&#x5A01;&#x5EC9;&#x59C6;&#x65AF; Beuren &#x7EFC;&#x5408;&#x5F81;&#x76F8;&#x5173;&#x87BA;&#x65CB;-&#x73AF;-&#x87BA;&#x65CB;&#x542B;&#x6709;&#x57DF;&#x7684;&#x8F6C;&#x5F55;&#x56E0;&#x5B50;&#x6FC0;&#x6D3B;&#x7D20;/&#x8282;&#x70B9;&#x4FE1;&#x53F7;&#x3002;

We investigated the regulation of the activin/nodal-inducible distal element (DE) of the Xenopus goosecoid (gsc) promoter. On the basis of its interaction with the DE, we isolated a Xenopus homolog of the human Williams-Beuren syndrome critical region 11 (XWBSCR11), and further, show that it interacts with pathway-specific Smad2 and Smad3 in a ligand-dependent manner. Interestingly, we also find that XWBSCR11 functions cooperatively with FoxH1 (Fast-1) to stimulate DE-dependent transcription. We propose a mechanism in which FoxH1 functions together with Smads as a cofactor for the recruitment of transcription factors like XWBSCR11 in the process of activin/nodal-mediated gsc-specific induction. This mechanism provides considerable opportunities for modulation of transcription across a variety of activin/nodal-inducible genes, increasing diversity in promoter selection, thus leading to the differential induction of activin/nodal target genes.

The role of a Williams-Beuren syndrome-associated helix-loop-helix domain-containing transcription factor in activin/nodal signaling.

Nous avons &#xE9;tudi&#xE9; la r&#xE9;gulation de l&#x26;#39;&#xE9;l&#xE9;ment activine / nodale inductible distale (DE) de la goosecoid Xenopus (CGC) promoteur. Sur la base de son interaction avec le DE, nous avons isol&#xE9; un homologue de Xenopus de l&#x26;#39;&#xEA;tre humain le syndrome de Williams-Beuren r&#xE9;gion critique 11 (XWBSCR11), et en outre, d&#xE9;montrer qu&#x26;#39;il interagit avec la voie sp&#xE9;cifique Smad2 et Smad3 d&#x26;#39;une mani&#xE8;re ligand-d&#xE9;pendante. Fait int&#xE9;ressant, nous constatons &#xE9;galement que XWBSCR11 fonctions en collaboration avec FoxH1 (Fast-1) afin de stimuler la transcription d&#xE9;pendante DE. Abbiamo studiato il regolamento dell&#39;elemento activin/nodale-inducible distale (DE) del promotore Xenopus goosecoid (SGC). Sulla base delle sue interazioni con il DE, abbiamo isolato un omologo di Xenopus della regione critica di sindrome Williams-Beuren umano 11 (XWBSCR11) e ulteriormente, mostrano che interagisce con specifici pathway Smad2 e Smad3 in modo ligando-dipendente. Interessante, troviamo anche che XWBSCR11 funzioni in modo cooperativo con FoxH1 (Fast-1) per stimolare la trascrizione di DE-dependent. Proponiamo un meccanismo in cui FoxH1 funziona insieme a SMAD come cofattore per l&#39;assunzione di fattori di trascrizione come XWBSCR11 nel processo di induzione gsc-specifiche activin/nodale-mediata. Questo meccanismo fornisce una notevole opportunit&agrave; per la modulazione della trascrizione attraverso una variet&agrave; di geni activin/nodale-inducible, aumentando la diversit&agrave; nella selezione del promotore, determinando in tal modo l&#39;induzione differenziale dei geni bersaglio activin/nodale.

Hemos investigado la regulaci&#xF3;n del elemento de la activina / nodales inducibles distal (DE) de la goosecoid Xenopus (SGC) promotor. Sobre la base de su interacci&#xF3;n con la DE, hemos aislado un hom&#xF3;logo del Xenopus humano de Williams-Beuren regi&#xF3;n cr&#xED;tica del s&#xED;ndrome 11 (XWBSCR11) y, adem&#xE1;s, muestran que interact&#xFA;a con la v&#xED;a espec&#xED;fica de Smad2 y Smad3 en una forma dependiente de ligando. Curiosamente, tambi&#xE9;n encontramos que las funciones de cooperaci&#xF3;n con XWBSCR11 FoxH1 (Fast-1) para estimular la DE-dependen de la transcripci&#xF3;n. Se propone un mecanismo en el que FoxH1 funciones, junto con Smads como un cofactor para la contrataci&#xF3;n de los factores de transcripci&#xF3;n como el XWBSCR11 en el proceso de la activina / nodales mediada por la inducci&#xF3;n SGC espec&#xED;fico. Este mecanismo ofrece oportunidades considerables para la modulaci&#xF3;n de la transcripci&#xF3;n a trav&#xE9;s de una variedad de genes activina / nodales inducibles, el aumento de la diversidad en la selecci&#xF3;n del promotor, lo que conduce a la inducci&#xF3;n diferencial de los genes diana de activina / ganglionar.

The complex gene regulatory networks governed by growth factor signaling are still poorly understood. In order to accelerate the rate of progress in uncovering these networks, we explored the usefulness of interspecies sequence comparison (phylogenetic footprinting) to identify conserved growth factor response elements. The promoter regions of two direct target genes of Bone Morphogenetic Protein (BMP) signaling in Xenopus, Xvent2 and XId3, were compared with the corresponding human and/or mouse counterparts to identify conserved sequences. A comparison between the Xenopus and human Vent2 promoter sequences revealed a highly conserved 21 bp sequence that overlaps the previously reported Xvent2 BMP response element (BRE). Reporter gene assays using Xenopus animal pole ectodermal explants (animal caps) revealed that this conserved 21 bp BRE is both necessary and sufficient for BMP responsiveness. We combine the same phylogenetic footprinting approach with luciferase assays to identify a highly conserved 49 bp BMP responsive region in the Xenopus Id3 promoter. GFP reporters containing multimers of either the Xvent2 or XId3 BREs appear to recapitulate endogenous BMP signaling activity in transgenic Xenopus embryos. Comparison of the Xvent2 and the XId3 BRE revealed core sequence features that are both necessary and sufficient for BMP responsiveness: a Smad binding element (SBE) and a GC-rich element resembling an OAZ binding site. Based on these findings, we have implemented genome scanning to identify over 100 additional putative target genes containing 2 or more BRE-like sequences which are conserved between human and mouse. RT-PCR and in situ analyses revealed that this in silico approach can effectively be used to identify potential BMP target genes.

Phylogenetic footprinting and genome scanning identify vertebrate BMP response elements and new target genes.

Les r&#xE9;seaux de g&#xE8;nes r&#xE9;gulateurs complexes r&#xE9;gis par la signalisation du facteur de croissance sont encore mal compris. Afin d&#x26;#39;acc&#xE9;l&#xE9;rer le rythme des progr&#xE8;s dans la d&#xE9;couverte de ces r&#xE9;seaux, nous avons explor&#xE9; l&#x26;#39;utilit&#xE9; de la comparaison de s&#xE9;quences entre esp&#xE8;ces (empreinte phylog&#xE9;n&#xE9;tique) pour identifier les &#xE9;l&#xE9;ments conserv&#xE9;s du facteur de croissance de r&#xE9;ponse. Les r&#xE9;gions promotrices de deux g&#xE8;nes cibles directs la prot&#xE9;ine morphog&#xE9;n&#xE9;tique osseuse (BMP) de signalisation chez Xenopus, Xvent2 et XId3, ont &#xE9;t&#xE9; compar&#xE9;s avec l&#x26;#39;&#xEA;tre humain correspondant et / ou ses homologues de la souris afin d&#x26;#39;identifier des s&#xE9;quences conserv&#xE9;es. Sur la base de ces constatations, nous avons mis en place g&#xE9;nome balayage pour identifier plus de 100 autres g&#xE8;nes cibles putatifs contenant 2 ou plusieurs s&#xE9;quences de type BRE qui sont conserv&#xE9;es entre l&#x26;#39;homme et la souris. Le reti di gene complesso normativo governate da segnalazione del fattore di crescita sono ancora poco conosciute. Al fine di accelerare la velocit&agrave; di avanzamento nella scoperta di queste reti, abbiamo esplorato l&#39;utilit&agrave; del confronto di sequenza interspecie (filogenetica dell&#39;impronta) per identificare gli elementi di risposta conservata al fattore di crescita. Le regioni del promotore di due geni bersaglio diretto di Bone Morphogenetic Protein (BMP) di segnalazione in Xenopus, Xvent2 e XId3, sono stati confrontati con il corrispondente umano e/o controparti del mouse per identificare sequenze conservate. Un confronto tra l&#39;umane Vent2 promotore sequenze e Xenopus ha rivelato una sequenza di bp 21 altamente conservato che si sovrappone l&#39;elemento precedentemente riportato per la risposta Xvent2 BMP (BRE). Dosaggi di gene reporter utilizzando Xenopus Polo animale ectodermiche espianti (protezioni animali) ha rivelato che questo conservato 21 bp BRE &egrave; necessaria e sufficiente per la reattivit&agrave; BMP. Combiniamo lo stesso approccio filogenetico dell&#39;impronta con dosaggi di luciferase per identificare un altamente conservato 49 bp BMP reattivo nella regione del promotore di Xenopus Id3. Reporter GFP contenente multimeri di Xvent2 o XId3 BREs sembrano riassumere endogeno attivit&agrave; segnalazione BMP in embrioni di Xenopus transgenici. Confronto tra il Xvent2 e la XId3 BRE rivelato core sequenza caratteristiche che sono necessarie e sufficienti per la reattivit&agrave; BMP: un elemento di associazione di Smad (SBE) e un elemento GC-ricco, simile a un sito di legame OAZ. Sulla base di questi risultati, abbiamo implementato genoma scansione per identificare oltre 100 geni bersaglio putativo aggiuntive contenenti sequenze di 2 o pi&ugrave; BRE-like che sono conservate tra umano e del mouse. Bas&#xE1;ndose en estos hallazgos, se ha implementado de barrido del genoma para identificar m&#xE1;s de 100 genes adicionales objetivo putativo que contienen 2 o m&#xE1;s BRE-igual que las secuencias que se conservan entre los humanos y de rat&#xF3;n. Bone Morphogenetic Proteins (Bmps) are secreted growth factors that play crucial roles in animal development across the phylogenetic spectrum. Bmp signaling results in the phosphorylation and nuclear translocation of Smads, downstream signal transducers that bind DNA. In Drosophila, the zinc finger protein Schnurri (Shn) plays a key role in signaling by the Bmp2/Bmp4 homolog Decapentaplegic (Dpp), by forming a Shn/Smad repression complex on defined promoter elements in the brinker (brk) gene. Brk is a transcriptional repressor that downregulates Dpp target genes. Thus, brk inhibition by Shn results in the upregulation of Dpp-responsive genes. We present evidence that vertebrate Shn homologs can also mediate Bmp responsiveness through a mechanism similar to Drosophila Shn. We find that a Bmp response element (BRE) from the Xenopus Vent2 promoter drives Dpp-dependent expression in Drosophila. However, in sharp contrast to its activating role in vertebrates, the frog BRE mediates repression in Drosophila. Remarkably, despite these opposite transcriptional polarities, sequence changes that abolish cis-element activity in Drosophila also affect BRE function in Xenopus. These similar cis requirements reflect conservation of trans-acting factors, as human Shn1 (hShn1; HIVEP1) can interact with Smad1/Smad4 and assemble an hShn1/Smad complex on the BRE. Furthermore, both Shn and hShn1 activate the BRE in Xenopus embryos, and both repress brk and rescue embryonic patterning defects in shn mutants. Our results suggest that vertebrate Shn proteins function in Bmp signal transduction, and that Shn proteins recruit coactivators and co-repressors in a context-dependent manner, rather than acting as dedicated activators or repressors.

Drosophila spielt das Zink-Finger-Protein Schnurri (Shn) eine Schl&uuml;sselrolle bei der Signalisierung durch die Bmp2/Bmp4 Homolog Decapentaplegic (Dpp), durch die Bildung einer komplexen Shn/Smad-Unterdr&uuml;ckung auf definierten Promotor Elemente im Brinker (Brk)-Gen. BRK ist ein transkriptionelle Verdr&auml;nger, dass Downregulates Dpp Gene Ziel. Brk-Hemmung durch Shn f&uuml;hrt somit die Hochregulation der Dpp-responsive Gene. Proteine morfogenetiche ossee (Bmps) sono secrete fattori di crescita che giocano un ruolo cruciale nello sviluppo degli animali attraverso lo spettro filogenetico. Segnalazione BMP provoca la fosforilazione e la traslocazione nucleare di SMAD, trasduttori di segnale a valle che legano il DNA. The molecular mechanisms governing the cell behaviors underlying morphogenesis remain a major focus of research in both developmental biology and cancer biology. TGF-beta ligands control cell fate specification via Smad-mediated signaling. However, their ability to guide cellular morphogenesis in a variety of biological contexts is poorly understood. We report on the discovery of a novel TGF-beta signaling-mediated cellular morphogenesis occurring during vertebrate gastrulation. Activin/nodal members of the TGF-beta superfamily induce the expression of two genes regulating cell adhesion during gastrulation: Fibronectin Leucine-rich Repeat Transmembrane 3 (FLRT3), a type I transmembrane protein containing extracellular leucine-rich repeats, and the small GTPase Rnd1. FLRT3 and Rnd1 interact physically and modulate cell adhesion during embryogenesis by controlling cell surface levels of cadherin through a dynamin-dependent endocytosis pathway. Our model suggests that cell adhesion can be dynamically regulated by sequestering cadherin through internalization, and subsequent redeploying internalized cadherin to the cell surface as needed. As numerous studies have linked aberrant expression of small GTPases, adhesion molecules such as cadherins, and TGF-beta signaling to oncogenesis and metastasis, it is tempting to speculate that this FLRT3/Rnd1/cadherin pathway might also control cell behavior and morphogenesis in adult tissue homeostasis.

The FLRT family of transmembrane proteins has been implicated in the regulation of FGF signalling, neurite outgrowth, homotypic cell sorting and cadherin-mediated adhesion. In an expression screen we identified the Netrin receptors Unc5B and Unc5D as high-affinity FLRT3 interactors. Upon overexpression, Unc5B phenocopies FLRT3 and both proteins synergize in inducing cell deadhesion in Xenopus embryos. Morpholino knock-downs of Unc5B and FLRT3 synergistically affect Xenopus development and induce morphogenetic defects. The small GTPase Rnd1, which transmits FLRT3 deadhesion activity, physically and functionally interacts with Unc5B, and mediates its effect on cell adhesion. The results suggest that FLRT3, Unc5B and Rnd1 proteins interact to modulate cell adhesion in early Xenopus development.

Sur la surexpression, Unc5B ph&#xE9;nocopies FLRT3 et les deux prot&#xE9;ines en synergie pour induire deadhesion cellules dans des embryons de x&#xE9;nope. Die Familie FLRT Transmembranproteine hat bei der Regulierung der FGF Signalisierung, Neurit Auswuchs, homotypischen Zellsortierung und Cadherin-vermittelte Adh&auml;sion verwickelt. In einem Ausdruck-Bildschirm haben wir die Netrin-Rezeptoren, Unc5B und Unc5D als hoher Affinit&auml;t FLRT3 Interaktoren identifiziert. We used somatic cell nuclear transfer (SCNT) to generate a mouse from the nucleus of an IgG1(+) ovalbumin-specific B cell. The resulting OBI mice show generally normal B-cell development, with elevated percentages of marginal zone B cells and a reduction in B-1 B cells. Whereas OBI RAG1(-/-) mice have exclusively IgG1 anti-ovalbumin in their serum, OBI mice show elevated levels of anti-ovalbumin of nearly all isotypes 3' of the Î³1 constant region in the IgH locus, indicating that class switch recombination (CSR) occurs in the absence of immunization with ovalbumin. This CSR is associated with the presence of IgM(+)IgG1(+) double producer B cells that represent <1% of total B cells, accumulate in the peritoneal cavity, and account for near-normal levels of serum IgM and IgG3.

We have generated, via somatic cell nuclear transfer, two independent lines of transnuclear (TN) mice, using as nuclear donors CD8 T cells, sorted by tetramer staining, that recognize the endogenous melanoma antigen TRP1. These two lines of nominally identical specificity differ greatly in their affinity for antigen (TRP1(high) or TRP1(low)) as inferred from tetramer dissociation and peptide responsiveness. Ex vivo-activated CD8 T cells from either TRP1(high) or TRP1(low) mice show cytolytic activity in 3D tissue culture and in vivo, and slow the progression of subcutaneous B16 melanoma. Although naïve TRP1(low) CD8 T cells do not affect tumor growth, upon activation these cells function indistinguishably from TRP1(high) cells in vivo, limiting tumor cell growth and increasing mouse survival. The anti-tumor effect of both TRP1(high) and TRP1(low) CD8 T cells is enhanced in RAG-deficient hosts. However, tumor outgrowth eventually occurs, likely due to T cell exhaustion. The TRP1 TN mice are an excellent model for examining the functional attributes of T cells conferred by TCR affinity, and they may serve as a platform for screening immunomodulatory cancer therapies.

Transnuclear TRP1-specific CD8 T cells with high or low affinity TCRs show equivalent antitumor activity.

We discovered a novel somatic gene fusion, CD74-NRG1, by transcriptome sequencing of 25 lung adenocarcinomas of never smokers. By screening 102 lung adenocarcinomas negative for known oncogenic alterations, we found four additional fusion-positive tumors, all of which were of the invasive mucinous subtype. Mechanistically, CD74-NRG1 leads to extracellular expression of the EGF-like domain of NRG1 III-β3, thereby providing the ligand for ERBB2-ERBB3 receptor complexes. Accordingly, ERBB2 and ERBB3 expression was high in the index case, and expression of phospho-ERBB3 was specifically found in tumors bearing the fusion (P < 0.0001). Ectopic expression of CD74-NRG1 in lung cancer cell lines expressing ERBB2 and ERBB3 activated ERBB3 and the PI3K-AKT pathway, and led to increased colony formation in soft agar. Thus, CD74-NRG1 gene fusions are activating genomic alterations in invasive mucinous adenocarcinomas and may offer a therapeutic opportunity for a lung tumor subtype with, so far, no effective treatment.
