followed by an increase in the number of BrdU-positive cells and glial cells, which were evident after 2 days. None of these pathophysiological changes occurred in the mice which showed no seizures, although they were injected with pilocarpine, nor in the activated epilepsy-prone EL mice, which experienced repeated severe seizures. Together, these results suggest that neuronal damage occurring in the brain of the mice manifesting tonic-clonic seizures is accompanied by neurogenesis. This sequence of events may be regulated through changes in expression of neurotrophic factors such as NGF, BDNF, HB-FGF, and NT-3.

Tonic-clonic seizures induce division of neuronal progenitor cells with concomitant changes in expression of neurotrophic factors in the brain of pilocarpine-treated mice.

Crises &eacute;pileptiques provoquent des &eacute;v&eacute;nements graves et durables sur l&#39;architecture du cerveau, y compris la neurogen&egrave;se accompagn&eacute;e, mort neuronale, gliose r&eacute;active et germination des fibres moussues. Toutefois, il demeure incertain si ces alt&eacute;rations fonctionnelles et anatomiques sont associ&eacute;es au d&eacute;veloppement d&#39;une hyperexcitabilit&eacute; ou processus inhibiteurs. Facteurs neurotrophiques sont des m&eacute;diateurs probables de ces manifestations physiopathologiques. La pr&eacute;sente &eacute;tude visait &agrave; clarifier le r&ocirc;le des diff&eacute;rents facteurs neurotrophiques sur le mod&egrave;le de la pilocarpine des saisies. 4 H apr&egrave;s les crises induites par pilocarpine, expression de NGF, BDNF, HB-EGF, et FGF-2 a augment&eacute; seulement chez les souris qui manifeste des convulsions tonico-cloniques et pas chez les souris sans convulsions. NT-3 expression a diminu&eacute; chez les souris trait&eacute;es pilocarpine touch&eacute;s par des crises tonico-cloniques ou non, par rapport aux souris &agrave; aucune saisie. Dommages des neurones, ce qui &eacute;tait &eacute;vident par coloration de Fluoro-Jade B, a &eacute;t&eacute; observ&eacute; dans les 24 heures chez les souris pr&eacute;sentant des convulsions tonico-cloniques, suivies d&#39;une augmentation du nombre de cellules positives BrdU et les cellules gliales, qui &eacute;taient visibles apr&egrave;s 2 jours. Aucun de ces changements pathophysiologiques s&#39;est produite chez les souris qui ont montr&eacute; aucune saisie, m&ecirc;me si elles ont &eacute;t&eacute; inject&eacute;s avec la pilocarpine, ni chez les souris activ&eacute;s dEL sujettes &agrave; l&#39;&eacute;pilepsie, qui a connu des crises graves r&eacute;p&eacute;t&eacute;s. Ensemble, ces r&eacute;sultats sugg&egrave;rent que des l&eacute;sions neuronales dans le cerveau des souris qui manifeste des crises tonico-cloniques g&eacute;n&eacute;ralis&eacute;es sont accompagn&eacute;e de la neurogen&egrave;se. Cette s&eacute;quence d&#39;&eacute;v&eacute;nements peut &ecirc;tre r&eacute;gl&eacute;e par le biais de changements dans l&#39;expression des facteurs neurotrophiques comme NGF, BDNF, HB-FGF et NT-3.

Crises tonico-cloniques g&eacute;n&eacute;ralis&eacute;es induisent la division des cellules souches neuronales avec des modifications concomitantes dans l&#39;expression des facteurs neurotrophiques dans le cerveau des souris trait&eacute;es pilocarpine.

Tonic-klonischen induzieren Bereich von neuronalen Vorl&#xE4;uferzellen unter gleichzeitiger Ver&#xE4;nderungen in der Expression von neurotrophen Faktoren im Gehirn von Pilocarpin-behandelten M&#xE4;usen.

Crisi epilettiche causano eventi gravi e di lunga durata sull&#39;architettura del cervello, tra cui morte delle cellule neuronali, accompagnata neurogenesi, gliosi reattiva e fibre muschiose che spuntano. Tuttavia, rimane incerto se queste alterazioni anatomiche e funzionali sono associate con lo sviluppo di ipereccitabilit&agrave;, o come processi inibitori. I fattori neurotrofici sono probabili mediatori di questi eventi fisiopatologici. Il presente studio &egrave; stato progettato per chiarire il ruolo dei vari fattori neurotrofici sul modello dei sequestri di pilocarpina. A 4 h dopo convulsioni indotte pilocarpina, espressione di NGF, BDNF, HB-EGF, FGF-2 maggiore solo in topi manifestare convulsioni tonico-cloniche e non nei topi senza convulsioni. NT-3 espressione ridotta nei topi trattati con pilocarpina verificano convulsioni, tonico-cloniche o non, rispetto a topi con nessuna crisi epilettiche. Danni delle cellule neuronali, che era evidenti dalla colorazione Fluoro-giada B, &egrave; stato osservato entro 24 h nei topi espositrici tonico-cloniche, seguite da un aumento del numero di cellule positive di BrdU e cellule gliali, che erano evidenti dopo 2 giorni. Nessuno di questi cambiamenti patofisiologici si &egrave; verificato nei topi che hanno mostrato nessuna crisi epilettiche, anche se essi sono stati iniettati con pilocarpina, n&eacute; nei topi EL inclini epilessia attivati, che ha sperimentato ripetute crisi epilettiche gravi. Insieme, questi risultati suggeriscono che danni neuronali che si verificano nel cervello dei topi manifestando crisi tonico-cloniche sono accompagnato da neurogenesi. Questa sequenza di eventi pu&ograve; essere regolata attraverso i cambiamenti nell&#39;espressione di fattori neurotrofici come NGF, BDNF, HB-FGF e NT-3.

Factores neurotr&oacute;ficos son mediadores probables de estos eventos patofisiol&oacute;gicos. El presente estudio fue dise&ntilde;ado para aclarar el papel de diversos factores neurotr&oacute;ficos en el modelo de pilocarpina de convulsiones. A las 4 h despu&eacute;s de las convulsiones inducidas por pilocarpina, expresi&oacute;n de NGF, BDNF, HB-EGF, y FGF-2 aument&oacute; s&oacute;lo en los ratones que manifiesta las convulsiones t&oacute;nico-cl&oacute;nicas y no en ratones sin convulsiones. New granule cells are continuously generated in the dentate gyrus of the adult hippocampus. During granule cell maturation, the mechanisms that differentiate new cells not only describe the degree of cell differentiation, but also crucially regulate the progression of cell differentiation. Here, we describe a gene, tryptophan 2,3-dioxygenase (TDO), whose expression distinguishes stem cells from more differentiated cells among the granule cells of the adult mouse dentate gyrus. The use of markers for proliferation, neural progenitors, and immature and mature granule cells indicated that TDO was expressed in mature cells and in some immature cells. In mice heterozygous for the alpha-isoform of calcium/calmodulin-dependent protein kinase II, in which dentate gyrus granule cells fail to mature normally, TDO immunoreactivity was substantially downregulated in the dentate gyrus granule cells. Moreover, a 5-bromo-2'-deoxyuridine labeling experiment revealed that new neurons began to express TDO between 2 and 4 wk after the neurons were generated, when the axons and dendrites of the granule cells developed and synaptogenesis occurred. These findings indicate that TDO might be required at a late-stage of granule cell development, such as during axonal and dendritic growth, synaptogenesis and its maturation.

A continuaci&#xF3;n, describimos un gen, el tript&#xF3;fano 2,3-dioxigenasa (TDO), cuya expresi&#xF3;n distingue a las c&#xE9;lulas madre de m&#xE1;s c&#xE9;lulas diferenciadas entre las c&#xE9;lulas granulares de la circunvoluci&#xF3;n dentada del rat&#xF3;n adulto. El uso de marcadores de proliferaci&#xF3;n, progenitores neuronales y c&#xE9;lulas granulosas inmaduras y maduras indic&#xF3; que TDO se expresa en las c&#xE9;lulas maduras y en algunas c&#xE9;lulas inmaduras. En los ratones heterocigotos para la isoforma alfa de calcio / calmodulina dependiente de la prote&#xED;na quinasa II, en el que las c&#xE9;lulas granulares dentadas gyrus no maduran normalmente, inmunoreactividad TDO fue sustancialmente regulados a la baja en las c&#xE9;lulas granulares del gyrus dentado. Adem&#xE1;s, un experimento de etiquetado de 5-bromo-2&#x26;#39;-desoxiuridina revel&#xF3; que las nuevas neuronas comenzaron a expresar TDO entre 2 y 4 semanas despu&#xE9;s de las neuronas se generaron, cuando los axones y dendritas de las c&#xE9;lulas granulares desarrollado y producido sinaptog&#xE9;nesis. The dentate gyrus produces new granule neurons throughout adulthood in mammals from rodents to humans. During granule cell maturation, defined markers are expressed in a highly regulated sequential process, which is necessary for directed neuronal differentiation. In the present study, we show that Î±-amino-3-hydroxy-5-methy-4-isoxazole propionate (AMPA) receptor subunits GluR1 and GluR2 are expressed in differentiated granule cells, but not in stem cells, in neonatal, and adult dentate gyrus. Using markers for neural progenitors, immature and mature granule cells, we found that GluR1 and GluR2 were expressed mainly in mature cells and in some immature cells. A time-course analysis of 5-bromo-2'-deoxyuridine staining revealed that granule cells express GluR1 around 3â€‰weeks after being generated. In mice heterozygous for the alpha-isoform of calcium/calmodulin-dependent protein kinase II, a putative animal model of schizophrenia and bipolar disorder in which dentate gyrus granule cells fail to mature normally, GluR1 and GluR2 immunoreactivities were substantially downregulated in the dentate gyrus granule cells. In the granule cells of mutant mice, the expression of both presynaptic and postsynaptic markers was decreased, suggesting that GluR1 and GluR2 are also associated with synaptic maturation. Moreover, GluR1 and GluR2 were also expressed in mature granule cells of the neonatal dentate gyrus. Taken together, these findings indicate that GluR1 and GluR2 expression closely correlates with the neuronal maturation state, and that GluR1 and GluR2 are useful markers for mature granule cells in the dentate gyrus.

El giro dentado produce nuevas neuronas granulares en la edad adulta en los mam&#xED;feros de los roedores a los humanos. Durante la maduraci&#xF3;n de c&#xE9;lulas granulares, marcadores definidos se expresan en un proceso altamente regulado secuencial, que es necesaria para la diferenciaci&#xF3;n neuronal dirigida. En el presente estudio, se muestra que los &#x3B1;-amino-3-hidroxi-5-metil-4-isoxazol propionato (AMPA) y subunidades del receptor GluR1 GluR2 se expresan en c&#xE9;lulas granulares diferenciados, pero no en c&#xE9;lulas madre, en neonatal y adulto giro dentado. Uso de marcadores de c&#xE9;lulas progenitoras neurales inmaduras, y las c&#xE9;lulas maduras de gr&#xE1;nulos, se encontr&#xF3; que GluR1 y GluR2 se expresa principalmente en c&#xE9;lulas maduras y en algunas c&#xE9;lulas inmaduras. Un an&#xE1;lisis de tiempo-por supuesto de la tinci&#xF3;n de 5-bromo-2&#x26;#39;-desoxiuridina revel&#xF3; que las c&#xE9;lulas granulares expresar GluR1 alrededor de 3 semanas despu&#xE9;s de que se generan.
