Die histologische Techniken liefern wichtige visuelle Hinweise in Studium Unterschiede innerhalb der sekundären Zellwände auf organisatorischer und zellulärer Ebene. Verschiedene histologische Techniken wurden entwickelt und kann in der Literatur gefunden werden, aber diese Techniken kann schwierig und zeitaufwendig für Anfänger sein, weil sehr ausführliche Protokolle mit einfachen visuellen Anweisungen sind selten, wenn überhaupt vorhanden. Das Ziel dieser Studie ist es, einfache Richtlinien zur histologischen Färbung Techniken bieten, um qualitativ hochwertige Bilder zu erhalten. Unterteilen des Schaftgewebe ist der erste Schritt bei der Visualisierung von Zellwänden und Zellformen. Obwohl von Hand geschnitten Abschnitte sind kostengünstig und weniger Zeit für die Vorbereitung, die Verwendung eines Vibratom bietet Kontinuität undliefert qualitativ hochwertige Bilder. Darüber hinaus wird es unmöglich, irgendeine biologische Aktivität der Probe zu messen, wenn es in ein Harz eingebettet sind. Eine einfache Technik, die Agarose und eine hausgemachte Form beschäftigt ist hilfreich für die Einbettung von Stammzellen Gewebe, und es kann auch in anderen Anwendungen Dissektion der Weichteile erfordern, genutzt werden. Schneiden Gewebe über einen Vibratoms ist sehr präzise und homogen erzeugtschiedenen Abschnitten, die je nach dem Ziel der Studie kann dann mit verschiedenen Färbetechniken verwendet werden. Die einfachsten Methoden, um Lignin und andere Aromaten visualisieren beschäftigen ultraviolettem (UV) Licht. Anregung der aromatischen Moleküle auf durch UV-Licht ist eine alte Technik, aber es ist immer noch eine der schnellsten Ansätze zur Visualisierung von Lignin. Doch tatsächlich ist die UV-Visualisierung nicht ideal für die Erkennung Lignin, weil UV-Licht werden andere Aromaten zu erregen. 1-3: Lignin besteht hauptsächlich aus drei Bausteinen, die Monolignole (Coniferylalkohol, Sinapylalkohol und p-Cumarylalkohol hydroxycinnamyl Alkohole) gemacht. Phloroglucin Flecken können Hinweise auf den Umfang der in das Xylem, Fasern und Luftröhrengewebe 4 vorhanden Zimtaldehyde bereitzustellen. Phloroglucinol ist ein guter Indikator für die allgemeine Zimtaldehyde und kann zwischen Zimtaldehyde und anderen Aromaten unterscheiden. Die Sinapylalkohol Monomere nachgewiesen werden kannund differenziert durch die Verwendung Maule Fleck. Toluidinblau O eine polychromatische Farbstoff und hat die Fähigkeit, verschiedene Elemente der Zellwand in verschiedenen Farben färben daher 5,6. Der primäre Einsatz von Toluidinblau O ist es, Pektin und Lignin 5,6 zu erkennen. Der Vorteil der Verwendung von Toluidinblau O ist, dass viele Elemente der Zellwand kann in einem einzigen Schritt dargestellt werden. Sowohl Calcofluor weiß und Kongorot sind leicht zu verarbeiten und kann verwendet werden, um Zellulose zu visualisieren. Calcofluor weißen Flecken Cellulose, Callose und andere nicht-substituierten oder schwach substituierten β-Glucane 6-9, während Kongorot Flecken direkt an β-(1 → 4)-Glucane und insbesondere zu Cellulose 10,11. Das Ziel dieser Studie ist es, einfache Richtlinien für die Verwendung der oben genannten Färbetechniken, um qualitativ hochwertige Bilder von A zu erhalten bieten thaliana stammt.

Arabidopsis thaliana is a model organism commonly used to understand and manipulate various cellular processes in plants, and it has been used extensively in the study of secondary cell wall formation. Secondary cell wall deposition occurs after the primary cell wall is laid down, a process carried out exclusively by specialized cells such as those forming vessel and fiber tissues. Most secondary cell walls are composed of cellulose (40&#8211;50%), hemicellulose (25&#8211;30%), and lignin (20&#8211;30%). Several mutations affecting secondary cell wall biosynthesis have been isolated, and the corresponding mutants may or may not exhibit obvious biochemical composition changes or visual phenotypes since these mutations could be masked by compensatory responses. Staining procedures have historically been used to show differences on a cellular basis. These methods are exclusively visual means of analysis; nevertheless their role in rapid and critical analysis is of great importance. Congo red and calcofluor white are stains used to detect polysaccharides, whereas M&#228;ule and phloroglucinol are commonly used to determine differences in lignin, and toluidine blue O is used to differentially stain polysaccharides and lignin. The seemingly simple techniques of sectioning, staining, and imaging can be a challenge for beginners. Starting with sample preparation using the A. thaliana model, this study details the protocols of a variety of staining methodologies that can be easily implemented for observation of cell and tissue organization in secondary cell walls of plants.
