La alt&iacute;sima tasa de karyotypic evoluci&oacute;n que caracteriza a las musara&ntilde;as del grupo Sorex araneus hace de este grupo un modelo excepcionalmente interesante para gen&eacute;tica de poblaciones y estudios evolutivos. Aqu&iacute;, hemos intentado mapa 46 marcadores microsat&eacute;lites en el nivel de brazo de cromosoma mediante los cromosomas ordenados por el flujo de tres karyotypically diferentes taxones del grupo Sorex araneus (S. granarius y las razas de cromosoma Cordon y Novosibirsk de S. araneus). Las localizaciones m&aacute;s probables fueron proporcionados para 35 marcadores, entre los cuales 25 fueron cada uno inequ&iacute;vocamente asignado a un locus en el cromosoma correspondiente en los tres taxa, cubriendo los tres cromosomas sexuales (XY1Y2) y nueve de los brazos de un autosoma 18 del Grupo S. araneus. Los resultados proporcionan m&aacute;s evidencia de un alto grado de conservaci&oacute;n en la organizaci&oacute;n del genoma en el grupo de araneus S. pese a la presencia de numerosos cambios de Robertsonian. The extremely high rate of karyotypic evolution that characterizes the shrews of the Sorex araneus group makes this group an exceptionally interesting model for population genetics and evolutionary studies. Here, we attempted to map 46 microsatellite markers at the chromosome arm level using flow-sorted chromosomes from three karyotypically different taxa of the Sorex araneus group (S. granarius and the chromosome races Cordon and Novosibirsk of S. araneus). The most likely localizations were provided for 35 markers, among which 25 were each unambiguously mapped to a single locus on the corresponding chromosomes in the three taxa, covering the three sexual chromosomes (XY1Y2) and nine of the 18 autosomal arms of the S. araneus group. The results provide further evidence for a high degree of conservation in genome organization in the S. araneus group despite the presence of numerous Robertsonian rearrangements. These markers can therefore be used to compare the genetic structure among taxa of the S. araneus group at the chromosome level and to study the role of chromosomal rearrangements in the genetic diversification and speciation process of this group.

Die extrem hohe Rate der karyotypic Evolution, die die Spitzm&auml;use der Sorex Araneus Gruppe charakterisiert dadurch ein besonders interessantes Modell f&uuml;r Populationsgenetik und evolution&auml;ren Studien zu gruppieren. Hier haben wir versucht, 46 Microsatellite Markierungen auf Chromosom Arm Ebene mithilfe von Fluss-sortiert Chromosomen von drei karyotypisch verschiedene Taxa der Sorex Araneus Gruppe (S. Granarius und die Chromosom-Rennen, Cordon und Nowosibirsk in S. Araneus) zuzuordnen. Die wahrscheinlichsten Lokalisierungen waren vorausgesetzt f&uuml;r 35 Marker, unter denen 25 waren jeweils eindeutig einem einzigen Locus auf den entsprechenden Chromosomen in den drei Taxa, die drei sexuellen Chromosomen (XY1Y2) und neun der 18 autosomal Arme der S. Araneus Gruppe zugeordnet. Die Ergebnisse liefern weitere Beweise f&uuml;r ein hohes Ma&szlig; an Erhaltung in Genom-Organisation in der S. Araneus Gruppe trotz der Anwesenheit der zahlreichen Robertsonian Umlagerungen. Diese Marker k&ouml;nnen daher die genetische Struktur zwischen den Taxa der S. Araneus Gruppe auf der Ebene der Chromosomen zu vergleichen und die Rolle der chromosomalen Rearrangements in die genetische Diversifikation und Artbildung Prozess dieser Gruppe lernen verwendet werden.

Il tasso estremamente alto di karyotypic evoluzione che caratterizza i toporagni del gruppo Sorex araneus rende questo gruppo un modello particolarmente interessante per la genetica di popolazione e di studi evoluzionistici. Qui, abbiamo tentato di mappa 46 marcatori microsatellite a livello di braccio cromosoma utilizzando cromosomi ordinati di flusso da tre taxa differenti aberrazioni del gruppo Sorex araneus (S. granarius e le gare di cromosoma Cordon e Novosibirsk di S. araneus). Le localizzazioni pi&ugrave; probabile sono stati forniti per 35 marcatori, tra i quali 25 ciascuno inequivocabilmente mappato a un singolo locus sui cromosomi corrispondenti in tre taxa, che coprono i tre cromosomi sessuali (XY1Y2) e nove di 18 braccia autosomal del gruppo S. araneus. I risultati forniscono ulteriori prove per un alto grado di conservazione nell&#39;organizzazione del genoma nel gruppo S. araneus nonostante la presenza di numerosi Robertsoniana riarrangiamenti. The great karyotypic differences between camel, cattle and pig, three important domestic animals, have been a challenge for comparative cytogenetic studies based on conventional cytogenetic approaches. To construct a genome-wide comparative chromosome map among these artiodactyls, we made a set of chromosome painting probes from the dromedary camel (Camelus dromedarius) by flow sorting and degenerate oligonucleotide primed-PCR. The painting probes were first used to characterize the karyotypes of the dromedary camel (C. dromedarius), the Bactrian camel (C. bactrianus), the guanaco (Lama guanicoe), the alpaca (L. pacos) and dromedary x guanaco hybrid karyotypes (all with 2n = 74). These FISH experiments enabled the establishment of a high-resolution GTG-banded karyotype, together with chromosome nomenclature and idiogram for C. dromedarius, and revealed that these camelid species have almost identical karyotypes, with only slight variations in the amount and distribution patterns of heterochromatin. Further cross-species chromosome painting between camel, cattle, pig and human with painting probes from the camel and human led to the establishment of genome-wide comparative maps. Between human and camel, pig and camel, and cattle and camel 47, 53 and 53 autosomal conserved segments were detected, respectively. Integrated analysis with previously published comparative maps of human/pig/cattle enabled us to propose a Cetartiodactyla ancestral karyotype and to discuss the early karyotype evolution of Cetartiodactyla. Furthermore, these maps will facilitate the positional cloning of genes by aiding the cross-species transfer of mapping information.

Les grandes diff&eacute;rences caryotypiques entre des chameaux, des bovins et des porcs, trois importantes des animaux domestiques, ont &eacute;t&eacute; un d&eacute;fi pour des &eacute;tudes cytog&eacute;n&eacute;tiques comparatives bas&eacute;e sur des approches de cytog&eacute;n&eacute;tiques conventionnelles. Pour construire une carte chromosomique comparative genome-wide parmi ces artiodactyles, nous avons fait un ensemble de sondes de peinture chromosomique de chameau dromadaire (Camelus dromedarius) par flux tri et oligonucl&eacute;otides d&eacute;g&eacute;n&eacute;r&eacute;s appr&ecirc;t&eacute;-PCR. Les sondes de peinture ont &eacute;t&eacute; utilis&eacute;es pour caract&eacute;riser les caryotypes de chameau dromadaire (dromedarius c.), le chameau de Bactriane (c. bactrianus), le guanaco (Lama guanicoe), alpaga (L. pacos) et dromadaire x caryotypes hybride guanaco (toutes avec 2n = 74). Die gro&szlig;e karyotypic Unterschiede zwischen Kamel, Rind und Schwein, drei wichtige Domestizierte Tiere, wurden eine Herausforderung f&uuml;r vergleichende zytogenetische Untersuchungen &uuml;ber konventionelle zytogenetische Ans&auml;tze. Konstruieren eine genomweiten vergleichende Chromosom-Karte unter diese Paarhufer haben wir eine Reihe von Chromosom Malerei Sonden aus dem Dromedar (Camelus Dromedarius) durch Str&ouml;mung zu sortieren und degenerierte Oligonukleotid-grundiert-PCR. Integrierte Analyse mit bereits ver&ouml;ffentlichten vergleichenden Maps Mensch/Schweine/Rinder konnten wir eine Cetartiodactyla ancestral Karyotyp vorzuschlagen und die fr&uuml;he Evolution der Karyotyp der Cetartiodactyla zu diskutieren. Le grandi differenze karyotypic tra cammelli, bovini e suini, tre importanti animali domestici, sono state una sfida per gli studi di citogenetici comparativi basata su approcci citogenetici convenzionali. Per costruire una mappa cromosomica comparative genome-wide tra questi artiodattili, abbiamo fatto un set di sonde painting cromosoma da cammello dromedario (Camelus dromedarius) dal flusso di ordinamento e degenere del oligonucleotide primer-PCR. Le sonde di pittura sono state utilizzate per caratterizzare il cariotipo di cammello dromedario (c. dromedarius), il cammello battriano (c. bactrianus), il guanaco (Lama guanicoe), l&#39;alpaca (L. pacos) e dromedario x cariotipo ibrido guanaco (tutte con 2n = 74). Questi esperimenti di pesce abilitato la creazione di un cariotipo ad alta risoluzione GTG banded, insieme al cromosoma nomenclatura e idiogram per c. dromedarius e ha rivelato che queste specie di camelide hanno cariotipi quasi identici, con solo lievi variazioni nei modelli di distribuzione e quantit&agrave; di eterocromatina. Las grandes diferencias karyotypic entre camellos, ganado y cerdo, tres importantes los animales dom&eacute;sticos, han sido un reto para los estudios citogen&eacute;ticos comparativos basados en enfoques citogen&eacute;ticos convencionales. Para construir un mapa del genoma comparativo cromosoma entre estos artiod&aacute;ctilos, hicimos un conjunto de sondas de pintura de cromosoma del camello dromedario (Camelus dromedarius) por flujo clasificaci&oacute;n y oligonucle&oacute;tidos degenerados preparado-PCR. Las sondas de pintura primero fueron utilizadas para caracterizar los cariotipos de camello dromedario (C. dromedarius), el Camello Bactriano (C. bactrianus), el guanaco (Lama guanicoe), la alpaca (L. pacos) y dromedario x cariotipos h&iacute;brido de guanaco (todas con 2n = 74). Estos experimentos de pescado permiti&oacute; el establecimiento de un cariotipo con bandas GTG alta resoluci&oacute;n, junto con la nomenclatura de cromosoma y idiogram para C. dromedarius y revelaron que estas especies de cam&eacute;lidos tienen cariotipos casi id&eacute;nticos, con s&oacute;lo ligeras variaciones en los patrones de la cantidad y distribuci&oacute;n de la heterocromatina. Otra especie cromosoma pintura entre camellos, ganado, cerdos y humano con sondas del camello y humanos conducido al establecimiento de mapas comparativos del genoma de la pintura. Entre humanos y camello, cerdo y camello y ganado y camello 47, se detectaron segmentos conservados de un autosoma 53 y 53, respectivamente. An&aacute;lisis integrado con mapas comparativos previamente publicados de humanos/cerdo/ganado permitido proponer un cariotipo ancestral Cetartiodactyla y discutir la evoluci&oacute;n temprana del cariotipo de Cetartiodactyla. Adem&aacute;s, estos mapas facilitar&aacute; la clonaci&oacute;n posicional de genes por ayudar a la transferencia de la especie de mapeo de informaci&oacute;n.

The mammalian ortholog of yeast Slx4, BTBD12, is an ATM substrate that functions as a scaffold for various DNA repair activities. Mutations of human BTBD12 have been reported in a new sub-type of Fanconi anemia patients. Recent studies have implicated the fly and worm orthologs, MUS312 and HIM-18, in the regulation of meiotic crossovers arising from double-strand break (DSB) initiating events and also in genome stability prior to meiosis. Using a Btbd12 mutant mouse, we analyzed the role of BTBD12 in mammalian gametogenesis. BTBD12 localizes to pre-meiotic spermatogonia and to meiotic spermatocytes in wildtype males. Btbd12 mutant mice have less than 15% normal spermatozoa and are subfertile. Loss of BTBD12 during embryogenesis results in impaired primordial germ cell proliferation and increased apoptosis, which reduces the spermatogonial pool in the early postnatal testis. During prophase I, DSBs initiate normally in Btbd12 mutant animals. However, DSB repair is delayed or impeded, resulting in persistent Î³H2AX and RAD51, and the choice of repair pathway may be altered, resulting in elevated MLH1/MLH3 focus numbers at pachynema. The result is an increase in apoptosis through prophase I and beyond. Unlike yeast Slx4, therefore, BTBD12 appears to function in meiotic prophase I, possibly during the recombination events that lead to the production of crossovers. In line with its expected regulation by ATM kinase, BTBD12 protein is reduced in the testis of Atm(-/-) males, and Btbd12 mutant mice exhibit increased genomic instability in the form of elevated blood cell micronucleus formation similar to that seen in Atm(-/-) males. Taken together, these data indicate that BTBD12 functions throughout gametogenesis to maintain genome stability, possibly by co-ordinating repair processes and/or by linking DNA repair events to the cell cycle via ATM.

Mammalian BTBD12 (SLX4) protects against genomic instability during mammalian spermatogenesis.

Los mam&iacute;fero ort&oacute;logo de levadura Slx4, BTBD12, es un sustrato de ATM que funciona como un andamio para diversas actividades de reparaci&oacute;n de ADN. Mutaciones de la BTBD12 humana se han divulgado en un nuevo subtipo de pacientes de la anemia de Fanconi. Estudios recientes han implicado la mosca y gusano ort&oacute;logos, MUS312 y &eacute;l-18, en la regulaci&oacute;n de cruces mei&oacute;ticas derivados de la rotura de la doble cadena (de diferencias OSD) iniciar eventos y tambi&eacute;n en la estabilidad del genoma antes de la meiosis. Utiliza un rat&oacute;n mutante Btbd12, hemos analizado el papel de BTBD12 en mam&iacute;fero gametog&eacute;nesis. BTBD12 localiza pre-meiotic espermatogonias y espermatocitos mei&oacute;ticas en varones de tipo salvaje. Btbd12 ratones mutantes tienen menos de 15% de espermatozoides normales y son subf&eacute;rtiles. P&eacute;rdida de BTBD12 durante la embriog&eacute;nesis resulta en la proliferaci&oacute;n de la c&eacute;lula de germen primordial deteriorada y apoptosis creciente, lo que reduce la piscina espermatogonias en el test&iacute;culo postnatal temprana.
