Development of normal colon epithelial cells proceeds through a systematic differentiation of cells that emerge from stem cells within the base of colon crypts. Genetic mutations in the adenomatous polyposis coli (APC) gene are thought to cause colon adenoma and carcinoma formation by enhancing colonocyte proliferation and impairing differentiation. We currently have a limited understanding of the cellular mechanisms that promote colonocyte differentiation. Herein, we present evidence supporting a lack of retinoic acid biosynthesis as a mechanism contributing to the development of colon adenomas and carcinomas. Microarray and reverse transcriptase-PCR analyses revealed reduced expression of two retinoid biosynthesis genes: retinol dehydrogenase 5 (RDH5) and retinol dehydrogenase L (RDHL) in colon adenomas and carcinomas as compared with normal colon. Consistent with the adenoma and carcinomas samples, seven colon carcinoma cell lines also lacked expression of RDH5 and RDHL. Assessment of RDH enzymatic activity within these seven cell lines showed poor conversion of retinol into retinoic acid when compared with normal cells such as normal human mammary epithelial cells. Reintroduction of wild type APC into an APC-deficient colon carcinoma cell line (HT29) resulted in increased expression of RDHL without affecting RDH5. APC-mediated induction of RDHL was paralleled by increased production of retinoic acid. Investigations into the mechanism responsible for APC induction of RDHL indicated that beta-catenin fails to repress RDHL. The colon-specific transcription factor CDX2, however, activated an RDHL promoter construct and induced endogenous RDHL. Finally, the induction of RDHL by APC appears dependent on the presence of CDX2. We propose a novel role for APC and CDX2 in controlling retinoic acid biosynthesis and in promoting a retinoid-induced program of colonocyte differentiation.

The zebrafish is an attractive vertebrate model for genetic studies of development, apoptosis, and cancer. Here we describe a transgenic zebrafish line in which T- and B-lymphoid cells express a fusion transgene that encodes the zebrafish bcl-2 protein fused to the enhanced green fluorescence protein (EGFP). Targeting EGFP-bcl-2 to the developing thymocytes of transgenic fish resulted in a 2.5-fold increase in thymocyte numbers and a 1.8-fold increase in GFP-labeled B cells in the kidney marrow. Fluorescent microscopic analysis of living rag2-EGFP-bcl-2 transgenic fish showed that their thymocytes were resistant to irradiation- and dexamethasone-induced apoptosis, when compared with control rag2-GFP transgenic zebrafish. To test the ability of bcl-2 to block irradiation-induced apoptosis in malignant cells, we compared the responsiveness of Myc-induced leukemias with and without EGFP-bcl-2 expression in living transgenic zebrafish. T-cell leukemias induced by the rag2-EGFP-Myc transgene were ablated by irradiation, whereas leukemias in double transgenic fish expressing both Myc and EGFP-bcl-2 were resistant to irradiation-induced apoptotic cell death. The forward genetic capacity of the zebrafish model system and the ability to monitor GFP-positive thymocytes in vivo make this an ideal transgenic line for modifier screens designed to identify genetic mutations or small molecules that modify bcl-2-mediated antiapoptotic pathways.

Suppression of apoptosis by bcl-2 overexpression in lymphoid cells of transgenic zebrafish.

Le poisson z&#xE8;bre est un mod&#xE8;le vert&#xE9;br&#xE9; attractif pour des &#xE9;tudes g&#xE9;n&#xE9;tiques du d&#xE9;veloppement, l&#x26;#39;apoptose et le cancer. Nous d&#xE9;crivons ici une ligne poisson-z&#xE8;bre transg&#xE9;nique dans laquelle les cellules T et B-lympho&#xEF;des exprimer un transg&#xE8;ne codant pour la fusion du poisson-z&#xE8;bre bcl-2 prot&#xE9;ine fusionn&#xE9;e &#xE0; la prot&#xE9;ine de fluorescence verte am&#xE9;lior&#xE9;e (EGFP). Ciblage EGFP-bcl-2 &#xE0; des thymocytes en d&#xE9;veloppement de poissons transg&#xE9;niques a entra&#xEE;n&#xE9; une augmentation de 2,5 fois le nombre de thymocytes et une augmentation de 1,8 fois en GFP marqu&#xE9;s cellules B dans la moelle des reins. Fluorescent analyse microscopique de la vie rag2-EGFP-bcl-2 poissons transg&#xE9;niques ont montr&#xE9; que leurs thymocytes &#xE9;taient r&#xE9;sistants &#xE0; l&#x26;#39;irradiation et l&#x26;#39;apoptose induite par la dexam&#xE9;thasone, en comparaison avec le contr&#xF4;le rag2-GFP transg&#xE9;nique poisson-z&#xE8;bre. T-cellules leuc&#xE9;mies induites par le transg&#xE8;ne rag2-EGFP-Myc ont &#xE9;t&#xE9; ablation par irradiation, alors que les leuc&#xE9;mies chez les poissons transg&#xE9;niques exprimant &#xE0; la fois &#xE0; double Myc et EGFP-bcl-2 &#xE9;taient r&#xE9;sistants &#xE0; l&#x26;#39;irradiation induite par la mort cellulaire par apoptose. Il Danio rerio &egrave; un attraente modello vertebrato per studi di genetica dello sviluppo, apoptosi e cancro. Qui descriviamo una linea transgenica di zebrafish in cui le cellule T e B-linfoidi esprimono un transgene di fusione che codifica la proteina bcl-2 Danio rerio fuse insieme alla proteina maggiore fluorescenza verde (EGFP). Targeting EGFP-bcl-2 per i timociti in via di sviluppo di pesci transgenici ha provocato un d&#39;aumento in numeri timocita e 1.8-fold in cellule GFP-etichettato B nel midollo renale. Fluorescente analisi microscopica del pesce transgenico vivo rag2-EGFP-bcl-2 ha mostrato che la loro timociti erano resistenti all&#39;apoptosi indotta da irradiazione e desametasone, quando confrontato con controllo rag2-GFP transgenici zebrafish. Per testare la capacit&agrave; di bloccare l&#39;apoptosi indotta da irradiazione in cellule maligne bcl-2, abbiamo confrontato la reattivit&agrave; di leucemie indotta da Myc con e senza espressione EGFP-bcl-2 in zebrafish transgenici vivente. Leucemie T-cellulare indotte da Myc-rag2-EGFP transgene erano ablazione di irradiazione, mentre leucemie in doppi pesci transgenici che esprimono Myc sia EGFP-bcl-2 erano resistenti alla morte cellulare apoptotica indotta da irradiazione. La capacit&agrave; genetica avanti del sistema modello zebrafish e la capacit&agrave; di monitorare i timociti GFP-positivi in vivo fanno di questo una linea transgenica ideale per schermi modificatore progettato per identificare le mutazioni genetiche o piccole molecole che modificano i percorsi antiapoptotici mediata da bcl-2.

Aqu&#xED; se describe una l&#xED;nea de pez cebra transg&#xE9;nico en el que las c&#xE9;lulas T y B-linfoides expresan un transg&#xE9;n que codifica la fusi&#xF3;n del pez cebra prote&#xED;na Bcl-2 fusionada a la prote&#xED;na de fluorescencia verde mayor (EGFP). Orientaci&#xF3;n EGFP-Bcl-2 a los timocitos en desarrollo de peces transg&#xE9;nicos como resultado un aumento de 2,5 veces en el n&#xFA;mero de timocitos y un aumento de 1,8 veces en las buenas pr&#xE1;cticas agrarias marcadas con las c&#xE9;lulas B en la m&#xE9;dula renal. The zebrafish (Danio rerio) has proven to be a powerful vertebrate model system for the genetic analysis of developmental pathways and is only beginning to be exploited as a model for human disease and clinical research. The attributes that have led to the emergence of the zebrafish as a preeminent embryological model, including its capacity for forward and reverse genetic analyses, provides a unique opportunity to uncover novel insights into the molecular genetics of cancer. Some of the advantages of the zebrafish animal model system include fecundity, with each female capable of laying 200-300 eggs per week, external fertilization that permits manipulation of embryos ex utero, and rapid development of optically clear embryos, which allows the direct observation of developing internal organs and tissues in vivo. The zebrafish is amenable to transgenic and both forward and reverse genetic strategies that can be used to identify or generate zebrafish models of different types of cancer and may also present significant advantages for the discovery of tumor suppressor genes that promote tumorigenesis when mutationally inactivated. Importantly, the transparency and accessibility of the zebrafish embryo allows the unprecedented direct analysis of pathologic processes in vivo, including neoplastic cell transformation and tumorigenic progression. Ultimately, high-throughput modifier screens based on zebrafish cancer models can lead to the identification of chemicals or genes involved in the suppression or prevention of the malignant phenotype. The identification of small molecules or gene products through such screens will serve as ideal entry points for novel drug development for the treatment of cancer. This review focuses on the current technology that takes advantage of the zebrafish model system to further our understanding of the genetic basis of cancer and its treatment.

Making waves in cancer research: new models in the zebrafish.

Evasion of DNA damage-induced cell death, via mutation of the p53 tumor suppressor or overexpression of prosurvival Bcl-2 family proteins, is a key step toward malignant transformation and therapeutic resistance. We report that depletion or acute inhibition of checkpoint kinase 1 (Chk1) is sufficient to restore gamma-radiation-induced apoptosis in p53 mutant zebrafish embryos. Surprisingly, caspase-3 is not activated prior to DNA fragmentation, in contrast to classical intrinsic or extrinsic apoptosis. Rather, an alternative apoptotic program is engaged that cell autonomously requires atm (ataxia telangiectasia mutated), atr (ATM and Rad3-related) and caspase-2, and is not affected by p53 loss or overexpression of bcl-2/xl. Similarly, Chk1 inhibitor-treated human tumor cells hyperactivate ATM, ATR, and caspase-2 after gamma-radiation and trigger a caspase-2-dependent apoptotic program that bypasses p53 deficiency and excess Bcl-2. The evolutionarily conserved "Chk1-suppressed" pathway defines a novel apoptotic process, whose responsiveness to Chk1 inhibitors and insensitivity to p53 and BCL2 alterations have important implications for cancer therapy.

The molecular events underlying the progression of T-lymphoblastic lymphoma (T-LBL) to acute T-lymphoblastic leukemia (T-ALL) remain elusive. In our zebrafish model, concomitant overexpression of bcl-2 with Myc accelerated T-LBL onset while inhibiting progression to T-ALL. The T-LBL cells failed to invade the vasculature and showed evidence of increased homotypic cell-cell adhesion and autophagy. Further analysis using clinical biopsy specimens revealed autophagy and increased levels of BCL2, S1P1, and ICAM1 in human T-LBL compared with T-ALL. Inhibition of S1P1 signaling in T-LBL cells led to decreased homotypic adhesion inÂ vitro and increased tumor cell intravasation inÂ vivo. Thus, blockade of intravasation and hematologic dissemination in T-LBL is due to elevated S1P1 signaling, increased expression of ICAM1, and augmented homotypic cell-cell adhesion.

Les &#xE9;v&#xE9;nements mol&#xE9;culaires qui sous-tendent la progression de la T-lymphoblastique lymphome (T-LBL) pour T-aigu&#xEB; leuc&#xE9;mie lymphoblastique (T-ALL) demeurent insaisissables. Dans notre mod&#xE8;le de poisson z&#xE8;bre, la surexpression concomitante de bcl-2 avec Myc acc&#xE9;l&#xE9;r&#xE9;e T-LBL apparition tandis inhiber la progression de T-ALL. Les cellules T-LBL pas &#xE0; envahir le syst&#xE8;me vasculaire et montraient des signes de plus homotypique adh&#xE9;sion cellule-cellule et l&#x26;#39;autophagie. In unserem Modell Zebrafisch beschleunigt damit einhergehende eine &Uuml;berexpression von Bcl-2 mit Myc T-LBL Ausbruch w&auml;hrend Hemmung Fortschreiten zu T-ALL. Die T-LBL-Zellen konnte das Gef&auml;&szlig;system einzudringen und zeigte Anzeichen f&uuml;r erh&ouml;hte homotypischen Zell-Zell-Adh&auml;sion und Autophagy. Weitere Analyse mit klinischen Biopsie Proben offenbart Autophagy und Steigerung des BCL2, S1P1 und ICAM1 in menschlichen T-LBL gegen&uuml;ber T-ALL. Hemmung der S1P1 signalisieren in T-LBL Zellen f&uuml;hrte zur sank homotypischen Adh&auml;sion in Vitro und erh&ouml;hte Tumor Zelle Intravasation in-vivo. En nuestro modelo de pez cebra, la sobreexpresi&#xF3;n simult&#xE1;nea de Bcl-2 con Myc acelerada T-LBL inicio, mientras que inhibir la progresi&#xF3;n de la LLA-T. Las c&#xE9;lulas T-LBL no invadir la vasculatura y mostraron evidencia de un aumento homotipica c&#xE9;lulas de adhesi&#xF3;n celular y la autofagia. Endoplasmic reticulum (ER) stress triggers tissue-specific responses that culminate in either cellular adaptation or apoptosis, but the genetic networks distinguishing these responses are not well understood. Here we demonstrate that ER stress induced in the developing zebrafish causes rapid apoptosis in the brain, spinal cord, tail epidermis, lens, and epiphysis. Focusing on the tail epidermis, we uncover an apoptotic response that depends on Puma, but not on p53 or Chop. puma is transcriptionally activated during this ER stress response in a p53-independent manner, and is an essential mediator of epidermal apoptosis. We demonstrate that the p63 transcription factor is upregulated to initiate this apoptotic pathway and directly activates puma transcription in response to ER stress. We also show that a mutation of human Connexin 31, which causes erythrokeratoderma variabilis, induces ER stress and p63-dependent epidermal apoptosis in the zebrafish embryo, thus implicating this pathway in the pathogenesis of inherited disease.

DNA double-strand breaks (DSBs) represent one of the most deleterious forms of DNA damage to a cell. In cancer therapy, induction of cell death by DNA DSBs by ionizing radiation (IR) and certain chemotherapies is thought to mediate the successful elimination of cancer cells. However, cancer cells often evolve to evade the cytotoxicity induced by DNA DSBs, thereby forming the basis for treatment resistance. As such, a better understanding of the DSB DNA damage response (DSB-DDR) pathway will facilitate the design of more effective strategies to overcome chemo- and radioresistance. To identify novel mechanisms that protect cells from the cytotoxic effects of DNA DSBs, we performed a forward genetic screen in zebrafish for recessive mutations that enhance the IR-induced apoptotic response. Here, we describe radiosensitizing mutation 7 (rs7), which causes a severe sensitivity of zebrafish embryonic neurons to IR-induced apoptosis and is required for the proper development of the central nervous system. The rs7 mutation disrupts the coding sequence of ccdc94, a highly conserved gene that has no previous links to the DSB-DDR pathway. We demonstrate that Ccdc94 is a functional member of the Prp19 complex and that genetic knockdown of core members of this complex causes increased sensitivity to IR-induced apoptosis. We further show that Ccdc94 and the Prp19 complex protect cells from IR-induced apoptosis by repressing the expression of p53 mRNA. In summary, we have identified a new gene regulating a dosage-sensitive response to DNA DSBs during embryonic development. Future studies in human cancer cells will determine whether pharmacological inactivation of CCDC94 reduces the threshold of the cancer cell apoptotic response.
