&#x4ECE;&#x6297;&#x539F;&#x7279;&#x5F02;&#x6027; CD8 T &#x7EC6;&#x80DE;&#x7684;&#x6297;&#x80BF;&#x7624;&#x6D3B;&#x6027;&#x751F;&#x6210;&#x4ECE;&#x8F6C;&#x57FA;&#x56E0;&#x9020;&#x8840;&#x5E72;&#x7EC6;&#x80DE;&#x5728;&#x4F53;&#x5185;&#x3002;

The goal of cancer immunotherapy is the generation of an effective, stable, and self-renewing antitumor T-cell population. One such approach involves the use of high-affinity cancer-specific T-cell receptors in gene-therapy protocols. Here, we present the generation of functional tumor-specific human T cells in vivo from genetically modified human hematopoietic stem cells (hHSC) using a human/mouse chimera model. Transduced hHSC expressing an HLA-A*0201-restricted melanoma-specific T-cell receptor were introduced into humanized mice, resulting in the generation of a sizeable melanoma-specific naÃ¯ve CD8(+) T-cell population. Following tumor challenge, these transgenic CD8(+) T cells, in the absence of additional manipulation, limited and cleared human melanoma tumors in vivo. Furthermore, the genetically enhanced T cells underwent proper thymic selection, because we did not observe any responses against non-HLA-matched tumors, and no killing of any kind occurred in the absence of a human thymus. Finally, the transduced hHSC established long-term bone marrow engraftment. These studies present a potential therapeutic approach and an important tool to understand better and to optimize the human immune response to melanoma and, potentially, to other types of cancer.

Antitumor activity from antigen-specific CD8 T cells generated in vivo from genetically engineered human hematopoietic stem cells.

Le but de l&#39;immunoth&eacute;rapie du cancer est la g&eacute;n&eacute;ration d&#39;une population de lymphocytes antitumorale autorenouvellement, stable et efficace. Une telle approche implique l&#39;utilisation de r&eacute;cepteurs de haute affinit&eacute; sp&eacute;cifique du cancer lymphocytes dans des protocoles de th&eacute;rapie g&eacute;nique. Nous pr&eacute;sentons ici la g&eacute;n&eacute;ration de fonctionnelles sp&eacute;cifique des cellules T in vivo &agrave; partir d&#39;organismes g&eacute;n&eacute;tiquement modifi&eacute;s cellules souches h&eacute;matopo&iuml;&eacute;tiques humaines (hHSC) en utilisant un mod&egrave;le de chim&egrave;re humain/souris. Transduite hHSC exprimant un r&eacute;cepteur des cellules T de m&eacute;lanome sp&eacute;cifiques restriction HLA-A * 0201 ont &eacute;t&eacute; introduits dans des souris humanis&eacute;es, ce qui entra&icirc;ne la g&eacute;n&eacute;ration d&#39;une population de lymphocytes T CD8 na&iuml;f de m&eacute;lanome sp&eacute;cifiques non n&eacute;gligeable. Apr&egrave;s le d&eacute;fi de la tumeur, ces cellules T CD8 transg&eacute;niques, en l&#39;absence de manipulation suppl&eacute;mentaire, limit&eacute; et effac&eacute; des tumeurs de m&eacute;lanome humain in vivo. En outre, les lymphocytes T g&eacute;n&eacute;tiquement am&eacute;lior&eacute;s a subi une s&eacute;lection thymique, parce que nous n&#39;avons pas observ&eacute; les r&eacute;ponses contre les tumeurs non-HLA-assortie, et aucun meurtre quelconque a eu lieu en l&#39;absence d&#39;un thymus humain. Enfin, le transduite hHSC &eacute;tabli &agrave; long terme prise de greffe de moelle osseuse. Ces &eacute;tudes pr&eacute;sentent une approche th&eacute;rapeutique potentielle et un outil important pour mieux comprendre et d&#39;optimiser la r&eacute;ponse immunitaire humaine m&eacute;lanome et, &eacute;ventuellement, &agrave; d&#39;autres types de cancer.

Das Ziel der Krebs-Immuntherapie ist die Generierung einer wirksamen, stabilen und erneuernde antitumor T-Zell-Bev&ouml;lkerung. Einen solchen Ansatz beinhaltet die Verwendung von hoher Affinit&auml;t Krebs-spezifische T-Zell Rezeptoren in Gentherapie Protokolle. Hier pr&auml;sentieren wir die Generation der funktionalen tumorspezifische menschlichen T-Zellen in-vivo aus gentechnisch ver&auml;nderten h&auml;matopoetische Stammzellen (hHSC) mit einem Mensch/Maus-Chim&auml;re-Modell. Transduzierten hHSC Ausdruck einer HLA-A * 0201 eingeschr&auml;nkten Melanom-spezifische T-Zell-Rezeptor eingef&uuml;hrt worden in humanisierte M&auml;use, wodurch die Generierung einer betr&auml;chtlichen Melanom-spezifische naiv-CD8(+) T-Zell-Population. Nach Tumor-Herausforderung diese transgenen CD8(+) T-Zellen, ohne zus&auml;tzliche Manipulation, begrenzt und deaktiviert menschlichen Melanom Tumoren in-vivo. Dar&uuml;ber hinaus wurde die genetisch verbesserten T-Zellen richtige Thymom Auswahl, denn wir keine Antworten gegen Tumoren nicht HLA-passende beobachten, und keine T&ouml;tung jeglicher Art aufgetreten, in Ermangelung eines menschlichen Thymus ist. Schlie&szlig;lich gr&uuml;ndete die transduzierten hHSC langfristige Knochenmark-Verteilung. Diese Studien stellen einen m&ouml;glichen therapeutischen Ansatz und ein wichtiges Instrument, um besser zu verstehen und die menschlichen Immunantwort zu Melanom und eventuell auch auf andere Arten von Krebs zu optimieren.

Presentiamo qui, la generazione di funzionale specifico tumore T cellule umane in vivo da geneticamente modificate umane cellule staminali ematopoietiche (hHSC) utilizzando un modello chimera umana e mouse. Infine, la hHSC trasdotte stabilito attecchimento a lungo termine di midollo osseo. Questi studi presentano un potenziale approccio terapeutico e un importante strumento per comprendere meglio e a ottimizzare la risposta immunitaria umana di melanoma e, potenzialmente, ad altri tipi di cancro.

El objetivo de inmunoterapia del c&aacute;ncer es la generaci&oacute;n de una poblaci&oacute;n de c&eacute;lulas T antitumoral eficaz, estable y renueva a s&iacute; misma. Uno de los enfoques implica el uso de receptores de alta afinidad espec&iacute;fica por c&aacute;ncer c&eacute;lula T en protocolos de terapia g&eacute;nica. Aqu&iacute;, presentamos la generaci&oacute;n de funcionales tumor-espec&iacute;ficos T c&eacute;lulas humanas in vivo de c&eacute;lulas madre hematopoy&eacute;ticas humanas gen&eacute;ticamente modificadas (hHSC) mediante un modelo de quimera humanos/rat&oacute;n. HHSC transduced expresando un receptor de c&eacute;lulas T espec&iacute;ficas melanoma restricci&oacute;n HLA-A * 0201 fueron introducidos en ratones humanizados, dando por resultado la generaci&oacute;n de una poblaci&oacute;n de c&eacute;lulas T CD8 considerable ingenuo de melanoma espec&iacute;fico. Tras el desaf&iacute;o de tumor, estas c&eacute;lulas CD8 T transg&eacute;nicas, en ausencia de manipulaci&oacute;n adicional, hab&iacute;a limitada y despejaron in vivo de tumores de melanoma humano. Adem&aacute;s, las c&eacute;lulas T mejoradas gen&eacute;ticamente experimentaron adecuada selecci&oacute;n t&iacute;mica, porque no observamos las respuestas contra tumores no-HLA-emparejado, y ning&uacute;n asesinato de cualquier tipo se produjo en ausencia de un timo humano. Por &uacute;ltimo, la hHSC transduced hab&iacute;a establecido injerto de m&eacute;dula &oacute;sea a largo plazo. Estos estudios presentan un enfoque terap&eacute;utico potencial y una herramienta importante para comprender mejor y optimizar la respuesta inmune humana al melanoma y, potencialmente, a otros tipos de c&aacute;ncer.
