The next generation of tissue engineered constructs (TECs) requires the incorporation of a controllable and optimized microstructure if they are to chemically, mechanically, and biologically mimic tissue function. In order to obtain TECs with optimized microstructures, a combination of spatiotemporally regulated mechanical and biochemical stimuli is necessary during the formation of the construct. While numerous efforts have been made to create functional tissue constructs, there are few techniques available to stimulate TECs in a localized manner. We herein describe the design of a microdevice which can stimulate TECs in a localized, inhomogeneous, and predefined anisotropic fashion using ferromagnetically doped polydimethylsiloxane microflaps (MFs). Specifically, a sequential magneto-structural finite element model of the proposed microdevice is constructed and utilized to understand how changes in magnetic and geometrical properties of the device affect MF deflection. Our study indicates that a relatively small density of ferromagnetic material is required to result in adequate force and MF defection (175 microm approximately 7% TEC strain). We also demonstrate that MF to magnet distance is more important than inherent MF magnetic permeability in determining resulting MF deflection. An experimental validation test setup was used to validate the computational solutions. The comparison shows reasonable agreement indicating a 5.9% difference between experimentally measured and computationally predicted MF displacement. Correspondingly, an apparatus with two MFs and two magnets has been made and is currently undergoing construct testing. The current study presents the design of a novel magnetic microactuator for tissue engineering applications. The computational results reported here will form the foundation in the design and optimization of a functional microdevice with multiple MFs and magnets capable of stimulating TECs in nonhomogenous and preferred directions with relevant spatial resolution.

La pr&#xF3;xima generaci&#xF3;n de construcciones de ingenier&#xED;a tisular (TEC) exige la incorporaci&#xF3;n de una microestructura controlable y optimizado para que puedan qu&#xED;mica, mec&#xE1;nica y biol&#xF3;gicamente imitar la funci&#xF3;n del tejido. Con el fin de obtener TEC con microestructuras optimizadas, una combinaci&#xF3;n de espaciotemporalmente regulados est&#xED;mulos mec&#xE1;nicos y bioqu&#xED;micos es necesario durante la formaci&#xF3;n de la construcci&#xF3;n. Mientras que los esfuerzos se han hecho numerosos para crear construcciones funcionales del tejido, hay pocas t&#xE9;cnicas disponibles para estimular la TEC de forma localizada. Se describen el dise&#xF1;o de un microdispositivo que puede estimular TEC de forma anisotr&#xF3;pica localizada, no homog&#xE9;nea, y predefinido utilizando ferromagnetically dopado polidimetilsiloxano microflaps (SFM). Espec&#xED;ficamente, un secuencial magneto-estructural modelo de elementos finitos del microdispositivo propuesto se construye y se utiliza para comprender c&#xF3;mo los cambios en las propiedades magn&#xE9;ticas y geom&#xE9;tricas del dispositivo afectar deflexi&#xF3;n MF. Nuestro estudio indica que una densidad relativamente peque&#xF1;a de material ferromagn&#xE9;tico se requiere para producir una fuerza adecuada y deserci&#xF3;n MF (175 micras aproximadamente 7% de deformaci&#xF3;n TCE). Tambi&#xE9;n demuestran que la MF a la distancia del im&#xE1;n es m&#xE1;s importante que la inherente a la permeabilidad magn&#xE9;tica en la determinaci&#xF3;n de MF MF deflexi&#xF3;n resultante. Una configuraci&#xF3;n de la validaci&#xF3;n de la prueba experimental se utiliz&#xF3; para validar las soluciones computacionales. La comparaci&#xF3;n muestra un acuerdo razonable que indica una diferencia de 5,9% entre los medidos experimentalmente y predecir computacionalmente el desplazamiento MF. En consecuencia, un aparato con dos FMs y dos imanes se ha hecho y est&#xE1; actualmente en fase de pruebas de construcci&#xF3;n. El presente estudio presenta el dise&#xF1;o de un nuevo microactuator magn&#xE9;tico para aplicaciones de ingenier&#xED;a de tejidos. Los resultados computacionales que aqu&#xED; se forman los cimientos en el dise&#xF1;o y optimizaci&#xF3;n de un micro funcional, con m&#xFA;ltiples y FMs imanes capaces de estimular los TECs en direcciones no homog&#xE9;nea y preferentes con resoluci&#xF3;n espacial relevante.

Cellular processes are regulated by various mechanical and physical factors in their local microenvironment such as geometric confinements, cell-substrate interactions, and cell-cell contact. Systematic elucidation of these regulatory mechanisms is crucial for fundamental understanding of cell biology and for rational design of biomedical devices and regenerative medicine. Here, we report a generally applicable plasma lithography technique, which performs selective surface functionalization on large substrate areas, for achieving long-term, stable confinements with length scales from 100 nm to 1 cm toward the investigation of cell-microenvironment interactions. In particular, we applied plasma lithography for cellular confinement of neuroblastomas, myoblasts, endothelial cells, and mammary gland epithelial cells, and examined the motion of mouse embryonic fibroblasts in directionality-confined environments for studying the effect of confinements on migratory behavior. In conjunction with live cell imaging, the distance traveled, velocity, and angular motion of individual cells and collective cell migration behaviors were measured in confined environments with dimensions comparable to a cell. A critical length scale that a cell could conceivably occupy and migrate to was also identified by investigating the behaviors of cells using confined environments with subcellular length scales.

Delucidazione sistematica di questi meccanismi di regolamentazione &egrave; cruciale per la fondamentale comprensione della biologia delle cellule e per la progettazione razionale di dispositivi biomedici e medicina rigenerativa. Riportiamo una tecnica di litografia in genere applicabili al plasma, che esegue la funzionalizzazione superficiale selettivo su aree grandi substrato, per il conseguimento di confini stabili, a lungo termine con scale di lunghezza da 100 nm a 1 cm verso l&#39;indagine delle interazioni cellula-microambiente. En particular, se aplic&#xF3; la litograf&#xED;a de plasma de confinamiento celular de los neuroblastomas, los mioblastos, c&#xE9;lulas endoteliales y las c&#xE9;lulas epiteliales de las gl&#xE1;ndulas mamarias, y se examina el movimiento de fibroblastos embrionarios de rat&#xF3;n en la direccionalidad de los ambientes cerrados para estudiar el efecto de los partos en el comportamiento migratorio. Sin embargo, &#xBF;c&#xF3;mo las c&#xE9;lulas procesan la informaci&#xF3;n geom&#xE9;trica a trav&#xE9;s de distancias mucho m&#xE1;s tiempo que las c&#xE9;lulas individuales de auto-organizarse en bien ordenado, alineados y fibras musculares multinucleadas sigue siendo una cuesti&#xF3;n central en la biolog&#xED;a del desarrollo y la medicina regenerativa. Usando micropatterning plasma la litograf&#xED;a para crear un entorno espacial de orientaci&#xF3;n celular, se muestra un mecanismo f&#xED;sico por el cual la informaci&#xF3;n de orientaci&#xF3;n se pueden propagar por una larga distancia del l&#xED;mite de una geom&#xE9;trica para guiar el desarrollo del tejido muscular. Myoblasts aggregate, differentiate and fuse to form skeletal muscle during both embryogenesis and tissue regeneration. For proper muscle function, long-range self-organization of myoblasts is required to create organized muscle architecture globally aligned to neighboring tissue. However, how the cells process geometric information over distances considerably longer than individual cells to self-organize into well-ordered, aligned and multinucleated myofibers remains a central question in developmental biology and regenerative medicine. Using plasma lithography micropatterning to create spatial cues for cell guidance, we show a physical mechanism by which orientation information can propagate for a long distance from a geometric boundary to guide development of muscle tissue. This long-range alignment occurs only in differentiating myoblasts, but not in non-fusing myoblasts perturbed by microfluidic disturbances or other non-fusing cell types. Computational cellular automata analysis of the spatiotemporal evolution of the self-organization process reveals that myogenic fusion in conjunction with rotational inertia functions in a self-reinforcing manner to enhance long-range propagation of alignment information. With this autocatalytic alignment feedback, well-ordered alignment of muscle could reinforce existing orientations and help promote proper arrangement with neighboring tissue and overall organization. Such physical self-enhancement might represent a fundamental mechanism for long-range pattern formation during tissue morphogenesis.

Tuttavia, come le cellule elaborano informazioni geometriche su distanze pi&ugrave; notevolmente di singole celle a si auto-organizzano in ben ordinato, allineati e multinucleate myofibers rimane una questione centrale nella biologia dello sviluppo e della medicina rigenerativa. Utilizzo di plasma litografia micropatterning per creare spunti spaziali per l&#39;orientamento delle cellule, mostriamo un meccanismo fisico che possono propagare informazioni orientamento per una lunga distanza da un confine geometrico per guidare lo sviluppo del tessuto muscolare. Questo allineamento lungo raggio si verifica solo nel differenziare mioblasti, ma non nella non-fusione myoblasts perturbato da microfluidica dispersioni o altri tipi di cellule non-fusione. Analisi computazionale automi cellulari dell&#39;evoluzione del processo di auto-organizzazione spatiotemporal rivelano che miogena fusion in combinazione con le funzioni di inerzia rotazionale in maniera autoalimentata per migliorare la propagazione a lunga distanza di informazioni sull&#39;allineamento. Con questo feedback allineamento autocatalitici, allineamento ben ordinato del muscolo potrebbe rafforzare gli orientamenti esistenti e contribuire a promuovere la corretta disposizione con tessuti limitrofi e l&#39;organizzazione complessiva. Tali self-enhancement fisico potrebbe rappresentare un meccanismo fondamentale per la formazione del modello a lungo raggio durante la morfogenesi dei tessuti.
