A long-standing problem in development is understanding how progenitor cells transiently expressing genes contribute to complex anatomical and functional structures. In the developing nervous system an additional level of complexity arises when considering how cells of distinct lineages relate to newly established neural circuits. To address these problems, we used both cumulative marking with Cre/loxP and Genetic Inducible Fate Mapping (GIFM), which permanently and heritably marks small populations of progenitors and their descendants with fine temporal control using CreER/loxP. A key component used in both approaches is a conditional phenotyping allele that has the potential to be expressed in all cell types, but is quiescent because of a loxP flanked Stop sequence, which precedes a reporter allele. Upon recombination, the resulting phenotyping allele is 'turned on' and then constitutively expressed. Thus, the reporter functions as a high fidelity genetic lineage tracer in vivo. Currently there is an array of reporter alleles that can be used in marking strategies, but their recombination efficiency and applicability to a wide array of tissues has not been thoroughly described. To assess the recombination/marking potential of the reporters, we utilized CreER(T) under the control of a Wnt1 transgene (Wnt1-CreER(T)) as well as a cumulative, non-inducible En1(Cre) knock-in line in combination with three different reporters: R26R (LacZ reporter), Z/EG (EGFP reporter), and Tau-Lox-STOP-Lox-mGFP-IRES-NLS-LacZ (membrane-targeted GFP/nuclear LacZ reporter). We marked the Wnt1 lineage using each of the three reporters at embryonic day (E) 8.5 followed by analysis at E10.0, E12.5, and in the adult. We also compared cumulative marking of cells with a history of En1 expression at the same stages. We evaluated the reporters by whole-mount and section analysis and ascertained the strengths and weaknesses of each of the reporters. Comparative analysis with the reporters elucidated complexities of how the Wnt1 and En1 lineages contribute to developing embryos and to axonal projection patterns of neurons derived from these lineages.

Un probl&#xE8;me de longue date dans le d&#xE9;veloppement est de comprendre comment les cellules prog&#xE9;nitrices de mani&#xE8;re transitoire des g&#xE8;nes exprimant contribuer &#xE0; des structures anatomiques complexes et fonctionnel. Dans le syst&#xE8;me nerveux en d&#xE9;veloppement un niveau suppl&#xE9;mentaire de complexit&#xE9; se pose lorsque l&#x26;#39;on consid&#xE8;re comment les cellules de lign&#xE9;es distinctes se rapportent &#xE0; nouvellement cr&#xE9;&#xE9;es circuits neuronaux. Pour r&#xE9;soudre ces probl&#xE8;mes, nous avons utilis&#xE9; &#xE0; la fois cumulatif marquage avec Cre / loxP et cartographie g&#xE9;n&#xE9;tique destin inductible (GIFM), qui marque de fa&#xE7;on permanente et heritably de petites populations de prog&#xE9;niteurs et de leurs descendants avec un contr&#xF4;le temporel pr&#xE9;cis avec CREER / loxP. Un &#xE9;l&#xE9;ment cl&#xE9; utilis&#xE9; dans les deux approches est un all&#xE8;le ph&#xE9;notypage conditionnelle qui a le potentiel pour &#xEA;tre exprim&#xE9; dans tous les types de cellules, mais est au repos en raison d&#x26;#39;une s&#xE9;quence loxP flanqu&#xE9;e d&#x26;#39;arr&#xEA;t, qui pr&#xE9;c&#xE8;de un all&#xE8;le journaliste. Nous avons marqu&#xE9; la lign&#xE9;e Wnt1 en utilisant chacun des trois journalistes &#xE0; jour embryonnaire (E) 8,5 suivie d&#x26;#39;une analyse &#xE0; E10.0, E12.5, et chez l&#x26;#39;adulte. Analyse comparative des all&#xE8;les Reporter conditionnelles dans le d&#xE9;veloppement de l&#x26;#39;embryon et embryonnaire du syst&#xE8;me nerveux

Ein anhaltendes Problem in der Entwicklung ist verstehen, wie Vorl&auml;uferzellen, die vor&uuml;bergehend Ausdr&uuml;cken Gene zu komplexen anatomischen und funktionalen Strukturen beitragen. In das sich entwickelnde Nervensystem entsteht ein zus&auml;tzliches Ma&szlig; an Komplexit&auml;t, wenn unter Ber&uuml;cksichtigung der Zellen unterschiedliche Linien neu Beziehung zu neuronale Schaltkreise gegr&uuml;ndet. Um diese Probleme zu beheben, haben wir beide kumulative Kennzeichnung mit Cre/LoxP und genetische Inducible Schicksal Mapping (GIFM), die dauerhaft und heritably kleine Populationen der Vorfahren und ihre Nachkommen mit feinen zeitliche Steuerung mit CreER/LoxP Marken verwendet. Eine Schl&uuml;sselkomponente in beide Ans&auml;tze verwendet ist eine bedingte pathologies-Allel, die das Potenzial hat, alle Zelltypen ausgedr&uuml;ckt werden, aber ruhige aufgrund einer LoxP flankiert Stop-Sequenz, die ein Reporter-Allel vorausgeht. Bei der Rekombination ist die resultierende pathologies Allel &#39;eingeschaltet&#39; und dann konstitutiv ausgedr&uuml;ckt. So funktioniert die Reporter als ein High-Fidelity-genetische Abstammung-Tracer, der in-vivo. Derzeit gibt es eine Reihe von Reporter-Allele, die in der Kennzeichnung, Strategien, sondern deren Rekombination Effizienz verwendet werden kann und Anwendbarkeit auf einer Vielzahl von Geweben nicht ausf&uuml;hrlich beschrieben. Um das Rekombination/Kennzeichnung der Reporter bewerten, haben wir CreER(T) unter der Kontrolle einer Wnt1 Transgene genutzt (Wnt1-CreER(T)) sowie eine kumulative-inducible En1(Cre) Knock-in Linie in Kombination mit drei verschiedenen Reportern: R26R (LacZ Reporter), Z/EG (EGFP Reporter) und Tau-Lox-STOP-Lox-mGFP-IRES-NLS-LacZ (Membran-bezogene GFP/nuklearen LacZ Reporter). Hier markieren wir die Wnt1-Linie, die mit den einzelnen der drei Reporter bei embryonalen Tag (E) 8.5 mit anschlie&szlig;ender Analyse auf E10.0, E12.5, und in den Erwachsenen. Wir verglichen auch kumulative Markierung von Zellen mit einer Geschichte von En1 Ausdruck in den gleichen Phasen. Wir die Reporter von ganz-Mount und Abschnitt Analyse ausgewertet und festgestellt, die St&auml;rken und Schw&auml;chen der einzelnen Reportern. Vergleichende Analyse mit den Reportern verdeutlicht die Komplexit&auml;t wie die Wnt1 und En1 &Uuml;bertragungslinien Embryonen und dem axonale Projektion Muster von Neuronen abgeleitet von diesen Linien beitragen.

Un annoso problema in via di sviluppo &egrave; capire come le cellule progenitrici transitoriamente espressione dei geni contribuiscono a complesse strutture anatomiche e funzionali. Nel sistema nervoso in via di sviluppo un ulteriore livello di complessit&agrave; si pone quando considerando come cellule di distinti lignaggi riguardano appena stabilito circuiti neurali. Per risolvere questi problemi, abbiamo usato sia cumulativo marcatura con Cre/loxP e genetica inducibile destino Mapping (GIFM), che contrassegna permanentemente e heritably piccole popolazioni di progenitori e dei loro discendenti con controllo temporale fine utilizzando CreER/loxP. Un componente chiave utilizzati in entrambi gli approcci &egrave; un allele fenotipi condizionale che ha il potenziale per essere espresso in tutti i tipi di cellule, ma &egrave; tranquillo a causa di una sequenza di Stop loxP affiancato, che precede un allele reporter. Abbiamo segnato il lignaggio di Wnt1 con ciascuno dei tre giornalisti a giorno embrionale (E) 8.5 seguita da analisi a E10.0, E12.5 e nell&#39;adulto. Abbiamo confrontato anche cumulativo marcatura delle cellule con una storia di En1 espressione presso le stesse fasi. Abbiamo valutato i reporter dall&#39;intero-monta e sezione analisi e accertata la forza e debolezza di ciascuno dei giornalisti. Analisi comparativa con la complessit&agrave; chiarito ai giornalisti di come i lignaggi Wnt1 ed En1 contribuiscono allo sviluppo di embrioni e di modelli di proiezione assonali dei neuroni derivati da questi lignaggi.

Para evaluar la recombinaci&#xF3;n / marcado potencial de los reporteros, hemos utilizado CREER (T) bajo el control de un transg&#xE9;n Wnt1 (Wnt1-CREER (T)), as&#xED; como un acumulativo, no inducible En1 (Cre) en cadena en l&#xED;nea en combinaci&#xF3;n con tres reporteros diferentes: R26R (lacZ reportero), Z / EG (EGFP reportero), y Tau-Lox-STOP-Lox-mGFP-IRES-NLS-LacZ (membrana orientada GFP / nuclear reportero LacZ). Se marc&#xF3; el linaje Wnt1 utilizando cada uno de los tres reporteros en d&#xED;a embrionario (E) 8,5 seguido por an&#xE1;lisis en E10.0, E12.5, y en el adulto. Asimismo, en comparaci&#xF3;n acumulada marcado de las c&#xE9;lulas con una historia de En1 expresi&#xF3;n en las mismas etapas. Se evalu&#xF3; a los periodistas por todo el montaje y el an&#xE1;lisis de la secci&#xF3;n y comprobar las fortalezas y debilidades de cada uno de los periodistas. El an&#xE1;lisis comparativo con los periodistas dilucidado complejidades de c&#xF3;mo los Wnt1 y en1 linajes contribuir a los embriones en desarrollo y los patrones de proyecci&#xF3;n axonal de las neuronas derivadas de estos linajes.

Neurons are born throughout adulthood in the hippocampus and show enhanced plasticity compared with mature neurons. However, there are conflicting reports on whether or not young neurons contribute to performance in behavioral tasks, and there is no clear relationship between the timing of maturation of young neurons and the duration of neurogenesis reduction in studies showing behavioral deficits. We asked whether these discrepancies could reflect differences in the properties of young neurons in mice and rats. We report that young neurons in adult rats show a mature neuronal marker profile and activity-induced immediate early gene expression 1-2 weeks earlier than those in mice. They are also twice as likely to escape cell death, and are 10 times more likely to be recruited into learning circuits. This comparison holds true in two different strains of mice, both of which show high rates of neurogenesis relative to other background strains. Differences in adult neurogenesis are not limited to the hippocampus, as the density of new neocortical neurons was 5 times greater in rats than in mice. Finally, in a test of function, we find that the contribution of young neurons to fear memory is much greater in rats than in mice. These results reveal substantial differences in new neuron plasticity and function between these two commonly studied rodent species.

Midbrain dopamine (MbDA) neurons are functionally heterogeneous and modulate complex functions through precisely organized anatomical groups. MbDA neurons are generated from Wnt1-expressing progenitors located in the ventral mesencephalon (vMes) during embryogenesis. However, it is unclear whether the progenitor pool is partitioned into distinct cohorts based on molecular identity and whether the timing of gene expression uniquely identifies subtypes of MbDA neurons. In this study we show that Wnt1-expressing MbDA progenitors from embryonic day (E)8.5-12.5 have dynamic molecular identities that correlate with specific spatial locations in the vMes. We also tested the hypothesis that the timing of Wnt1 expression in progenitors is related to the distribution of anatomically distinct cohorts of adult MbDA neurons using genetic inducible fate mapping (GIFM). We demonstrate that the Wnt1 lineage contributes to specific cohorts of MbDA neurons during a 7-day epoch and that the contribution to MbDA neurons predominates over other ventral Mb domains. In addition, we show that calbindin-, GIRK2-, and calretinin-expressing MbDA neuron subtypes are derived from Wnt1-expressing progenitors marked over a broad temporal window. Through GIFM and quantitative analysis we demonstrate that the Wnt1 lineage does not undergo progressive lineage restriction, which eliminates a restricted competence model of generating MbDA diversity. Interestingly, we uncover that two significant peaks of Wnt1 lineage contribution to MbDA neurons occur at E9.5 and E11.5. Collectively, our findings delineate the temporal window of MbDA neuron generation and show that lineage and timing predicts the terminal distribution pattern of MbDA neurons.

Molecular organization and timing of Wnt1 expression define cohorts of midbrain dopamine neuron progenitors in vivo.

MBDA neurones sont g&#xE9;n&#xE9;r&#xE9;s &#xE0; partir Wnt1 exprimant prog&#xE9;niteurs situ&#xE9;s dans le m&#xE9;senc&#xE9;phale ventral (VME) au cours de l&#x26;#39;embryogen&#xE8;se. Cependant, il est difficile de savoir si la piscine g&#xE9;niteur est divis&#xE9;e en cohortes distinctes bas&#xE9;es sur l&#x26;#39;identit&#xE9; mol&#xE9;culaire et si le moment de l&#x26;#39;expression g&#xE9;nique identifie de mani&#xE8;re unique sous-types de neurones MBDA. Tuttavia, non &egrave; chiaro se il pool di progenitrici &egrave; partizionato in coorti distinte basate sull&#39;identit&agrave; molecolare e se la tempistica dell&#39;espressione genica identifica in modo univoco i sottotipi di MbDA neuroni. In questo studio ci mostrano che Wnt1-esprimendo MbDA progenitori da giorno embrionale (E) 8,5-12,5 hanno dinamica molecolare identit&agrave; che correlano con le specifiche posizioni spaziali il vMes. Abbiamo testato anche l&#39;ipotesi che i tempi di Wnt1 espressione in progenitori sono correlato alla distribuzione delle coorti anatomicamente distinte di adulti MbDA neuroni mediante mappatura genetica destino inducibile (GIFM). Dimostriamo che il lignaggio Wnt1 contribuisce a specifiche coorti di MbDA neuroni durante un&#39;epoca di 7 giorni e che il contributo di MbDA neuroni predomina negli altri domini Mb ventrale. Inoltre, mostriamo che calbindin-, GIRK2-e che esprimono calretinina MbDA neurone sottotipi derivano da progenitori che esprimono Wnt1 segnati sopra una larga finestra temporale. Attraverso GIFM e analisi quantitativa ci dimostrano che il lignaggio Wnt1 non subire restrizione progressiva lignaggio, che elimina un modello di competenza limitata di generare diversit&agrave; MbDA. &Egrave; interessante notare, noi scoprire che due picchi significativi del contributo lignaggio Wnt1 a MbDA neuroni si verificano presso E9.5 ed E11.5. Collettivamente, i nostri risultati delineano la finestra temporale di generazione neurone MbDA e mostrano quel lignaggio e temporizzazione predice il pattern di distribuzione terminale dei neuroni MbDA.

Mesenc&#xE9;falo dopamina (MBDA), las neuronas son funcionalmente heterog&#xE9;neas y modular las funciones complejas a trav&#xE9;s de grupos organizados de precisi&#xF3;n anat&#xF3;mica. MBDA las neuronas se generan a partir Wnt1-expresando progenitores ubicados en el mesenc&#xE9;falo ventral (EMV) durante la embriog&#xE9;nesis. Sin embargo, no est&#xE1; claro si el reservorio progenitor se divide en distintas cohortes basadas en la identidad molecular y si el momento de la expresi&#xF3;n g&#xE9;nica identifica de forma &#xFA;nica los subtipos de neuronas MBDA. En este estudio mostramos que Wnt1-expresando progenitores MBDA desde el primer d&#xED;a embrionario (E) 08.05 a 12.05 tienen identidades din&#xE1;micas moleculares que se correlacionan con determinadas localizaciones espaciales en los ecosistemas marinos vulnerables. Tambi&#xE9;n probamos la hip&#xF3;tesis de que el momento de Wnt1 expresi&#xF3;n en los progenitores se relaciona con la distribuci&#xF3;n de las cohortes de adultos anat&#xF3;micamente distintos MBDA neuronas que utilizan la cartograf&#xED;a gen&#xE9;tica destino inducible (GIFM). Se demuestra que el linaje Wnt1 contribuye a cohortes espec&#xED;ficos de neuronas MBDA durante una &#xE9;poca de 7 d&#xED;as y que la contribuci&#xF3;n a MBDA neuronas predomina sobre otros dominios Mb ventrales. Adem&#xE1;s, nos muestran que calbindina, GIRK2-, y que expresan calretinina MBDA subtipos neuronales se derivan de Wnt1-que expresan las c&#xE9;lulas progenitoras marcadas por una ventana temporal m&#xE1;s amplio. A trav&#xE9;s del an&#xE1;lisis GIFM y cuantitativos que demuestran que el linaje Wnt1 no pasan por una restricci&#xF3;n progresiva de linaje, lo que elimina un modelo de competencia restringida de la generaci&#xF3;n de diversidad de MBDA. Curiosamente, hemos descubierto que hay dos picos importantes de la contribuci&#xF3;n de linaje Wnt1 a las neuronas MBDA se producen en E9.5 y E11.5.
