All procedures involving animal models have been reviewed by the local institutional animal care committee and the JoVE veterinary review board.
1. Electroencephalogram (EEG) Recordings
<ol>
	<li>Preparation of the EEG mini-cap
	<ol>
		<li>Immerse the electrode tips of the EEG mini-cap for at least 12 hrs in distilled water with 0.2% chloride. Rinse the EEG mini-cap gently in distilled water. Dry the cap and the electrodes in the air.</li>
		<li>Mix EEG electrode paste with 0.9% NaCl solution in the volume proportion of 2:1. Add a drop of methylene blue, which will help to visualize the electrode paste inside the electrodes and on the skin. Take the mixed paste in a syringe. Make sure there are no air bubbles in the syringe. Inject the gel into each of the 32 electrodes, filling them without introducing any air bubbles. It is recommended to inject from the bottom rather than the top. This provides better access to each electrode and reduces the possibility of the gel spilling over.</li>
		<li>Turn on the EEG and physiological recording system and open the corresponding recording software on the computer in use.</li>
	</ol>
	</li>
	<li>Animal preparation and anesthesia 
	NOTE: Chronic epilepsy was created using a protocol for focal cortical dysplasia (FCD)&#160;in Wistar rats. The EEG recordings were conducted in adult Wistar rats (8 weeks old, 300 - 400 g).
	<ol>
		<li>Record the rat&#39;s weight in an experiment sheet. Use this information to calculate the sedative dose (dexdomitor 0.25 mg/kg). Induce anesthesia in the rat with 5% isoflurane and 100% oxygen (1 L/min at 14.7 psi).</li>
		<li>After trimming the rat&#39;s head, reduce isoflurane to 2% and maintain it during the entire setting of the EEG mini-cap. Check rat reflexes are absent (toe-pinch).&#160;Place the rat on a heating pad in the stereotaxic apparatus by fixing the ear canals using ear bars. Make sure the anesthesia nose cone is secure.</li>
		<li>Apply lubricant ophthalmic ointment to each eye.</li>
		<li>Shave the extra hair on the rat&#39;s head and ears using a razor. Avoid any bleeding during shaving. 
		NOTE: Any hair left on the skin will produce noise in the EEG recordings. Rub the rat&#39;s skin with 90% isopropyl alcohol to stimulate the blood vessels and degrease the skin.</li>
		<li>Place a saline swab on the scalp and cover it completely to keep good skin conductance until the EEG mini-cap is ready to be placed.</li>
		<li>Connect temperature, respiration, and three lead electrocardiogram probes. Note that the temperature is measured by a rectal probe. Continuously monitor the physiology of the rat during the recording procedures. Ensure that the normal temperature is 37 &#176;C, respiration range is 30 - 60 breaths per minute, and heart rate is around 350 - 450 beats per minute.</li>
	</ol>
	</li>
	<li>Recording procedures
	<ol>
		<li>Remove the saline swab from the rat&#39;s scalp and place the prepared EEG mini-cap on its skin. Fix the mini-cap with rubber bands. Place one rubber band on the front side of the scalp, usually in front of the eyes, and another band at the back of the scalp between the ears and neck. Use a plastic protector under the neck to facilitate normal respiration.</li>
		<li>Put a layer of high-conductance electrode paste on both the ground and reference electrodes. Place them on the respective ear. 
		NOTE: The reference electrode can be possibly placed in other locations.</li>
		<li>Connect the EEG mini-cap to the amplifiers and observe a preview of the workbench for electrode impedance. Check the performance of all electrodes. For a high-quality recording, ensure that the impedance value is in the range of 5 - 30 k&#937;. If there are any noisy electrodes, provide better contact with the scalp by either moving them inside the scaffold toward the scalp or gently injecting more gel from the top of the electrode.</li>
		<li>Administer dexdomitor (0.25 mg/kg) intraperitoneally and immediately reduce the isoflurane rate to 0%. If the respiration rate is not within 30 - 60 breaths per minute range, start increasing the isoflurane rate gently. Do not exceed the value of 1% isoflurane. Monitor this step carefully because the mixture of isoflurane and dexdomitor could prompt the animals to a critical condition. 
		NOTE: In the preclinical model of focal epilepsy, isoflurane affects interictal epileptiform discharges (IEDs), whereas dexdomitor does not. Subjects under isoflurane have weaker epileptogenic properties,&#160;i.e.,&#160;relatively fewer IEDs can be detected compared to other conditions. The dexdomitor dose is effective for about 2 hrs. Thus, to save the time for its effect, the preparation was carried out under isoflurane.</li>
		<li>Conduct EEG recordings. After the recording, mark the positions of the three jutting circles of the EEG mini-cap on top of the skin by inserting a color pen inside them before the EEG mini-cap is removed. Use them as landmarks for MRI co-registration. Take a picture of the rat head with the landmarks. Place the rat back inside the cage and monitor it until complete recovery from dexdomitor&#39;s effect. 
		NOTE: In this experiment, red color (opponent color to green) was used to distinguish from the electrode positions (green). However, it is recommended to use other colors (purple/green) if small bleeding spots are observed in the skin.</li>
	</ol>

Take an anesthetized rat with focal epilepsy, characterized by abnormal brain electrical activity.
Secure the rat in a stereotaxic apparatus and administer anesthesia through a nose cone.&#160;
Completely remove the head fur to prevent signal interference during electroencephalogram, or EEG recording.
Clean the skin with alcohol and place saline swabs to enhance electrical conductance.
Apply EEG paste to each electrode of a multichannel EEG mini-cap to ensure optimal conductivity between the electrodes and the skin.
Fix the mini-cap on the scalp.
Position the ground and reference electrodes on the ears.
Connect the mini-cap to the amplifier. Visualize the EEG signals to identify loosely connected electrodes and improve their connectivity.
Administer a sedative and stop the anesthetic flow to minimize its interference with brain activity.
Record the EEG and identify the abnormal electrical activity events.
Mark the mini-cap&#39;s position on the skin for spatial correlation during subsequent brain imaging.

Non-restraining EEG Radiotelemetry: Epidural and Deep Intracerebral Stereotaxic EEG Electrode Placement

Implantable EEG radiotelemetry is of central relevance in the neurological characterization of transgenic mouse models of neuropsychiatric and neurodegenerative diseases as well as epilepsies. This powerful technique does not only provide valuable insights into the underlying pathophysiological mechanisms, i.e., the etiopathogenesis of CNS related diseases, it also facilitates the development of new translational, i.e., therapeutic approaches. Whereas competing techniques that make use of recorder systems used in jackets or tethered systems suffer from their unphysiological restraining to semi-restraining character, radiotelemetric EEG recordings overcome these disadvantages. Technically, implantable EEG radiotelemetry allows for precise and highly sensitive measurement of epidural and deep, intracerebral EEGs under various physiological and pathophysiological conditions. First, we present a detailed protocol of a straight forward, successful, quick and efficient technique for epidural (surface) EEG recordings resulting in high-quality electrocorticograms. Second, we demonstrate how to implant deep, intracerebral EEG electrodes, e.g., in the hippocampus (electrohippocampogram). For both approaches, a computerized 3D stereotaxic electrode implantation system is used. The radiofrequency transmitter itself is implanted into a subcutaneous pouch in both mice and rats. Special attention also has to be paid to pre-, peri- and postoperative treatment of the experimental animals. Preoperative preparation of mice and rats, suitable anesthesia as well as postoperative treatment and pain management are described in detail.

Non-restraining EEG radiotelemetry is a valuable methodological approach to record in vivo long-term electroencephalograms from freely moving rodents. This detailed protocol describes stereotaxic epidural and deep intracerebral electrode placement in different brain regions in order to obtain reliable recordings of CNS rhythmicity and CNS related behavioral stages.

Electrophysiological Assessment of Murine Atria with High-Resolution Optical Mapping

Recent genome-wide association studies targeting atrial fibrillation (AF) have indicated a strong association between the genotype and electrophysiological phenotype in the atria. That encourages us to utilize a genetically-engineered mouse model to elucidate the mechanism of AF. However, it is difficult to evaluate the electrophysiological properties in murine atria due to their small size. This protocol describes the electrophysiological evaluation of atria using an optical mapping system with a high temporal and spatial resolution in Langendorff perfused murine hearts. The optical mapping system is assembled with dual high-speed complementary metal oxide semiconductor cameras and high magnification objective lenses, to detect the fluorescence of a voltage-sensitive dye and Ca2+ indicator. To focus on the assessment of murine atria, optical mapping is performed with an area of 2 mm &#215; 2 mm or 10 mm x 10 mm, with a 100 &#215; 100 resolution (20 &#181;m/pixel or 100 &#181;m/pixel) and sampling rate of up to 10 kHz (0.1 ms) at maximum. A 1-French size quadripolar electrode pacing catheter is placed into the right atrium through the superior vena cava avoiding any mechanical damage to the atrium, and pacing stimulation is delivered through the catheter. An electrophysiological study is performed with programmed stimulation including constant pacing, burst pacing, and up to triple extrastimuli pacing. Under a spontaneous or pacing rhythm, the optical mapping recorded the action potential duration, activation map, conduction velocity, and Ca2+ transient individually in the right and left atria. In addition, the programmed stimulation also determines the inducibility of atrial tachyarrhythmias. Precise activation mapping is performed to identify the propagation of the excitation in the atrium during an induced atrial tachyarrhythmia. Optical mapping with a specialized setting enables a thorough electrophysiological evaluation of the atrium in murine pathological models.

This protocol describes the electrophysiological evaluation of murine atria utilizing an optical mapping system with a high temporal and spatial resolution, including dual recordings of the membrane voltage and Ca2+ transient under programmed stimulation through a specialized electrode catheter.

Concurrent EEG and Functional MRI Recording and Integration Analysis for Dynamic Cortical Activity Imaging

Electroencephalography (EEG) and functional magnetic resonance imaging (fMRI) are two of the fundamental noninvasive methods for identifying brain activity. Multimodal methods have sought to combine the high temporal resolution of EEG with the spatial precision of fMRI, but the complexity of this approach is currently in need of improvement. The protocol presented here describes the recently developed spatiotemporal fMRI-constrained EEG source imaging method, which seeks to rectify source biases and improve EEG-fMRI source localization through the dynamic recruitment of fMRI sub-regions. The process begins with the collection of multimodal data from concurrent EEG and fMRI scans, the generation of 3D cortical models, and independent EEG and fMRI processing. The processed fMRI activation maps are then split into multiple priors, according to their location and surrounding area. These are taken as priors in a two-level hierarchical Bayesian algorithm for EEG source localization. For each window of interest (defined by the operator), specific segments of the fMRI activation map will be identified as active to optimize a parameter known as model evidence. These will be used as soft constraints on the identified cortical activity, increasing the specificity of the multimodal imaging method by reducing cross-talk and avoiding erroneous activity in other conditionally active fMRI regions. The method generates cortical maps of activity and time-courses, which may be taken as final results, or used as a basis for further analyses (analyses of correlation, causation, etc.) While the method is somewhat limited by its modalities (it will not find EEG-invisible sources), it is broadly compatible with most major processing software, and is suitable for most neuroimaging studies.

An EEG-fMRI multimodal imaging method, known as the spatiotemporal fMRI-constrained EEG source imaging method, is described here. The presented method employs conditionally-active fMRI sub-maps, or priors, to guide EEG source localization in a manner that improves spatial specificity and limits erroneous results.
