1. 创建重组矢量
<ol><li>识别150 bp长侧翼的同源区域（称为左右臂），需要N-或C-最终标记感兴趣的病毒基因（见图1b）。</li>

</ol>2. 重组病毒生成
<ol><li>根据 图2，在无血清介质中感染 BS-C-1 细胞的单层细胞，在多种感染 （MOI） > 1。</li>
	<li>感染后1小时，使用Dulbecco的改良鹰介质（DMEM）进行抢救细胞，并在无血清介质中以3：1的比例与重组载体质粒和转染试剂的混合物发生变异。</li>
