Tissue Targeted Embryonic Chimeras: Zebrafish Gastrula Cell Transplantation
Elizabeth R. Deschene 1, Michael J. Barresi 1
1Department of Biological Sciences, Smith College

Zebrafish cell transplantation enables the combination of genetics and embryology to generate tissue specific chimeras. This video demonstrates gastrula staged cell transplantations that have allowed our lab to investigate the roles of astroglial populations and specific guidance cues during commissure formation in the forebrain.

A straight-forward set of methods to isolate and determine the identity of the most abundant proteins expressed in skeletal muscle. About 800 spots are discerned on a two-dimensional gel from 10 mg muscle; this allows for the determination of gender-specific protein expression. These methods will give equivalent results in most tissues.

Production of Dynein and Kinesin Motor Ensembles on DNA Origami Nanostructures for Single Molecule Observation
Jingjie Hu 1, Nathan D. Derr 1,2
1Department of Biological Sciences, Smith College, 2Center for Microscopy and Imaging, Smith College

The goal of this protocol is to form ensembles of molecular motors on DNA origami nanostructures and observe the ensemble motility using total internal reflection fluorescence microscopy.
