1. Skull Penetrazione <ol><li> Anestetizzare ratto con isofluorano (induzione 5%, 1,5-2% di manutenzione) in 70% N 2 e 30% O 2. Dopo l&#39;induzione dell&#39;anestesia, posizionare il topo prona su una piastra riscaldante, posizionando il naso del topo in un cono di naso anestetico. </li><li> Pur mantenendo l&#39;anestesia, fissare la testa in un frame stereotassico, inserendo le orecchie-bar fino a quando la testa si è stabilizzata. Assicurarsi che la respirazione non è compromessa. (Figura 2-A). </li><li> Iniettare cuoio capelluto per via sottocutanea con anestetico locale a lunga durata, bupivacaina 0,3 ml 0,5% (Pfizer, Australia) prima di prendere una skin 1,5 centimetri un&#39;incisione mediana testa. (Strumenti sterili e guanti devono essere utilizzati.) </li><li> Smussare sezionare i tessuti molli e dei muscoli circostanti per individuare in modo chiaro e Lambda Bregma. Ritrarre la pelle e del tessuto connettivo. </li><li> Stelo qualsiasi sanguinamento applicando pressione al cranio esposto. Cranio sanguinamento eccessivo può essere cauterizzato. </li><li> Utilizzo di un dentale drill con una punta di 1 mm radica, radica di 2 mm ampio foro nell&#39;osso parietale destro. Burr da 2 mm foro laterale e 2 mm posteriormente da Bregma per evitare il seno longitudinale superiore e assicurare il posizionamento del sensore ICP è sul territorio ischemico, ictus per studi. Ubicazioni alternative sia ugualmente adatta per altre applicazioni. Burr il foro ad una profondità dove il cranio sopra la dura diventa traslucida. (Figura 2-B). </li><li> Sostituire la bava con una punta da 0,5 mm bave per rimuovere il cranio alla base del foro. </li><li> Quando il cranio comincia a incrinarsi, usare pinze a 45 ° per rimuovere tutti i rimanenti cranio, assicurando la base del foro è liberata dai detriti. (Figura 3). </li></ol> 2. Vite Modifica e inserimento <ol><li> Praticare un foro di 0,7 millimetri attraverso una vite con testa esagonale usando un tornio e lo 0,7 punta mm. </li><li> Inserire la vite di monitoraggio nel foro ruotandolo in circa 1,5 giri (usare la quantità minima didiventa necessario a garantire la vite nel cranio in modo da non danneggiare il tessuto sottostante). (Figura 2-C e Figura 4). </li><li> Burr un secondo foro per una vite di ancoraggio nell&#39;osso parietale sinistro, 2 mm e 2 mm laterale posteriore da Bregma. Questo foro non richiede la penetrazione completa del cranio, in modo che il mm 1 punta bava viene usato per fluidificare il cranio per l&#39;inserimento della vite. </li><li> Inserire un x 2 4 a testa esagonale da mm vite nel secondo foro. Questa vite aiuta ad ancorare il cemento dentale e quindi la vite di monitoraggio al cranio. </li><li> Utilizzare una pipetta di trasferimento per mescolare e applicare monomero e polimero cemento dentale alla base della testa delle viti. </li><li> Lasciare cemento dentale ad asciugare per almeno 10 minuti. </li></ol> 3. Inserimento del trasduttore di pressione intracranica <ol><li> Utilizzo di liquido bianco correzione, segnare la fibra ottica sensore 4 mm dalla punta. </li><li> Riempire il foro della vite di monitoraggio con soluzione salina sterile (0,9%) e non garantisconobolle d&#39;aria sono presenti all&#39;interno della vite. </li><li> Inserire la ICP sonda 4 mm nella vite in modo che la punta della sonda è a livello con l&#39;estremità della vite. Assicurarsi che la punta non perforare la dura. </li><li> Regolare la punta della sonda all&#39;interno della vite fino a una traccia ICP riflettente ventilazione e sangue onde a impulsi di pressione può essere osservato. (Figura 5). </li></ol> 4. Formare una chiusura ermetica <ol><li> Una chiusura ermetica è indispensabile per una lettura accurata ICP. Mescolare un viscoso monomero biocompatibile materiale di sigillatura e polimero nel rapporto di 1:1. Poiché il sensore di pressione è sulla punta della sonda, e questo è all&#39;interno della vite coppellate, l&#39;applicazione di materiale di sigillatura all&#39;albero della fibra ottica sonda ha alcun effetto sulla sensibilità del sensore di pressione. </li><li> Applicare uno strato sottile della sonda e la testa della vite di monitoraggio. Evitare di spostare la sonda ICP. </li><li> Lasciare riposare per 5 minuti. </li><li> Applicare un laico secondoer del materiale di sigillatura attorno alla vite di monitoraggio intera e sonda. Assicurarsi che non vi siano perdite di liquido da eventuali crepacci nel materiale di coibentazione. (Figura 2-D). </li><li> Rimuovere le orecchie-bar. </li><li> Il ratto può rimanere in posizione prona, o essere attentamente ruotato nella posizione supina durante il monitoraggio ICP. </li><li> Uno schema della procedura completata è rappresentato in (Figura 6). </li></ol> 5. Trasduttore di pressione intracranica rimozione e il reinserimento <ol><li> Al completamento monitoraggio ICP, il sensore ICP può essere rimosso tirando delicatamente il catetere dal materiale vite e sigillatura. </li><li> Il sensore di SAMBA deve essere immediatamente posto in 1% TERG-A-Zyme soluzione per prevenire la corrosione della punta. </li><li> Il foro rimanente nel materiale di sigillatura deve essere coperto con uno strato aggiuntivo di materiale di sigillatura. (Rat può essere svegliato in questa fase). </li><li> Per reinserire il catetere SAMBA per il monitoraggio aggiuntivo, affettareil materiale di sigillatura a livello della testa della vite. </li><li> Ripetere i passaggi 3.2 - 5.3. </li></ol> 6. Risultati rappresentativi Figura 5 è una rappresentazione di letture ICP oltre dieci secondi. Al basale, l&#39;ICP medio in un ratto Wistar è di 6 mmHg. Gli eventi di più breve periodicità illustrato nella figura 5 riflettere onde a impulso di pressione del sangue. Gli eventi di più lunga periodicità visualizzare gli eventi di ventilazione. Si noti che il sensore SAMBA riflette una ampiezza di ventilazione mmHg 3-4 e ampiezza dell&#39;impulso di 1-2 mmHg. Per convalidare la posizione del sensore SAMBA in ciascun esperimento, le tracce ICP devono essere testati per la reattività di compressioni addominali ed eventi respiratorie, come i periodi di apnea. Una compressione addominale è illustrato in Figura 7. Periodi di apnea (illustrata nella figura 8) si osservano in più esperimenti Invo<html> lving respiro spontaneo animali. Questi eventi sono identificati sulle registrazioni fisiologiche, l&#39;assenza di deviazioni su respiratorie delle vie respiratorie (trasduttore diaframma) e tracce di pressione arteriosa. Un&#39;alterazione equivalente nella traccia ICP convalida il posizionamento della sonda ICP. Figura 9 illustra una tipica traccia ICP dopo la rimozione delle barre auricolari (passo 4.6). L&#39;inserimento delle barre di auricolari in fase 1.2 risultati in una leggera compressione del cranio e la conseguente interruzione del volume intracranico e quindi un aumento della ICP. Se il sensore è posizionato correttamente, ICP scenderà almeno 4-5 mmHg con la rimozione delle barre dell&#39;orecchio. L&#39;analisi istologica può essere utilizzato per verificare eventuali danni alla zona corticale immediatamente sotto il sensore di pressione e la vite. La procedura qui presentata consente una registrazione molto sensibile e preciso della pressione intracranica. Questa tecnica minimamente invasiva evita traumi significativi cervello posizionando il sensore di pressione nello spazio epidurale e non il tessuto cerebrale o ventricoli. I passaggi critici sono: 1) di perforazione attraverso il cranio - bisogna fare attenzione a non forare la durata o danno sotteso tessuto cerebrale, 2) garantire una tenuta perfetta con il materiale di sigillatur...

epidural intracranial pressure measurement in rats using a fiber-optic pressure transducer

L&#39;elevata pressione intracranica (ICP) è un problema significativo in diverse forme di danno cerebrale ischemico tra cui ictus, trauma cranico e arresto cardiaco. Questo aumento può causare un ulteriore danno neurologico, in forma di ernia transtentoriale 1,2,3,4, compressione mesencefalo, deficit neurologici o infarto cerebrale è aumentato 2,4. Le attuali terapie sono spesso inadeguati per controllare elevata ICP in ambito clinico 5,6,7. Quindi vi è la necessità di metodi accurati di misura ICP in modelli animali per migliorare la nostra comprensione dei meccanismi di base e di sviluppare nuovi trattamenti per elevata ICP. In entrambi i casi l&#39;impostazione clinica e sperimentale ICP non può essere definita senza misurazione diretta. Diversi metodi di inserimento del catetere ICP esistono attualmente. Di questi il catetere intraventricolare è diventata la clinica &#39;gold standard&#39; di misura ICP negli esseri umani 8. Questo metodo involves l&#39;asportazione parziale del cranio e la strumentazione del catetere attraverso il tessuto cerebrale. Di conseguenza, cateteri intraventricolari hanno un tasso di infezione del 6-11% 9. Per questo motivo, cannulazioni subdurali ed epidurale sono diventati i metodi preferiti in modelli animali di danno ischemico. Diverse tecniche di misurazione ICP sono stati adattati per i modelli animali, e di questi, piene di liquido cateteri di telemetria 10 e cateteri a stato solido sono le più frequentemente utilizzate 11,12,13,14,15. Le piene di liquido sistemi sono inclini a sviluppare bolle d&#39;aria nella linea, con conseguente falsi letture ICP. Sonde a stato solido evitare questo problema (Figura 1). Un ulteriore problema è cateteri di montaggio sotto il cranio o nei ventricoli senza causare alcun danno cerebrale che potrebbero alterare i risultati sperimentali. Pertanto, abbiamo sviluppato un metodo che inserisce un catetere ICP contiguo con lo spazio epidurale, ma evita il NEed inserirlo tra cranio e cervello. Una fibra ottica catetere di pressione (420LP, sensori SAMBA, Svezia) è stato usato per misurare ICP nella posizione epidurale perché la posizione del sensore di pressione (sulla punta del catetere) è stato trovato per produrre un segnale ad alta fedeltà ICP in questo modello . Ci sono altri produttori di simili tecnologie a fibre ottiche 13 che possono essere utilizzati con la metodologia. Alternative cateteri stato solido, che hanno il sensore di pressione collocato sul lato della punta del catetere, non sarebbe appropriato per questo modello come il segnale sarebbe smorzata dalla presenza della vite di monitoraggio. Qui vi presentiamo un metodo relativamente semplice ed accurato per misurare la ICP. Questo metodo può essere utilizzato in una vasta gamma di modelli animali ICP correlati.

Pressure Controlled Ventilation to Induce Acute Lung Injury in Mice

Murine models are extensively used to investigate acute injuries of different organs systems (1-34). Acute lung injury (ALI), which occurs with prolonged mechanical ventilation, contributes to morbidity and mortality of critical illness, and studies on novel genetic or pharmacological targets are areas of intense investigation (1-3, 5, 8, 26, 30, 33-36). ALI is defined by the acute onset of the disease, which leads to non-cardiac pulmonary edema and subsequent impairment of pulmonary gas exchange (36). We have developed a murine model of ALI by using a pressure-controlled ventilation to induce ventilator-induced lung injury (2). For this purpose, C57BL/6 mice are anesthetized and a tracheotomy is performed followed by induction of ALI via mechanical ventilation. Mice are ventilated in a pressure-controlled setting with an inspiratory peak pressure of 45 mbar over 1 - 3 hours. As outcome parameters, pulmonary edema (wet-to-dry ratio), bronchoalveolar fluid albumin content, bronchoalveolar fluid and pulmonary tissue myeloperoxidase content and pulmonary gas exchange are assessed (2). Using this technique we could show that it sufficiently induces acute lung inflammation and can distinguish between different treatment groups or genotypes (1-3, 5). Therefore this technique may be helpful for researchers who pursue molecular mechanisms involved in ALI using a genetic approach in mice with gene-targeted deletion.

Automated Midline Shift and Intracranial Pressure Estimation based on Brain CT Images

In this paper we present an automated system based mainly on the computed tomography (CT) images consisting of two main components: the midline shift estimation and intracranial pressure (ICP) pre-screening system. To estimate the midline shift, first an estimation of the ideal midline is performed based on the symmetry of the skull and anatomical features in the brain CT scan. Then, segmentation of the ventricles from the CT scan is performed and used as a guide for the identification of the actual midline through shape matching. These processes mimic the measuring process by physicians and have shown promising results in the evaluation. In the second component, more features are extracted related to ICP, such as the texture information, blood amount from CT scans and other recorded features, such as age, injury severity score to estimate the ICP are also incorporated. Machine learning techniques including feature selection and classification, such as Support Vector Machines (SVMs), are employed to build the prediction model using RapidMiner. The evaluation of the prediction shows potential usefulness of the model. The estimated ideal midline shift and predicted ICP levels may be used as a fast pre-screening step for physicians to make decisions, so as to recommend for or against invasive ICP monitoring.

Ascending Aortic Constriction in Rats for Creation of Pressure Overload Cardiac Hypertrophy Model

Ascending aortic constriction is the most common and successful surgical model for creating pressure overload induced cardiac hypertrophy and heart failure. Here, we describe a detailed surgical procedure for creating pressure overload and cardiac hypertrophy in rats by constriction of the ascending aorta using a small metallic clip. After anesthesia, the trachea is intubated by inserting a cannula through a half way incision made between two cartilage rings of trachea. Then a skin incision is made at the level of the second intercostal space on the left chest wall and muscle layers are cleared to locate the ascending portion of aorta. The ascending aorta is constricted to 50&#8211;60% of its original diameter by application of a small sized titanium clip. Following aortic constriction, the second and third ribs are approximated with prolene sutures. The tracheal cannula is removed once spontaneous breathing was re-established. The animal is allowed to recover on the heating pad by gradually lowering anesthesia. The intensity of pressure overload created by constriction of the ascending aorta is determined by recording the pressure gradient using trans-thoracic two dimensional Doppler-echocardiography. Overall this protocol is useful to study the remodeling events and contractile properties of the heart during the gradual onset and progression from compensated cardiac hypertrophy to heart failure stage.

We describe a stepwise procedure for creating pressure overload and left ventricular hypertrophy in Wistar rats by constriction of the ascending aorta using a small metallic clip. This model is extensively used for studying remodeling changes during cardiac hypertrophy and for identifying and evaluating strategies for regression of such changes.

Assessing Myogenic Response and Vasoactivity In Resistance Mesenteric Arteries Using Pressure Myography

Small resistance arteries constrict and dilate respectively in response to increased or decreased intraluminal pressure; this phenomenon known as myogenic response is a key regulator of local blood flow. In isobaric conditions small resistance arteries develop sustained constriction known as myogenic tone (MT), which is a major determinant of systemic vascular resistance (SVR). Hence, ex vivo&#160;pressurized preparations of small resistance arteries are major tools to study microvascular function in near-physiological states. To achieve this, a freshly isolated intact segment of a small resistance artery (diameter ~260 &#956;m) is mounted onto two small glass cannulas and pressurized. These arterial preparations retain most in vivo characteristics and permit assessment of vascular tone in real-time. Here we provide a detailed protocol for assessing vasoactivity in pressurized small resistance mesenteric arteries from rats; these arteries develop sustained vasoconstriction - approximately 25% of maximal diameter - when pressurized at 70 mmHg. These arterial preparations may be used to study the effect of investigational compounds on relationship between intra-arterial pressure and vasoactivity and determine changes in microvascular function in animal models of various diseases.

Measurement of the Pressure-volume Curve in Mouse Lungs

In recent decades the mouse has become the primary animal model of a variety of lung diseases. In models of emphysema or fibrosis, the essential phenotypic changes are best assessed by measurement of the changes in lung elasticity. To best understand specific mechanisms underlying such pathologies in mice, it is essential to make functional measurements that can reflect the developing pathology. Although there are many ways to measure elasticity, the classical method is that of the total lung pressure-volume (PV) curve done over the whole range of lung volumes. This measurement has been made on adult lungs from nearly all mammalian species dating back almost 100 years, and such PV curves also played a major role in the discovery and understanding of the function of pulmonary surfactant in fetal lung development. Unfortunately, such total PV curves have not been widely reported in the mouse, despite the fact that they can provide useful information on the macroscopic effects of structural changes in the lung. Although partial PV curves measuring just the changes in lung volume are sometimes reported, without a measure of absolute volume, the nonlinear nature of the total PV curve makes these partial ones very difficult to interpret. In the present study, we describe a standardized way to measure the total PV curve. We have then tested the ability of these curves to detect changes in mouse lung structure in two common lung pathologies, emphysema and fibrosis. Results showed significant changes in several variables consistent with expected structural changes with these pathologies. This measurement of the lung PV curve in mice thus provides a straightforward means to monitor the progression of the pathophysiologic changes over time and the potential effect of therapeutic procedures.

Here we present a protocol to simply and reliably measure the lung pressure-volume curve in mice, showing that it is sufficiently sensitive to detect phenotypic parenchymal changes in two common lung pathologies, pulmonary fibrosis and emphysema. This metric provides a means to quantify the lung&#8217;s structural changes with developing pathology.

Intrauterine Telemetry to Measure Mouse Contractile Pressure In Vivo

A complex integration of molecular and electrical signals is needed to transform a quiescent uterus into a contractile organ at the end of pregnancy. Despite the discovery of key regulators of uterine contractility, this process is still not fully understood. Transgenic mice provide an ideal model in which to study parturition. Previously, the only method to study uterine contractility in the mouse was ex vivo isometric tension recordings, which are suboptimal for several reasons. The uterus must be removed from its physiological environment, a limited time course of investigation is possible, and the mice must be sacrificed. The recent development of radiometric telemetry has allowed for longitudinal, real-time measurements of in vivo intrauterine pressure in mice. Here, the implantation of an intrauterine telemeter to measure pressure changes in the mouse uterus from mid-pregnancy until delivery is described. By comparing differences in pressures between wild type and transgenic mice, the physiological impact of a gene of interest can be elucidated. This technique should expedite the development of therapeutics used to treat myometrial disorders during pregnancy, including preterm labor.

Trabecular Meshwork Response to Pressure Elevation in the Living Human Eye

The mechanical characteristics of the trabecular meshwork (TM) are linked to outflow resistance and intraocular pressure (IOP) regulation. The rationale behind this technique is the direct observation of the mechanical response of the TM to acute IOP elevation. Prior to scanning, IOP is measured at baseline and during IOP elevation. The limbus is scanned by spectral-domain optical coherence tomography at baseline and during IOP elevation (ophthalmodynamometer (ODM) applied at 30 g force). Scans are processed to enhance visualization of the aqueous humor outflow pathway using ImageJ. Vascular landmarks are used to identify corresponding locations in baseline and IOP elevation scan volumes. Schlemm canal (SC) cross-sectional area (SC-CSA) and SC length from anterior to posterior along its long axis are measured manually at 10 locations within a 1 mm segment of SC. Mean inner to outer wall distance (short axis length) is calculated as the area of SC divided by its long axis length. To examine the contribution of adjacent tissues to the effect IOP elevations, measurements are repeated without and with smooth muscle relaxation with instillation of tropicamide. TM migration into SC is resisted by TM stiffness, but is enhanced by the support of its attachment to adjacent smooth muscle within the ciliary body. This technique is the first to measure the living human TM response to pressure elevation in situ under physiological conditions within the human eye.

Long-term Blood Pressure Measurement in Freely Moving Mice Using Telemetry

During the development of new vasoactive agents, arterial blood pressure monitoring is crucial for evaluating the efficacy of the new proposed drugs. Indeed, research focusing on the discovery of new potential therapeutic targets using genetically altered mice requires a reliable, long-term assessment of the systemic arterial pressure variation. Currently, the gold standard for obtaining long-term measurements of blood pressure in ambulatory mice uses implantable radio-transmitters, which require&#160;artery cannulation. This technique eliminates the need for tethering, restraining, or anesthetizing the animals which introduce stress and artifacts during data sampling. However, arterial blood pressure monitoring in mice via catheterization can be rather challenging due to the small size of the arteries. Here we present a step-by-step guide to illustrate the crucial key passages for a successful subcutaneous implantation of radio-transmitters and carotid artery cannulation in mice. We also include examples of long-term blood pressure activity taken from freely moving mice after a period of post-surgery recovery. Following this procedure will allow reliable direct blood pressure recordings from multiple animals simultaneously.

Spatial Measurements of Perfusion, Interstitial Fluid Pressure and Liposomes Accumulation in Solid Tumors

The heterogeneous intra-tumoral accumulation of liposomes is a critical determinant of their efficacy. Both the chaotic tumor microcirculation and elevated IFP are linked to the heterogeneous intra-tumoral distribution of nanotechnology-based drug delivery systems such as liposomes. In the present study, the relationship between tumor microcirculation, elevated IFP, and accumulation of nanoparticles was investigated through in vivo experimentation. This was accomplished by evaluation of the tumor microcirculation using dynamic contrast enhanced computed tomography (DCE-CT) and measurement of tumor IFP using a novel image-guided robotic needle placement system connected to the micro-CT scanner. The intra-tumoral accumulation of liposomes was determined by CT image-based assessment of a nanoparticle liposomal formulation that stably encapsulate the contrast agent iohexol (CT-liposomes). CT imaging allowed for co-localization of the spatial distribution of tumor hemodynamics, IFP and CT-liposome accumulation in an individual subcutaneous xenograft mouse model of breast cancer. Measurements led to the discovery that perfusion and plasma volume fraction are strong mediators of the intra-tumoral distribution of liposomes. Furthermore, the results suggest that IFP plays an indirect role in mediating liposome distribution through modulating blood flow.

Cavernous Nerve Stimulation and Recording of Intracavernous Pressure in a Rat

The stimulation of the cavernous nerve (CN) and measurement of intracavernous pressure (ICP) have been used extensively to test and evaluate therapies for erectile dysfunction. However, the methods used vary between laboratories, and pitfalls still exist. The goal of this study was to describe a surgical technique that would provide a reliable and reproducible model. By exposing the ischiocavernosus muscle at its point of insertion on the ischial tuberosity, the penile crus could be cannulated with minimal dissection and injury to the structures involved in erectile function. Repeated stimulation of the CN, without the need for lifting and drying, was achieved by using a 125 &#181;m bipolar silver electrode and biocompatible silicon glue to isolate the electrode-nerve complex. This method prevents neuropraxia by reducing stretching and drying the nerve and provides complete isolation of the nerve, negating electrical leakage and preventing stimulation of alternative pathways.
