Although the treatment outcome of lymphoid malignancies has improved in recent years by the introduction of transplantation and antibody-based therapeutics, relapse remains a major problem. Therefore, new therapeutic options are urgently needed. One promising approach is the selective activation of apoptosis in tumor cells by the tumor necrosis factor-related apoptosis-inducing ligand (TRAIL). This study investigated the pro-apoptotic potential of a novel TRAIL fusion protein designated scFvCD19:sTRAIL, consisting of a CD19-specific single-chain Fv antibody fragment (scFv) fused to the soluble extracellular domain of TRAIL (sTRAIL). Potent apoptosis was induced by scFvCD19:sTRAIL in several CD19-positive tumor cell lines, whereas normal blood cells remained unaffected. In mixed culture experiments, selective binding of scFvCD19:sTRAIL to CD19-positive cells resulted in strong induction of apoptosis in CD19-negative bystander tumor cells. Simultaneous treatment of CD19-positive cell lines with scFvCD19:sTRAIL and valproic acid (VPA) or Cyclosporin A induced strongly synergistic apoptosis. Treatment of patient-derived acute B-lymphoblastic leukemia (B-ALL) and chronic B-lymphocytic leukemia (B-CLL) cells resulted in strong tumoricidal activity that was further enhanced by combination with VPA. In addition, scFvCD19:sTRAIL prevented engraftment of human Nalm-6 cells in xenotransplanted NOD/Scid mice. The pre-clinical data presented here warrant further investigation of scFvCD19:sTRAIL as a potential new therapeutic agent for CD19-positive B-lineage malignancies.

Bien que les r&eacute;sultats du traitement des malignit&eacute;s lympho&iuml;des a am&eacute;lior&eacute; ces derni&egrave;res ann&eacute;es par l&#39;introduction de la transplantation et de produits th&eacute;rapeutiques &agrave; base d&#39;anticorps, rechute demeure un probl&egrave;me majeur. Par cons&eacute;quent, nouvelles options th&eacute;rapeutiques sont urgent. Une approche prometteuse est l&#39;activation s&eacute;lective de l&#39;apoptose dans les cellules tumorales par le ligand induisant l&#39;apoptose ax&eacute;s sur le facteur de n&eacute;crose tumorale (TRAIL). En outre, scFvCD19:sTRAIL emp&ecirc;ch&eacute; de greffe de cellules humaines de Nalm-6 chez les souris NOD/Scid transplant&eacute;es. Les donn&eacute;es pr&eacute;cliniques pr&eacute;sent&eacute;es ici justifient plus approfondi de scFvCD19:sTRAIL comme un potentiel nouvel agent th&eacute;rapeutique pour les tumeurs malignes lign&eacute;e B CD19-positif.

Neue therapeutische Optionen sind daher dringend erforderlich. Ein vielversprechender Ansatz ist die selektive Aktivierung der Apoptose in Tumorzellen durch den Tumor-Nekrose-Faktor-bezogene Apoptose induzierenden Liganden (TRAIL). Diese Studie untersucht das Potenzial von Pro-apoptotischen eines Romans TRAIL Schmelzverfahren Protein benannten ScFvCD19:sTRAIL, bestehend aus einem CD19-spezifische Einzel-Kette Fv Antik&ouml;rper Fragment (ScFv) der l&ouml;slichen extrazellul&auml;ren Dom&auml;ne des Trails (sTRAIL) verschmolzen. Dar&uuml;ber hinaus verhindert die ScFvCD19:sTRAIL Verteilung menschlicher Nalm-6-Zellen in Xenotransplanted NOD/Scid M&auml;use. Die hier vorgestellten pr&auml;klinischen Daten garantieren weitere Untersuchungen des ScFvCD19:sTRAIL als potenzielle neue Therapeutikum f&uuml;r CD19-positiven B-Linie Malignome.

Anche se il risultato del trattamento di neoplasie linfoidi &egrave; migliorata negli ultimi anni dall&#39;introduzione di trapianti e terapie basate su anticorpi, ricaduta rimane un grave problema. Quindi, nuove opzioni terapeutiche sono urgentemente necessarie. Un approccio promettente &egrave; l&#39;attivazione selettiva di apoptosi nelle cellule tumorali dal fattore di necrosi tumorale-correlati che inducono apoptosi ligand (sentiero). Trattamento del paziente-derivato B-leucemia linfoblastica acuta (B-ALL) e le cellule della leucemia B-lymphocytic cronica (B-CLL) portato in attivit&agrave; tumoricidal forte che &egrave; stata ulteriormente rafforzata dalla combinazione con VPA. Inoltre, scFvCD19:sTRAIL ha impedito di attecchimento delle cellule umane Nalm-6 in topi NOD/Scid xenotrapiantati. I dati preclinici presentati qui giustificano ulteriori indagini di scFvCD19:sTRAIL come un potenziale nuovo agente terapeutico per i tumori maligni CD19 positivi B-lineage.

Aunque el resultado del tratamiento de malignidades linfoides ha mejorado en los &uacute;ltimos a&ntilde;os por la introducci&oacute;n de trasplante y terapias basadas en anticuerpos, reca&iacute;da sigue siendo un problema importante. Por lo tanto, se necesitan urgentemente nuevas opciones terap&eacute;uticas. Un enfoque prometedor es la activaci&oacute;n selectiva de apoptosis en las c&eacute;lulas tumorales por los relacionados con el factor de necrosis tumoral que inducen apoptosis ligando (TRAIL). Este estudio investig&oacute; el potencial pro-apopt&oacute;tica de una novela de prote&iacute;na de fusi&oacute;n del sendero se&ntilde;alada scFvCD19:sTRAIL, consistiendo en un CD19 espec&iacute;fica cadena simple Fv anticuerpo fragmento (scFv) fundido al dominio extracelular soluble del camino (sTRAIL).
