In mobile opportunistic networks, the social relationship among nodes has an important impact on data transmission efficiency. Motivated by the strong share ability of "circles of friends" in communication networks such as Facebook, Twitter, Wechat and so on, we take a real-life example to show that social relationships among nodes consist of explicit and implicit parts. The explicit part comes from direct contact among nodes, and the implicit part can be measured through the "circles of friends". We present the definitions of explicit and implicit social relationships between two nodes, adaptive weights of explicit and implicit parts are given according to the contact feature of nodes, and the distributed mechanism is designed to construct the "circles of friends" of nodes, which is used for the calculation of the implicit part of social relationship between nodes. Based on effective measurement of social relationships, we propose a social-based clustering and routing scheme, in which each node selects the nodes with close social relationships to form a local cluster, and the self-control method is used to keep all cluster members always having close relationships with each other. A cluster-based message forwarding mechanism is designed for opportunistic routing, in which each node only forwards the copy of the message to nodes with the destination node as a member of the local cluster. Simulation results show that the proposed social-based clustering and routing outperforms the other classic routing algorithms.

Effective Social Relationship Measurement and Cluster Based Routing in Mobile Opportunistic Networks.

Remote clouds are gradually unable to achieve ultra-low latency to meet the requirements of mobile users because of the intolerable long distance between remote clouds and mobile users and the network congestion caused by the tremendous number of users. Mobile edge computing, a new paradigm, has been proposed to mitigate aforementioned effects. Existing studies mostly assume the edge servers have been deployed properly and they just pay attention to how to minimize the delay between edge servers and mobile users. In this paper, considering the practical environment, we investigate how to deploy edge servers effectively and economically in wireless metropolitan area networks. Thus, we address the problem of minimizing the number of edge servers while ensuring some QoS requirements. Aiming at more consistence with a generalized condition, we extend the definition of the dominating set, and transform the addressed problem into the minimum dominating set problem in graph theory. In addition, two conditions are considered for the capacities of edge servers: one is that the capacities of edge servers can be configured on demand, and the other is that all the edge servers have the same capacities. For the on-demand condition, a greedy based algorithm is proposed to find the solution, and the key idea is to iteratively choose nodes that can connect as many other nodes as possible under the delay, degree and cluster size constraints. Furthermore, a simulated annealing based approach is given for global optimization. For the second condition, a greedy based algorithm is also proposed to satisfy the capacity constraint of edge servers and minimize the number of edge servers simultaneously. The simulation results show that the proposed algorithms are feasible.

Cost-Effective Edge Server Placement in Wireless Metropolitan Area Networks.

A complete understanding of the structural and functional potential of RNA requires understanding of chemical modifications and non-canonical bases; this in turn requires advances in current sequencing methods to be able to sequence not only canonical ribonucleotides, but at the same time directly sequence these non-standard moieties. Here, we present the first direct and modification type-independent RNA sequencing method via introduction of a 2-dimensional hydrophobic end-labeling strategy into traditional mass spectrometry-based sequencing (2D HELS MS Seq) to allow de novo sequencing of RNA mixtures and enhance sample usage efficiency. Our method can directly read out the complete sequence, while identifying, locating, and quantifying base modifications accurately in both single and mixed RNA samples containing multiple different modifications at single-base resolution. Our method can also quantify stoichiometry/percentage of modified RNA versus its canonical counterpart RNA, simulating a real biological sample where modifications exist but may not be 100% at a particular site in the RNA. This method is a critical step towards fully sequencing real complex cellular RNA samples of any type and containing any modification type and can also be used in the quality control of modified therapeutic RNAs.

A general LC-MS-based RNA sequencing method for direct analysis of multiple-base modifications in RNA mixtures.

The continuous data-flow application in the IoT integrates the functions of fog, edge, and cloud computing. Its typical paradigm is the E-Health system. Like other IoT applications, the energy consumption optimization of IoT devices in continuous data-flow applications is a challenging problem. Since the anomalous nodes in the network will cause the increase of energy consumption, it is necessary to make continuous data flows bypass these nodes as much as possible. At present, the existing research work related to the performance of continuous data-flow is often optimized from system architecture design and deployment. In this paper, a mathematical programming method is proposed for the first time to optimize the runtime performance of continuous data flow applications. A lightweight anomaly detection method is proposed to evaluate the reliability of nodes. Then the node reliability is input into the optimization algorithm to estimate the task latency. The latency-aware energy consumption optimization for continuous data-flow is modeled as a mixed integer nonlinear programming problem. A block coordinate descend-based max-flow algorithm is proposed to solve this problem. Based on the real-life datasets, the numerical simulation is carried out. The simulation results show that the proposed strategy has better performance than the benchmark strategy.

Anomaly Detection Based Latency-Aware Energy Consumption Optimization For IoT Data-Flow Services.

Post-transcriptional modifications are intrinsic to RNA structure and function. However, methods to sequence RNA typically require a cDNA intermediate and are either not able to sequence these modifications or are tailored to sequence one specific nucleotide modification only. Interestingly, some of these modifications occur with <100% frequency at their particular sites, and site-specific quantification of their stoichiometries is another challenge. Here, we report a direct method for sequencing tRNA without cDNA by integrating a two-dimensional hydrophobic RNA end-labeling strategy with an anchor-based algorithm in mass spectrometry-based sequencing (2D-HELS-AA MS Seq). The entire tRNA was sequenced and the identity, location, and stoichiometry of all eleven different RNA modifications was determined, five of which were not 100% modified, including a 2'-O-methylated G (Gm) in the wobble anticodon position as well as an -dimethylguanosine (mG), a 7-methylguanosine (mG), a 1-methyladenosine (mA), and a wybutosine (Y), suggesting numerous post-transcriptional regulations in tRNA. Two truncated isoforms at the 3'-CCA tail of the tRNA (75 nt with a 3'-CC tail (80% abundance) and 74 nt with a 3'-C tail (3% abundance)) were identified in addition to the full-length 3'-CCA-tailed tRNA (76 nt, 17% abundance). We discovered a new isoform with A-G transitions/editing at the 44 and 45 positions in the tRNA variable loop, and discuss possible mechanisms related to the emergence and functions of the isoforms with these base transitions or editing. Our method revealed new isoforms, base modifications, and RNA editing as well as their stoichiometries in the tRNA that cannot be determined by current cDNA-based methods, opening new opportunities in the field of epitranscriptomics.
