Leukocytes and leukocyte-derived microparticles contain low levels of tissue factor (TF) and incorporate into forming thrombi. Although this circulating pool of TF has been proposed to play a key role in thrombosis, its functional significance relative to that of vascular wall TF is poorly defined. We tested the hypothesis that leukocyte-derived TF contributes to thrombus formation in vivo. Compared to wild-type mice, mice with severe TF deficiency (ie, TF(-/-), hTF-Tg+, or "low-TF") demonstrated markedly impaired thrombus formation after carotid artery injury or inferior vena cava ligation. A bone marrow transplantation strategy was used to modulate levels of leukocyte-derived TF. Transplantation of low-TF marrow into wild-type mice did not suppress arterial or venous thrombus formation. Similarly, transplantation of wild-type marrow into low-TF mice did not accelerate thrombosis. In vitro analyses revealed that TF activity in the blood was very low and was markedly exceeded by that present in the vessel wall. Therefore, our results suggest that thrombus formation in the arterial and venous macrovasculature is driven primarily by TF derived from the blood vessel wall as opposed to leukocytes.

P-selectin inhibition has been shown to decrease thrombogenesis in multiple animal species. In this study, we show that a novel oral small-molecule inhibitor of P-selectin, PSI-697, promotes thrombus resolution and decreases inflammation in a baboon model of venous thrombosis. Experimental groups consisted of the following: 1) primates receiving a single oral dose of PSI-697 (30 mg/kg) daily starting three days pre-iliac vein balloon occlusion, and continued for six days; 2) primates receiving a single treatment dose of a low-molecular-weight-heparin (LMWH) (1.5 mg/kg) daily starting one day pre-iliac balloon occlusion, and continued for six days; and 3) primates receiving a single oral dose of a vehicle control daily starting three days pre-iliac vein balloon occlusion, and continued for six days. Animals receiving PSI-697, although thrombosed after balloon deflation, demonstrated greater than 80% vein lumen opening over time, with no opening (0%) for vehicle control (p < 0.01). LMWH opening evident after balloon deflation slightly deteriorated over time compared to PSI-697. PSI-697 therapy also significantly decreased vein wall inflammation determined by magnetic resonance venography (MRV). Importantly, this beneficial opening occurred without measured anticoagulation. Animals receiving PSI-697 demonstrated significantly increased plasma D-dimer levels versus LMWH and control animals six hours post thrombus induction (p < 0.01). This study is the first to demonstrate the effectiveness of oral P-selectin inhibition to modify venous thrombogenesis, increase vein lumen opening, and decrease inflammation in a large animal model.

Resolution of venous thrombosis using a novel oral small-molecule inhibitor of P-selectin (PSI-697) without anticoagulation.

Los animales que recibieron PSI-697, a pesar de trombosis despu&#xE9;s de desinflar el bal&#xF3;n, demostr&#xF3; que la luz de la vena mayor del 80% de abrir el paso del tiempo, sin apertura (0%) para el control del veh&#xED;culo (p &#x26;lt;0,01). Microparticles (MP) are lipid vesicles from platelets, leukocytes and endothelial cells that are involved in early thrombogenesis. We evaluated a detailed time-course analysis of MPs on thrombogenesis and the associated tissue factor (TF) activity in wild-type, in gene-deleted for E- and P-selectins and with high levels of P-selectin expression after the initiation of venous thrombosis in mice. Inferior vena cava (IVC) ligation was performed on C57BL/6 mice (n = 191, 59 = wild-type [WT], 55 = gene-deleted for E- and P - selectins [knock-outs, EPKO] and 77 = elevated levels of soluble P-selectin, named Delta Cytoplasmic Tail (DeltaCT). Animals were euthanised at various time points to assess MP production, origin and thrombus weight. MPs were re-injected into separate mice at concentrations of 80,000 and 160,000 units, as well as from different ages. In addition, MPs from thrombosed animals were pooled and TF activity quantitated using a chromogenic assay. Thrombus weight correlated negatively with MPs derived from leukocytes, and positively with MPs derived from platelets for WT animals (p < 0.05), while MPs from platelets presented a positive correlation to thrombus weight in the WT and EPKO groups (p < 0.01). Total MPs correlated negatively with thrombus weight in the DeltaCT group (p < 0.05). MP re-injections led to greater thrombus weight, while older MP reinjections tended to form larger thrombus than younger. Finally, TF bearing MPs showed a significant correlation to MP concentrations (R = 0.99). In conclusion, MPs appear to be an important element in venous thrombogenesis.

Vena Cava inferior (IVC) Verbindung wurde am C57BL/6 M&auml;usen durchgef&uuml;hrt (n = 191, 59 = Wildtyp [WT], 55 f&uuml;r E- und P - Selectins [Knock-Outs, EPKO] = gen gel&ouml;scht und 77 = erh&ouml;hte Niveaus des l&ouml;slichen P-selectin, Delta zellplasmatisches Endst&uuml;ck (DeltaCT) benannt. Tiere waren Beseitigungsanstalt zu verschiedenen Zeitpunkten MP Produktion, Herkunft und Thrombus Gewicht beurteilen. Gesamt MPs korreliert negativ mit dem Gewicht der Thrombus in der DeltaCT-Gruppe (p &lt; 0,05). MP Re-injections f&uuml;hrte zu gr&ouml;&szlig;eren Thrombus Gewicht, w&auml;hrend &auml;ltere MP Reinjections tendenziell gr&ouml;&szlig;ere Thrombus als j&uuml;ngere bilden. Schlie&szlig;lich TF mit MPs ergab eine signifikante Korrelation zu MP-Konzentrationen (R = 0.99). Zusammenfassend erscheinen MPs ein wichtiges Element im ven&ouml;sen Thrombogenese.

BACKGROUND: Hyperlipidemia increases the level of blood plasminogen activator inhibitor-1 (PAI-1) that is responsible for regulating fibrinolysis by inhibiting both urokinase-type plasminogen activator (u-PA) and tissue-type plasminogen activator (t-PA). While this fibrinolytic pathway is well known, the role of PAI-1 in venous thrombosis (VT) under hyperlipidemic conditions has not been fully established. We sought to determine the effects of PAI-1 in an in vivo hyperlipidemic model of VT. METHODS: C57BL/6 wild-type (WT) mice, apolipoprotein E gene-deleted mice (ApoE-/-) having hyperlipidemia, and PAI-1 gene-deleted (PAI-1-/-) mice were used in this study. Inferior vena cava (IVC) ligation below the level of the renal veins was performed to create a stasis VT. Endpoints included measuring acute thrombosis (day 2) and chronic thrombosis (days 6 and 14). At euthanasia, blood samples were collected for plasmin and PAI-1 activity. In addition, the IVC and its thrombus were evaluated for thrombus weight (TW), u-PA activity, and differential leukocyte count while the vein wall only was analyzed for monocyte chemoattractant protein-1 (MCP-1), matrix metalloproteinase (MMP) 2, and MMP-9. RESULTS: Compared to WT at day 2, ApoE-/-mice demonstrated a statistically significant 14% increase in TW (P < .05) and a significant 41% increase in circulating PAI-1 activity (P < .05), while showing a trend of decreased plasmin activity. In addition, TW in ApoE-/-mice was 45% higher than PAI-1-/-mice at day 2 (P < .05), 33% at day 6 (P < .01), and 41% at day 14 (P < .01). ApoE-/-mice exhibited undetectable levels of u-PA in both vein wall and thrombus, compared to WT, at all time points. Also, vein wall MMP-2 was significantly decreased by 64% at day 6 (P < .01) and 58% at day 14 (P < .05). MMP-9 was significantly decreased by 71% at day 2 (P < .01) and 48% at day 6 (P < .01), in ApoE-/-mice compared to WT mice. In addition, in ApoE-/-mice, MCP-1 was significantly decreased by 38% at day 2 (P < .01) and 67% at day 6 (P < .01) vs WT mice. As expected in ApoE mice, following a decrease in MCP-1, monocyte recruitment was significantly decreased at days 6 (P < .01) and 14 (P < .05). CONCLUSIONS: A significant increase of circulating PAI-1 levels in hyperlipidemic mice correlated with an early increase in TW due to impaired fibrinolysis. The undetectable levels of u-PA in ApoE-/-mice correlated to a decrease in vein wall MMP-2, MMP-9, MCP-1, and a decrease in monocyte recruitment diminishing thrombus resolution.

Impaired fibrinolytic system in ApoE gene-deleted mice with hyperlipidemia augments deep vein thrombosis.

CONTEXTE: L&#x26;#39;hyperlipid&#xE9;mie augmente le niveau de sang plasminogen activator inhibitor-1 (PAI-1) qui est charg&#xE9; de r&#xE9;glementer la fibrinolyse en inhibant &#xE0; la fois un activateur plasminog&#xE8;ne de type urokinase (u-PA) et de tissus de type activateur de plasminog&#xE8;ne (t-PA). Bien que cette voie fibrinolytique est bien connu, le r&#xF4;le de PAI-1 dans la thrombose veineuse (VT) dans des conditions hyperlipid&#xE9;miques n&#x26;#39;a pas &#xE9;t&#xE9; compl&#xE8;tement &#xE9;tablie. Nous avons cherch&#xE9; &#xE0; d&#xE9;terminer les effets de PAI-1 dans un mod&#xE8;le in vivo de hyperlipid&#xE9;mique VT. L&#x26;#39;affaiblissement du syst&#xE8;me fibrinolytique chez les souris ApoE Gene-supprim&#xE9;s avec hyperlipid&#xE9;mie Augments thrombose veineuse profonde

Hintergrund: Hyperlipid&auml;mie erh&ouml;ht das Blut Plasminogen Aktivator Inhibitor-1 (PAI-1), verantwortlich f&uuml;r die Regulierung der Fibrinolyse durch die Hemmung der Urokinase Plasminogen Aktivator (u-PA) und Gewebe-Plasminogen Aktivator (t-PA) ist. W&auml;hrend dieses Fibrinolytikum Weg bekannt ist, hat die Rolle des PAI-1 bei Venenthrombose (VT) unter hyperlipidemic Bedingungen nicht vollst&auml;ndig eingerichtet wurde. Wollten wir die Auswirkungen der PAI-1 in einem in-vivo-hyperlipidemic-Modell des VT bestimmen Methoden: C57BL/6 Wildtyp (WT) M&auml;use, Apolipoprotein E-gen-gel&ouml;scht-M&auml;use (ApoE-/-), Hyperlipid&auml;mie, und PAI-1-Gen-gel&ouml;scht (PAI - 1-/-) M&auml;use wurden in dieser Studie verwendet. Gest&ouml;rter fibrinolytischen System ApoE-gen-gel&ouml;scht-M&auml;usen mit Hyperlipid&auml;mie erweitert die tiefe Venenthrombose.

Come previsto nei topi ApoE, seguendo una diminuzione di MCP-1, reclutamento di monociti era significativamente diminuito a 6 giorni (P &lt;,01) e 14 (P &lt;,05 dentellatura CONCLUSIONI: Un aumento significativo dei livelli di PAI-1 nei topi iperlipidemici correlati con un aumento precoce della TW a causa della ridotta fibrinolisi circolanti. I livelli rilevabili di u-PA in ApoE-/-mice correlato ad una diminuzione della parete venosa MMP-2, MMP-9, MCP-1 e una diminuzione nel reclutamento di monociti diminuendo risoluzione trombo.

Sistema fibrinolitica alterata in alcuni topi eliminato gene ApoE con iperlipidemia aumenta la trombosi venosa profonda.

&#xC7A5;&#xC560;&#xC778;&#xB41C; &#xB610;&#xD55C; &#xC2DC;&#xC2A4;&#xD15C; ApoE &#xC720;&#xC804;&#xC790; &#xC0AD;&#xC81C; &#xC950; &#xD608;&#xACFC; &#xAE4A;&#xC740; &#xC815; &#xB9E5; &#xD608;&#xC804; &#xC99D;&#xC744; &#xAC15;&#xD654;&#xD569;&#xB2C8;&#xB2E4;.

Antecedentes: La hiperlipidemia aumenta el nivel de la sangre del activador del plasmin&#xF3;geno-1 (PAI-1) que es responsable de regular mediante la inhibici&#xF3;n de la fibrin&#xF3;lisis tanto activador plasmin&#xF3;geno de tipo uroquinasa (u-PA) y de tipo tisular del plasmin&#xF3;geno (t-PA). Si bien esta v&#xED;a fibrinol&#xED;tica es bien sabido, el papel de PAI-1 en la trombosis venosa (VT), bajo condiciones hiperlipid&#xE9;micos no se ha establecido completamente. Hemos tratado de determinar los efectos de PAI-1 en un modelo in vivo de la TV hiperlipidemia. M&#xC9;TODOS: C57BL / 6 de tipo salvaje (WT) los ratones, los ratones apolipoprote&#xED;na E genes eliminados (ApoE-/ -) con hiperlipidemia y PAI-1 genes eliminados (PAI-1-/ -) ratones fueron utilizados en este estudio. Vena cava inferior (VCI) ligadura por debajo del nivel de las venas renales se realiz&#xF3; para crear un VT estasis. Los puntos finales incluyeron la medici&#xF3;n de la trombosis aguda (d&#xED;a 2) y trombosis cr&#xF3;nica (d&#xED;as 6 y 14). En la eutanasia, las muestras de sangre fueron recolectadas por la plasmina y la actividad de PAI-1. Adem&#xE1;s, la VCI y sus trombo se evaluaron para el peso del trombo (TW), la actividad de u-PA, y el recuento de leucocitos diferencial, mientras que la pared de la vena s&#xF3;lo se analiz&#xF3; para determinar matriz MCP-1 (MCP-1), metaloproteinasa (MMP) 2 y MMP-9. RESULTADOS: En comparaci&#xF3;n con WT en el d&#xED;a 2, ApoE-/-mice demostr&#xF3; un aumento estad&#xED;sticamente significativo del 14% en TW (P &#x26;lt;.05) y un aumento significativo del 41% en la circulaci&#xF3;n de actividad PAI-1 (P &#x26;lt;.05), mientras se muestra una tendencia de la actividad de plasmina disminuido. Adem&#xE1;s, TW en ApoE-/-mice fue del 45% m&#xE1;s alto que PAI-1-/-mice en el d&#xED;a 2 (p &#x26;lt;0,05), el 33% en el d&#xED;a 6 (P &#x26;lt;.01), y el 41% a los 14 d&#xED;as (P &#x26;lt;.01). ApoE-/-mice mostraron niveles indetectables de u-PA en la pared de la vena y el trombo tanto, en comparaci&#xF3;n con el WT, en todos los tiempos. Adem&#xE1;s, la pared venosa de MMP-2 se redujo significativamente en un 64% el d&#xED;a 6 (P &#x26;lt;.01) y el 58% a los 14 d&#xED;as (P &#x26;lt;.05). MMP-9 se redujo significativamente en un 71% el d&#xED;a 2 (p &#x26;lt;.01) y 48% el d&#xED;a 6 (P &#x26;lt;.01), en comparaci&#xF3;n con los ratones ApoE-/-mice WT. Adem&#xE1;s, en ApoE-/-mice, MCP-1 se redujo significativamente en un 38% el d&#xED;a 2 (p &#x26;lt;.01) y el 67% el d&#xED;a 6 (p &#x26;lt;0,01) frente a los ratones WT. Como era de esperar en los ratones ApoE, a ra&#xED;z de una disminuci&#xF3;n de la MCP-1, el reclutamiento de monocitos fue significativamente menor en los d&#xED;as 6 (P &#x26;lt;.01) y 14 (P &#x26;lt;.05). CONCLUSIONES: Hubo un aumento significativo de la circulaci&#xF3;n de los niveles de PAI-1 en ratones hiperlipid&#xE9;micos correlacionado con un aumento a principios de TW debido a la alteraci&#xF3;n de la fibrin&#xF3;lisis. Los niveles indetectables de u-PA en ApoE-/-mice correlacionada con una disminuci&#xF3;n en la vena pared MMP-2, MMP-9, MCP-1, y una disminuci&#xF3;n en el reclutamiento de monocitos disminuyendo la resoluci&#xF3;n del trombo.
