Glycoprotein fucosylation enables fringe-dependent modulation of signal transduction by Notch transmembrane receptors, contributes to selectin-dependent leukocyte trafficking, and is faulty in leukocyte adhesion deficiency (LAD) type II, also known as congenital disorder of glycosylation (CDG)-IIc, a rare human disorder characterized by psychomotor defects, developmental abnormalities, and leukocyte adhesion defects. We report here that mice with an induced null mutation in the FX locus, which encodes an enzyme in the de novo pathway for GDP-fucose synthesis, exhibit a virtually complete deficiency of cellular fucosylation, and variable frequency of intrauterine demise determined by parental FX genotype. Live-born FX(-/-) mice exhibit postnatal failure to thrive that is suppressed with a fucose-supplemented diet. FX(-/-) adults suffer from an extreme neutrophilia, myeloproliferation, and absence of leukocyte selectin ligand expression reminiscent of LAD-II/CDG-IIc. Contingent restoration of leukocyte and endothelial selectin ligand expression, general cellular fucosylation, and normal postnatal physiology is achieved by modulating dietary fucose to supply a salvage pathway for GDP-fucose synthesis. Conditional control of fucosylation in FX(-/-) mice identifies cellular fucosylation events as essential concomitants to fertility, early growth and development, and leukocyte adhesion.

R&eacute;tablissement &eacute;ventuel des leucocytes et expression de ligand de SELECTINE endoth&eacute;liale, g&eacute;n&eacute;rale fucosylation cellulaire et physiologie postnatale normale est obtenu en modulant le fucose alimentaire pour fournir une voie de sauvegarde pour la synth&egrave;se de GDP-fucose. Glicoproteina fucosylation permette di frangia-dipendente dalla modulazione della trasduzione del segnale dai recettori transmembrana Notch, contribuisce al traffico dei leucociti-selectina-dipendente ed &egrave; difettoso nel tipo di deficit (LAD) adesione leucocitaria II, noto anche come congenita della glicosilazione (CDG)-IIc, una rara patologia umana caratterizzata da anomalie dello sviluppo psicomotori difetti e adesione leucocitaria difetti. Segnaliamo qui che topi con una mutazione indotta null nel locus di FX, che codifica per un enzima nella via de novo per la sintesi del PIL-fucosio, mostrano un deficit praticamente completo di cellulare fucosylation e frequenza variabile di morte intrauterina determinato dal genotipo parentale di FX. Nati vivi FX(-/-) topi mostrano postnatale ritardo di crescita che &egrave; stata soppressa con una dieta integrata fucosio. FX(-/-) adulti soffrono di un&#39;estrema neutrofilia, myeloproliferation e assenza di leucociti selectina espressione del ligando ricorda LAD-II/CDG-IIc. Restauro contingente del leucocita e selectin ligand endoteliali espressione, generale fucosylation cellulare e normale fisiologia postnatale &egrave; ottenuto modulando fucosio dietetica per fornire un percorso di salvataggio per la sintesi del PIL-fucosio. Controllo condizionale di fucosylation nei topi FX(-/-) identifica eventi cellulari fucosylation come essenziale concomitanti alla fertilit&agrave;, crescita precoce e sviluppo e adesione dei leucociti.

The FX locus encodes an essential enzyme in the de novo pathway of GDP-fucose biosynthesis. Mice homozygous for a targeted mutation of the FX gene manifest a host of pleiotropic abnormalities including a lethal phenotype that is almost completely penetrant in heterozygous intercrosses on a mixed genetic background. Here we have investigated genetic suppression of FX-mediated lethality. Reduced recovery of heterozygous mice was observed while backcrossing the null FX allele to C57BL/6J (B6), but was less dramatic in an outcross to CASA/Rk and absent in an outcross to 129S1/SvImJ, indicating that genetic background modifies survival of FX+/- progeny. Substantial strain-specific differences in pre- and postnatal survival of FX-/- progeny were also detected in heterozygous crosses of C57BL/6J congenic, 129S1B6F1, and B6CASAF1 mice. Specifically, intrauterine survival of FX-/- mice was greatly increased during a heterozygous intercross on a uniform C57BL/6J genetic background compared with survival on a hybrid genetic background consisting of a mixture of C57BL/6J and 129S2/SvPas. In addition, statistically significant clustering of FX-/- progeny into litters and specific breeding cages was noted during a B6CASAF1 FX+/- intercross, suggesting a rare mechanism for modifier gene action in which parentally expressed genes define the phenotype, in this case the survival potential, of mutant offspring. Our results disclose that lethality in FX mutant mice is determined by one or more strain-specific modifier loci.

Souris homozygotes pour la mutation du g&egrave;ne FX cibl&eacute;e manifestent une foule d&#39;anomalies pl&eacute;iotropes dont un ph&eacute;notype l&eacute;tal qui est presque totalement p&eacute;n&eacute;trant dans les croisements h&eacute;t&eacute;rozygotes sur un fond g&eacute;n&eacute;tique mixte. Nous avons examin&eacute; suppression g&eacute;n&eacute;tique de l&eacute;talit&eacute; induite par FX. R&eacute;cup&eacute;ration r&eacute;duite chez les souris h&eacute;t&eacute;rozygotes a &eacute;t&eacute; observ&eacute;e alors que l&#39;all&egrave;le nul de FX de C57BL/6J (B6) des r&eacute;trocroisements, mais a &eacute;t&eacute; moins spectaculaire dans une retrempe &agrave; CASA/Rk et absents dans une retrempe &agrave; 129S1/SvImJ, ce qui indique que le bagage g&eacute;n&eacute;tique modifie survie FX +/-prog&eacute;niture. Diff&eacute;rences sp&eacute;cifiques &agrave; la souche pr&eacute;- et postnatale de la survie des FX/de la prog&eacute;niture ont aussi &eacute;t&eacute;s dans les croisements h&eacute;t&eacute;rozygotes de cong&eacute;niques C57BL/6J, 129S1B6F1 et souris de b6casaf1. Plus pr&eacute;cis&eacute;ment, survie intra-ut&eacute;rine de souris/FX augmente fortement pendant une intercross h&eacute;t&eacute;rozygotes sur un fond g&eacute;n&eacute;tique de C57BL/6J uniform par rapport &agrave; la survie sur un fond g&eacute;n&eacute;tique hybride compos&eacute;e d&#39;un m&eacute;lange de C57BL/6J et 129S2/SvPas. En outre, clustering statistiquement significatif de FX/de la prog&eacute;niture dans les liti&egrave;res et les cages d&#39;&eacute;levage sp&eacute;cifiques a &eacute;t&eacute; not&eacute; durant une FX B6CASAF1 +/-intercroisements, sugg&eacute;rant un m&eacute;canisme rare d&#39;action g&egrave;ne modificateur dans quels g&egrave;nes parental d&eacute;finir le ph&eacute;notype, dans ce cas la survie potentielle, de descendance mutante. Nos r&eacute;sultats divulguer que la l&eacute;talit&eacute; chez les souris mutantes FX est d&eacute;termin&eacute;e par un ou plusieurs des locus modificateur sp&eacute;cifique &agrave; la souche.

Il locus FX codifica per un enzima essenziale nella via de novo della biosintesi del PIL-fucosio. Topi omozigoti per una mutazione del gene FX mirata si manifestano una serie di anomalie pleiotropica compreso un fenotipo letale che &egrave; quasi completamente penetrante in eterozigoti intercrosses su un background genetico misto. Qui abbiamo studiato genetica soppressione della letalit&agrave; FX-mediata. Recupero ridotto di topi eterozigoti &egrave; stata osservata mentre retroincroci l&#39;allele null FX di C57BL/6J (B6), ma &egrave; stato meno drammatico in un outcross a CASA/Rk e assente in un outcross a 129S1/SvImJ, che indica che il background genetico modifica sopravvivenza di FX + progenie. Differenze sostanziali del ceppo-specifici nella sopravvivenza pre- e postnatale di FX/progenie sono state rilevate anche in eterozigoti croci di B6CASAF1 topi C57BL/6J congenici e 129S1B6F1. In particolare, sopravvivenza intrauterina di FX/topi &egrave; stato notevolmente aumentato durante un re eterozigoti su uno sfondo uniforme di genetico C57BL/6J confrontato con la sopravvivenza su un background genetico ibrido costituito da una miscela di C57BL/6J e 129S2/SvPas. Inoltre, cluster statisticamente significativo della FX/progenie in cucciolate e gabbie di allevamento specifiche fu notato durante una B6CASAF1 FX + re, suggerendo un meccanismo raro per l&#39;azione del gene modificatore nel quale geni espressi corre definire il fenotipo, in questo caso il potenziale, di sopravvivenza della prole mutante. I nostri risultati rivelare che letalit&agrave; nei topi mutanti FX &egrave; determinata da uno o pi&ugrave; loci ceppo specifico modificatore.

El lugar geom&eacute;trico de FX codifica una enzima esencial en la v&iacute;a de novo de la bios&iacute;ntesis de PIB-fucosa. Ratones homocig&oacute;ticos para una mutaci&oacute;n espec&iacute;fica del gen FX manifiestan una serie de anormalidades pleiotr&oacute;picos incluyendo un fenotipo letal que es casi totalmente penetrante en heterozig&oacute;ticos intercrosses en un fondo gen&eacute;tico mixto. Aqu&iacute; hemos investigado supresi&oacute;n gen&eacute;tica de letalidad FX-mediada. Recuperaci&oacute;n reducidos de ratones heterocigotos observ&oacute; mientras retrocruzamiento el alelo nulo del FX a C57BL/6J (B6), pero fue menos dram&aacute;tica en un outcross a CASA/Rk y ausente en un outcross a 129S1/SvImJ, indicando que la gen&eacute;tica modifica supervivencia de FX + /-progenie. Tambi&eacute;n se detectaron diferencias substanciales de la cepa espec&iacute;fica en la supervivencia de pre- y postnatal de FX-/-progenie en cruces heterozig&oacute;ticos Cong&eacute;nicos C57BL/6J, 129S1B6F1 y ratones de b6casaf1. Espec&iacute;ficamente, supervivencia intrauterina de FX-/-ratones aument&oacute; considerablemente durante un heterozig&oacute;tico cruzan en un fondo gen&eacute;tico de C57BL/6J uniforme en comparaci&oacute;n con la supervivencia en un fondo gen&eacute;tico h&iacute;brido compuesto por una mezcla de C57BL/6J y 129S2/SvPas. Adem&aacute;s, estad&iacute;sticamente significativa agrupamiento de FX-/-progenie en camadas y jaulas de cr&iacute;a espec&iacute;fica fue observado durante un FX B6CASAF1 +/-cruzan, sugiriendo un raro mecanismo de acci&oacute;n g&eacute;nica de modificador en que genes expresados parental definir el fenotipo, en este caso la supervivencia potencial, de descendencia mutante. Nuestros resultados revelan que mortalidad en ratones mutantes FX est&aacute; determinada por uno o m&aacute;s loci de modificador de la cepa espec&iacute;fica.

High-grade gliomas (World Health Organization grade III anaplastic astrocytoma and grade IV glioblastoma multiforme), the most prevalent primary malignant brain tumors, display a cellular hierarchy with self-renewing, tumorigenic cancer stem cells (CSCs) at the apex. While the CSC hypothesis has been an attractive model to describe many aspects of tumor behavior, it remains controversial due to unresolved issues including the use of ex vivo analyses with differential growth conditions. A CSC population has been confirmed in malignant gliomas by preferential tumor formation from cells directly isolated from patient biopsy specimens. However, direct comparison of multiple tumor cell populations with analysis of the resulting phenotypes of each population within a representative tumor environment has not been clearly described. To directly test the relative tumorigenic potential of CSCs and non-stem tumor cells in the same microenvironment, we interrogated matched tumor populations purified from a primary human tumor transplanted into a xenograft mouse model and monitored competitive in vivo tumor growth studies using serial in vivo intravital microscopy. While CSCs were a small minority of the initial transplanted cancer cell population, the CSCs, not the non-stem tumor cells, drove tumor formation and yielded tumors displaying a cellular hierarchy. In the resulting tumors, a fraction of the initial transplanted CSCs maintained expression of stem cell and proliferation markers, which were significantly higher compared to the non-stem tumor cell population and demonstrated that CSCs generated cellular heterogeneity within the tumor. These head-to-head comparisons between matched CSCs and non-stem tumor cells provide the first functional evidence using live imaging that in the same microenvironment, CSCs more than non-stem tumor cells are responsible for tumor propagation, confirming the functional definition of a CSC.

Direct in vivo evidence for tumor propagation by glioblastoma cancer stem cells.

Gliomas de alto grado (Organizaci&#xF3;n Mundial de la Salud de grado III astrocitoma anapl&#xE1;sico y glioblastoma multiforme grado IV), los m&#xE1;s frecuentes los tumores cerebrales primarios malignos, mostrar una jerarqu&#xED;a de celular con auto-renovaci&#xF3;n, tumorig&#xE9;nicos c&#xE9;lulas madre del c&#xE1;ncer (CCC) en el &#xE1;pice. Si bien la hip&#xF3;tesis de CSC ha sido un modelo atractivo para describir muchos aspectos del comportamiento del tumor, que sigue siendo pol&#xE9;mica debido a problemas no resueltos, incluyendo el uso de an&#xE1;lisis ex vivo con las condiciones de crecimiento diferenciales. Una poblaci&#xF3;n CSC ha sido confirmada en los gliomas malignos por la formaci&#xF3;n preferencial del tumor a partir de c&#xE9;lulas aisladas directamente de muestras de biopsias de pacientes. The fundamental mechanism of photodynamic therapy (PDT)-induced cell death has been characterized, but early critical PDT events in vivo remain incompletely defined. With the recent development in advanced fluorescence imaging modalities, such as intravital 2-photon laser scanning microscopy (2P-LSM), researchers are now able to investigate and visualize biological processes with high resolution in real time. This powerful imaging technology allows deep tissue visualization with single-cell resolution, thus providing dynamic information on the 3-dimensional architectural makeup of the tissue. The main goal of this study was to determine the cutaneous penetration of a topically applied photosensitizer, the silicon phthalocyanine Pc 4, into the skin of live animals and to assess the effective absorption of Pc 4 through the skin barrier. Our 2P-LSM images indicate that Pc 4 penetrates to the epidermal/dermal junction of mouse skin. The data also indicate that the degree of Pc 4 absorption is dose dependent. These findings represent initial steps that may help in improving the clinical utilization of topical Pc 4-PDT.

Viewing transplantation immunology through today's lens: new models, new imaging, and new insights.

Human mesenchymal stem cells (hMSCs) have gained intense research interest due to their immune-modulatory, tissue differentiating, and homing properties to sites of inflammation. Despite evidence demonstrating the biodistribution of infused hMSCs in target organs using static fluorescence imaging or whole-body imaging techniques, surprisingly little is known about how hMSCs behave dynamically within host tissues on a single-cell level in vivo. Here, we infused fluorescently labeled clinical-grade hMSCs into immune-competent mice in which neutrophils and monocytes express a second fluorescent marker under the lysozyme M (LysM) promoter. Using intravital two-photon microscopy (TPM), we were able for the first time to capture dynamic interactions between hMSCs and LysM(+) granulocytes in the calvarium bone marrow of recipient mice during systemic LPS challenge in real time. Interestingly, many of the infused hMSCs remained intact despite repeated cellular contacts with host neutrophils. However, we were able to observe the destruction and subsequent phagocytosis of some hMSCs by surrounding granulocytes. Thus, our imaging platform provides opportunities to gain insight into the biology and therapeutic mechanisms of hMSCs in vivo at a single-cell level within live hosts.

Dynamic Imaging of Marrow-Resident Granulocytes Interacting with Human Mesenchymal Stem Cells upon Systemic Lipopolysaccharide Challenge.

After traumatic spinal cord injury, functional deficits increase as axons die back from the center of the lesion and the glial scar forms. Axonal dieback occurs in two phases: an initial axon intrinsic stage that occurs over the first several hours and a secondary phase which takes place over the first few weeks after injury. Here, we examine the secondary phase, which is marked by infiltration of macrophages. Using powerful time-lapse multi-photon imaging, we captured images of interactions between Cx3cr1(+/GFP) macrophages and microglia and Thy-1(YFP) axons in a mouse dorsal column crush spinal cord injury model. Over the first few weeks after injury, axonal retraction bulbs within the lesion are static except when axonal fragments are lost by a blebbing mechanism in response to physical contact followed by phagocytosis by mobile Cx3Cr1(+/GFP) cells. Utilizing a radiation chimera model to distinguish marrow-derived cells from radio-resistant CNS-resident microglia, we determined that the vast majority of accumulated cells in the lesion are derived from the blood and only these are associated with axonal damage. Interestingly, CNS-resident Cx3Cr1(+/GFP) microglia did not increasingly accumulate nor participate in neuronal destruction in the lesion during this time period. Additionally, we found that the blood-derived cells consisted mainly of singly labeled Ccr2(+/RFP) macrophages, singly labeled Cx3Cr1(+/GFP) macrophages and a small population of double-labeled cells. Since all axon destructive events were seen in contact with a Cx3Cr1(+/GFP) cell, we infer that the CCR2 single positive subset is likely not robustly involved in axonal dieback. Finally, in our model, deletion of CCR2, a chemokine receptor, did not alter the position of axons after dieback. Understanding the in vivo cellular interactions involved in secondary axonal injury may lead to clinical treatment candidates involving modulation of destructive infiltrating blood monocytes.

High-resolution intravital imaging reveals that blood-derived macrophages but not resident microglia facilitate secondary axonal dieback in traumatic spinal cord injury.

The clinical application of Mesenchymal Stem Cells (MSCs) for the treatment of a variety of diseases is the focus of intense research. Despite large research efforts many questions regarding MSC biology in vivo remain unanswered. For instance, we do not know for certain whether MSCs exert their therapeutic effects directly within the target tissue or indirectly by influencing the polarization of other cell types, such as macrophages, which can then home to the target tissue microenvironment. To help address this issue, the application of intravital multiphoton microscopy allows for the determination of the dynamic action of intact MSCs versus endogenous host cells at the target tissue site in real time.
