Todos os camundongos foram obtidos comercialmente e mantidos em um ciclo claro/12 h escuro de 12 h (luz acesa às 08:00 horas locais) a uma temperatura ambiente de 22-25 °C e umidade relativa de 50%-60%. Os ratos tiveram acesso a um suprimento contínuo de comida e água. Todos os experimentos foram realizados de acordo com as Diretrizes para Cuidados e Uso de Animais de Laboratório da South China Normal University e aprovados pelo Comitê Institucional de Ética em Animais. Camundongos C57BL/6J machos com idade entre 3-5 meses de idade foram utilizados para os experimentos.
1. Montagem do sistema de micro-acionamento
<ol><li>Conecte duas placas projetadas por computador usando dois stulls e um parafuso que prende o micro-drive móvel e conecte o conector a uma placa (Figura 1A, Bi-iii). Acione o micro-drive torcendo um parafuso (0,5 mm/círculo).</li>
	<li>Certifique-se de que o micro-drive possa transportar dois conjuntos de oito tubos guia (~3 cm de comprimento, ~50 μm de diâmetro interno, ~125 μm de diâmetro externo) para cada lado da região MC e, em seguida, corte-o com o mesmo comprimento (pelo menos 15 mm; Figura 1Biv, v).</li>
	<li>Cortar 16 fios de cromo Ni (~5 cm de comprimento) com diâmetro de 35 μm e, em seguida, carregá-los sucessivamente nos tubos guia, seguido da aplicação da cola para fixá-los (Figura 1Bi, vi, vii).</li>
	<li>Retire o isolamento do fio, amarre sucessivamente cada fio exposto a cada pino do conector seguindo o mapa do canal, bem como os eletrodos de referência e terra, e então rebata lentamente a tinta condutora em cada pino (Figura 1Bviii-x).</li>
	<li>Cubra os pinos com resina epóxi (Figura 1Axi, xii) e, em seguida, realize o revestimento a ouro através de um testador de impedância para reduzir a impedância das pontas dos eletrodos para ~350 kOhm (Figura 1Bxiii, xiv). Defina os parâmetros do testador de impedância da seguinte forma: −10,08 μA de corrente contínua para 1 s com solução de revestimento de ouro, incluindo 5 mM PtCl4.</li>
	<li>Finalmente, mova o micro-drive para o topo torcendo um parafuso. Verifique o tamanho total do sistema de microacionamento modificado conforme mostrado na Figura 1A (aproximadamente 15 mm de comprimento, 10 mm de largura, 20 mm de altura, ~1 g de peso). Verifique as especificações detalhadas da placa projetada por computador e do componente móvel na Figura 1Ai, ii.</li>
</ol>2. Implantação de arranjo de eletrodos
<ol><li>Esterilize os kits de cirurgia, use luvas estéreis e coloque um jaleco branco estéril do médico antes do início da operação.</li>
	<li>Para controlar a dor, use injeção subcutânea (s.c.) de meloxicam injetável (5 mg/kg) para o camundongo em uma câmara de indução. Em seguida, anestesiar o camundongo por injeção intraperitoneal (i.p.) de pentobarbital (80 mg/kg) em câmara de indução18,19. Aplicar uma dose suplementar de pentobarbital (20 mg/kg/h) se o reflexo de pinça do dedo ainda estiver presente.</li>
	<li>Fixe o rato num aparelho estereotáxico e mantenha a sua temperatura rectal a 37 °C utilizando um controlador de temperatura.</li>
	<li>Aplique pomada ocular de tetraciclina em ambos os olhos do rato e troque as luvas estéreis novamente antes da cirurgia.</li>
	<li>Raspar a pele do camundongo e desinfetar o sítio cirúrgico com três rodadas alternadas de betadina e álcool usando um aplicador estéril com ponta de algodão em círculos concêntricos, começando no centro e movendo-se para fora (Figura 2i, ii). Faça uma pequena incisão na linha média (~15 mm) para expor seu crânio. Imediatamente, aplique a lidocaína a 1% localmente nos músculos do pescoço para alívio da dor. Em seguida, remova o tecido residual com tesoura e limpe o crânio com cotonetes revestidos com soro fisiológico (Figura 2iii).</li>
	<li>Com um microeletrodo de vidro preenchido com tinta, marque os locais desejados do CM bilateral para implantação (Figura 2iv, v). Com base em estudoprévio20, as localizações das CM bilaterais são as seguintes: 0,74 mm anterior ao bregma e 1,25 mm lateral à linha média.</li>
	<li>Implantar quatro parafusos de aço inoxidável (0,8 mm de diâmetro) para proteger o sistema de micro-acionamento e, em seguida, unir todos os parafusos com os eletrodos de referência e terra, seguido de cobertura com cimento dentário misto para formar paredes (Figura 2vi-xi).</li>
	<li>Faça cuidadosamente dois pequenos orifícios (~1,5mm2) com uma broca de crânio nos lados esquerdo e direito do crânio coordenado nas regiões do CM (Figura 2xii). Utilizar as coordenadas estereotáxicas do CM bilateral: 0,74 mm anterior ao bregma, 1,25 mm lateral à linha média e 0,5 mm ventral à dura-máter.</li>
	<li>Retire cuidadosamente a dura-máter dos orifícios com pinça fina (Figura 2xiii).</li>
	<li>Insira o sistema de micro-acionamento no centro dos orifícios usando um micromanipulador a 10 μm/s (Figura 2xiv-xvii).</li>
	<li>Preencher a vaselina nas paredes de cimento dental após finalizar a inserção do sistema de micro-acionamento (Figura 2xviii).</li>
	<li>Unir a placa inferior do sistema de micro-acionamento e as paredes de cimento dental com o cimento dental misto (Figura 2xix)</li>
	<li>Lavar a incisão com soro fisiológico seguido de tratamento local com gel contendo cloridrato de lincomicina e cloridrato de lidocaína para alívio da dor pós-cirúrgica.</li>
	<li>Enrole a fita condutora de folha de cobre ao redor do sistema de microacionamento implantado (Figura 2xx).</li>
	<li>Mova o rato para uma gaiola mantida a 31-33 °C e monitorize o rato para recuperação da anestesia.</li>
	<li>Permita que os ratos se recuperem por 1 semana com alimentação separada. Verificar e tratar a incisão com 3 dias de aplicação contínua de um gel contendo cloridrato de lincomicina e cloridrato de lidocaína.</li>
</ol>3. Gravação multicanal no CM bilateral em camundongos em movimento livre
<ol><li>Segure a cabeça de um rato acordado com leveza e cuidado. Desça as matrizes de eletrodos (~0,1 mm de profundidade) torcendo o parafuso na parte móvel do sistema de microacionamento (Figura 1Aii) com pelo menos 1 dia de antecedência.</li>
	<li>Segure a cabeça do rato acordado com leveza e cuidado. Conecte o centro do headstage e um balão de hélio (preenchido com aproximadamente 0,02 L de hélio) com uma rosca para compensar o peso do headstage e do sistema de micro-acionamento (Figura 3A, B).</li>
	<li>Capture sinais brutos usando os eletrodos de gravação e sistemas multicanal por amostragem a 30 kHz no software de gravação e, em seguida, digitalize usando um conversor analógico-digital (DA) dos sistemas multicanal.</li>
	<li>Extraia os sinais LFP dos dados brutos reamostrando a 10 kHz no software de gravação e, em seguida, use um filtro de entalhe do software de gravação para remover o ruído da linha de 50 Hz.</li>
	<li>Registre dados brutos em um estado estável a partir de um mouse em movimento livre por pelo menos 60 s. Depois de terminar a gravação, remova lentamente a conexão entre o headstage e o sistema de micro-drive e retorne o mouse de volta à sua gaiola inicial.</li>
	<li>Armazene os dados gravados no computador e analise-os offline (Figura 4 e Figura 5).</li>
	<li>Após o término do experimento, realizar a eutanásia de acordo com as orientações do instituto e, em seguida, confirmar a localização dos eletrodos usando a fonte de alimentação na saída de 3 V para realizar uma lesão eletrolítica de 1 min, seguido de análise histológica. Cortar o cérebro do camundongo em fatias de 30 μm usando um micrótomo de congelamento, coletar as seções MC e, em seguida, capturar as imagens com um microscópio (Figura 3C, D).</li>
</ol>4. Classificação e análise de spikes
<ol><li>Clique em Arquivo > Abrir arquivos > Nev no software de classificação de spike para abrir os dados de spike amostrados em 30 kHz (Figura 4Ai).</li>
	<li>Clique em Informações para selecionar o canal não classificado e, em seguida, selecione Classificar > Alterar Método de Classificação > Usar K-Means. Pressione o botão Valley-Seeking Sort > K-Means Sorting para obter as unidades classificadas (Figura 4Aii, iii).</li>
	<li>Clique em Arquivo > Salvar como, salve os dados de pico classificados com uma extensão de nome de arquivo .nev e selecione Arquivo > Exportar dados por forma de onda para exportar os resultados da PCA com uma extensão de nome de arquivo .txt (Figura 4Aiv).</li>
	<li>Clique em File > Import Data > Blackrock File no software para análise de dados neurofisiológicos para abrir o arquivo spike classificado (Figura 4Bi).</li>
	<li>Clique em Análise > Autocorrelogramas para obter o autocorrelograma para a unidade selecionada e, em seguida, defina os parâmetros da seguinte forma: o valor X Mínimo em −0,05 s, o valor X Máximo em 0,05 s e o valor Bin em 0,001 (Figura 4Bii, iii).</li>
	<li>Carregue os dados de spike classificados, clique em Analysis > Interspike Interval Histograms para obter o histograma de intervalo inter-spike e, em seguida, defina os parâmetros da seguinte maneira: o valor do intervalo mínimo em 0 s, o valor do intervalo máximo em 0,1 s e o valor do Bin em 0,001 (Figura 4Biv, v).</li>
	<li>Clique em Análise > Correlogramas Cruzados para obter o correlograma cruzado entre dois eventos unitários classificados e, em seguida, defina os eventos e parâmetros de referência da seguinte forma: o valor X Mínimo em −0,1 s, o valor X Máximo em 0,1 s e o valor Bin em 0,001 (Figura 4Bvi, vii).</li>
	<li>Clique em Resultados > Resultados Numéricos para salvar os resultados do autocorrelograma, histograma de intervalo entre espículas e correlação cruzada com .xls extensões de nome de arquivo (Figura 4Bviii, ix). Analise os dados e desenhe o gráfico.</li>
</ol>5. Análise LFP
<ol><li>Clique em File Import Data > Blackrock File no software para análise dos dados neurofisiológicos para abrir os dados de sinal contínuo amostrados a 10 kHz (Figura 5Ai).</li>
	<li>Clique em Analysis > Spectrum for Continuous para analisar o espectro de potência para o LFP a partir do canal selecionado. Defina os parâmetros da seguinte forma: o Número de Valores de Frequência em 8.192, o valor de Múltiplas Faixas em 3-5, a Normalização da porcentagem da densidade espectral de potência total (PSD) e a Faixa de Frequência de 1 Hz a 100 Hz (Figura 5Aii, iii).</li>
	<li>Clique em Análise > Coerência para Contínuo para analisar a coerência de dois LFPs dos lados esquerdo e direito do CM. Defina o canal de referência e os parâmetros da seguinte maneira: Calcule em valores de coerência, o número de valores de frequência em 8.192, o valor de Multiple Tapers em 3-5 e a faixa de frequência de 1 Hz a 100 Hz (Figura 5Aiv, v).</li>
	<li>Clique em Análise > Corr. com Variáveis Contínuas para analisar a correlação entre dois LFPs dos lados esquerdo e direito do MC. Defina o canal de referência (dados LFP) e os parâmetros da seguinte maneira: o valor X Mínimo em −0,1 s, o valor X Máximo em 0,1 s e o valor Bin em 0,001 (Figura 5Avi, vii).</li>
	<li>Clique em Resultados > Resultados Numéricos para salvar os resultados do PSD, coerência e correlação com uma extensão de nome de arquivo .xls (Figura 5Aviii, ix).</li>
	<li>Selecione o canal para o qual os traços representativos precisam ser extraídos para cada faixa de frequência, clique em Editar > Filtragem Digital de Variáveis Contínuas para obter cada faixa de frequência e, em seguida, defina os parâmetros da seguinte forma: a Resposta de Frequência de Filtro como Passa-Banda, a Implementação de Filtro em Resposta de Impulso Infinito (IIR) Butterworth e o valor de Ordem de Filtro em 2. Por fim, defina a faixa de frequência de interesse (Figura 5Bi-iv). NOTA: As faixas de frequência utilizadas aqui foram: oscilações delta (δ, 1-4 Hz), (θ, 5-12 Hz), beta (β, 13-30 Hz), gama baixa (baixa γ, 30-70 Hz) e gama alta (alta γ, 70-100 Hz).</li>
	<li>Analise os dados e desenhe o gráfico.</li>
</ol>6. Correlações entre a espícula e a LFP
<ol><li>Clique em Arquivo > Importar Dados > Arquivo Blackrock no software para a análise de dados neurofisiológicos para abrir os dados de sinal contínuo e dados de pico.</li>
	<li>Clique em Análise > Análise de Coerência para analisar a coerência entre os picos e LFP do canal selecionado. Defina a variável de referência (tempo de pico) e os parâmetros da seguinte forma: Calcule em Valores de Coerência, o Número de Valores de Frequência em 512, o valor de Múltiplas Tapers em 3-5 e a faixa de frequência de 1 Hz a 100 Hz (Figura 5Ci, ii).</li>
	<li>Clique em Resultados > Resultados Numéricos para salvar o resultado da coerência do campo spike com uma extensão de nome de arquivo .xls (Figura 5Ciii, iv).</li>
	<li>Analise os dados e desenhe o gráfico.</li>

O potencial de campo local (LFP), um importante componente dos sinais extracelulares, reflete a atividade sináptica de grandes populações de neurônios, que formam o código neural para múltiploscomportamentos1. Considera-se que as espículas geradas pela atividade neuronal contribuem para a LFP e são importantes para a codificação neural2. Alterações em spikes e LFPs comprovadamente mediam diversas doenças cerebrais, como a doença de Alzheimer, bem como emoções como medo, etc.3,4. Vale ressaltar que muitos estudos têm destacado que a atividade da espícula difere significativamente entre os estados acordado e anestesiado em animais5. Embora os registros em animais anestesiados ofereçam uma oportunidade de avaliar PBL com artefatos mínimos em estados de sincronização cortical altamente definidos, os resultados diferem em certa medida do que pode ser encontrado em indivíduos acordados 6,7,8. Assim, é mais significativo detectar atividade neural em longas escalas de tempo e grandes escalas espaciais em várias doenças em estado de vigília usando eletrodos implantados no cérebro. Este manuscrito fornece informações para iniciantes sobre como fazer o sistema de micro-drive e definir os parâmetros usando um software comum para calcular os sinais de pico e LFP de forma rápida e direta, a fim de iniciar a gravação e análise.
Embora o registro não invasivo de funções cerebrais, como o uso de eletroencefalogramas (EEGs) e potenciais relacionados a eventos (ERPs) registrados a partir do couro cabeludo, tenha sido amplamente utilizado em estudos com humanos e roedores, os dados de EEG e ERP têm baixas propriedades espaciais e temporais e, portanto, não conseguem detectar os sinais precisos produzidos pela atividade sináptica dendrítica próxima dentro deuma área específica do cérebro1. Atualmente, aproveitando o registro multicanal em animais conscientes, a atividade neural nas camadas mais profundas do cérebro pode ser registrada crônica e progressivamente pela implantação de um sistema de micro-drive no cérebro de primatas ou roedores durante múltiplos testes comportamentais 1,2,3,4,5,6,7,8,9 . Resumidamente, os pesquisadores podem construir um sistema de micro-drive que pode ser usado para o posicionamento independente dos eletrodos ou tetrodos para atingir diferentes partes do cérebro10,11. Por exemplo, Chang e col. descreveram técnicas para registrar spikes e LFPs em camundongos montando um micro-drive leve e compacto12. Além disso, sondas de silício microusinadas com componentes acessórios feitos sob medida estão comercialmente disponíveis para o registro de múltiplos neurônios individuais e LFPs em roedores durante tarefas comportamentais13. O protocolo visa desvendar as propriedades do disparo neuronal e dos potenciais de campo local (LFPs) em camundongos comportados realizando tarefas específicas, correlacionando os sinais eletrofisiológicos com o comportamento espontâneo e/ou específico. Esta técnica representa uma ferramenta valiosa no estudo da atividade da rede neuronal subjacente a esses comportamentos. O artigo fornece um procedimento detalhado e completo para o implante de eletrodos e consequente registro extracelular em camundongos conscientes em movimento livre. O estudo inclui um método detalhado para implantar os arranjos de microeletrodos, capturar os sinais de LFP e spiking neuronal no córtex motor (CM) usando um sistema multicanal, e a subsequente análise de dados off-line. A vantagem do registro multicanal em animais conscientes é que um maior número de neurônios de spiking e subtipos neuronais pode ser obtido e comparado, o que permite avaliar a relação entre um comportamento específico e os sinais eletrofisiológicos associados. Notavelmente, a técnica de registro extracelular multicanal e o procedimento de análise de dados descrito no presente estudo podem ser aplicados a outras áreas cerebrais ao realizar experimentos em camundongos com comportamento.

The local field potential (LFP), an important component of extracellular signals, reflects the synaptic activity of large populations of neurons, which form the neural code for multiple behaviors1. Spikes generated by neuronal activity are considered to contribute to the LFP and are important for neural coding2. Alterations in spikes and LFPs have been proven to mediate several brain diseases, such as Alzheimer&#39;s disease, as well as emotions such as fear, etc.3,4. It is worth noting that many studies have highlighted that spike activity significantly differs between awake and anesthetized states in animals5. Although recordings in anesthetized animals offer an opportunity to assess LFPs with minimal artifacts in highly defined cortical synchronization states, the results differ to some extent from what can be found in awake subjects6,7,8. Thus, it is more meaningful to detect neural activity over long time scales and large spatial scales in various diseases in an awake brain state using electrodes implanted in the brain. This manuscript provides information for beginners on how to make the micro-drive system and set the parameters using common software for computing the spike and LFP signals in a fast and straightforward way in order to get the recording and analysis started.
Although the non-invasive recording of brain functions, such as by using electroencephalograms (EEGs) and event-related potentials (ERPs) recorded from the scalp, has been used extensively in human and rodent studies, EEG and ERP data have low spatial and temporal properties and, thus, cannot detect the precise signals produced by nearby dendritic synaptic activity within a specific brain area1. Currently, by taking advantage of multichannel recording in conscious animals, neural activity in the deeper layers of the brain can be recorded chronically and progressively by implanting a micro-drive system into the brains of primates or rodents during multiple behavioral tests1,2,3,4,5,6,7,8,9. Briefly, researchers can construct a micro-drive system that can be used for the independent positioning of the electrodes or tetrodes to target different parts of the brain10,11. For example, Chang et al. described techniques to record spikes and LFPs in mice by assembling a light and compact micro-drive12. In addition, micro-machined silicon probes with custom-made accessory components are commercially available for recording multiple single neurons and LFPs in rodents during behavioral tasks13. Although various designs have been used for assembling micro-drive systems, these still have limited success in terms of the complexity and weight of the whole micro-drive system. For example, Lansink et al. showed a multichannel micro-drive system with a complex structure for recording from a single brain region14. Sato et al. reported a multichannel micro-drive system displaying an automatic hydraulic positioning function15. The main disadvantages of these micro-drive systems are that they are too heavy for mice to move freely and are difficult to assemble for beginners. Although multichannel extracellular recording has been shown to be a suitable and efficient technology for measuring neural activity during behavioral tests, it is not easy for beginners to record and analyze the signals acquired by the complex micro-drive system. Given that it is difficult to get the entire operation process of the multichannel extracellular recording and data analysis started in freely moving mice16,17, this present article presents simplified guidelines to introduce the detailed process of making the micro-drive system using commonly available components and settings; the parameters in the common software for computing the spike and LFP signals in a fast and straightforward way are also provided. Additionally, in this protocol, the mouse can move freely due to the use of a helium balloon, which contributes to offsetting the weight of headstage and micro-drive system. Generally, in the present study, we describe how to easily build a micro-drive system and optimize the processes of recording and data analysis.

We aim to uncover the properties of neuronal firing and network local field potentials in behaving mice carrying out specific tasks by correlating the electrophysiological signal with behaviors. Multichannel extracellular recording by the micro-drive system has been shown to be a suitable and a efficient technology for neural activity during behavioral tests. Multichannel recording in free-moving mice has been deemed to be a useful technology in neuroscience studies, but it's still quite challenging for beginners to acquire and analyze the signals.We introduced how to perform the multichannel extracellular recording in freely-moving mice with a more steady and lightweight micro-drive system, and optimize the processes of recording and data analysis for the beginners. We are going to increase the number of channels and reduce the volume of micro-driver system in a new version.

A gravação multicanal em camundongos em movimento livre tem sido considerada uma tecnologia útil em estudos de neurociência, mas ainda é bastante desafiadora para iniciantes adquirir e analisar os sinais.Apresentamos como realizar a gravação extracelular multicanal em mouses em movimento livre com um sistema de micro-drive mais estável e leve, e otimizar os processos de gravação e análise de dados para os iniciantes. Vamos aumentar o número de canais e reduzir o volume do sistema de micro-driver em uma nova versão.

The protocol aims to uncover the properties of neuronal firing and network local field potentials (LFPs) in behaving mice carrying out specific tasks by correlating the electrophysiological signals with spontaneous and/or specific behavior. This technique represents a valuable tool in studying the neuronal network activity underlying these behaviors. The article provides a detailed and complete procedure for electrode implantation and consequent extracellular recording in free-moving conscious mice. The study includes a detailed method for implanting the microelectrode arrays, capturing the LFP and neuronal spiking signals in the motor cortex (MC) using a multichannel system, and the subsequent offline data analysis. The advantage of multichannel recording in conscious animals is that a greater number of spiking neurons and neuronal subtypes can be obtained and compared, which allows the evaluation of the relationship between a specific behavior and the associated electrophysiological signals. Notably, the multichannel extracellular recording technique and the data analysis procedure described in the present study can be applied to other brain areas when conducting experiments in behaving mice.

The protocol describes the methodology of extracellular recording in the motor cortex (MC) to reveal extracellular electrophysiological properties in freely moving conscious mice, as well as the data analysis of local field potentials (LFPs) and spikes, which is useful for evaluating the network neural activity underlying behaviors of interest.

Micro-Drive System Assembly and Electrode Array Implantation for Multichannel Recording in Mice

Begin by assembling the Microdrive system. Connect two computer-designed boards using two stulls and a screw that holds the movable Microdrive, and attach the connector to one board. Ensure the Microdrive can carry two sets of eight guide tubes for each side of the motor cortex or MC region.Cut the guide tubes to the same length. Cut 16 nichrome wires, each measuring approximately five centimeters long and 35 micrometers in diameter. Load the wires into the guide tubes and apply glue to fix them.Strip the wire insulation and twine each exposed wire to each pin from the connector following the channel map and the reference and ground electrodes, then slowly coat conducting paint onto each pin. Cover the pins using epoxy resin, then perform gold plating via an impedance tester to reduce the impedance of the electrode tips to approximately 350 kiloohms. For electrode array implantation, fix the anesthetized mouse in a stereotaxic apparatus and use a temperature controller to maintain its rectal temperature at 37 degrees Celsius.Then make a small midline incision to expose the skull. Remove the residual tissue using scissors and clean the skull with sterile saline-soaked cotton buds. Using a glass microelectrode filled with ink, mark the desired locations of the bilateral MC for implantation.Next, using a skull drill, carefully drill two small holes on both the left and right sides of the coordinated skull in the MC regions. Gently remove the dura mater from the holes with fine forceps. Then insert the Microdrive system into the center of the holes, using a micromanipulator at 10 micrometers per second.After finishing the insertion, fill the petroleum jelly into the dental cement walls and join the bottom plate of the Microdrive system and the dental cement walls with the mixed dental cement.

Multichannel Recording in the Bilateral MC in Free-Moving Mice and Neurophysiological Data Analysis

For the multi-channel recording, move down the electrode arrays by twisting the screw on the movable part of the micro-drive system one day in advance. Hold the head of an awake mouse lightly and carefully. Then, link the center of the head stage and a helium balloon with a thread to offset the weight of the head stage and the micro-drive system.Capture raw signals using the recording electrodes and multi-channel systems by sampling at 30 kilohertz in the recording software. Then digitize using a digital analog converter from the multi-channel systems. Extract the local field potential, or LFP signals, from the raw data by re-sampling at 10 kilohertz.Then, using a notch filter, remove the 50-hertz line noise. For spike sorting and analysis, in the spike sorting software, click on File, then Open, and nev files to open the spike data sampled at 30 kilohertz. Click on Info to select the unsorted channel.Then choose Sort, Change Sort Method, and Use K-Means. Press the button Valley Seeking Sort, then K-Means Sorting to obtain the sorted units. Next, in the neurophysiological data analysis software, open the sorted spike file by clicking on File, Import Data, and BlackRock File.To obtain the autocorrelogram for the selected unit, click on Analysis, then Autocorrelograms, and set the parameters. Load the sorted spike data. Then, click on Analysis, and Interspike Interval Histograms to obtain the interspike interval histogram, and then set the desired parameters.Click on Analysis, then Crosscorrelograms to obtain the crosscorrelogram between two sorted unit events. And then set the reference events and parameters. For LFP analysis, click on File, Import Data, and BlackRock File, to open the continuous signal data sampled at 10 kilohertz.Then, by clicking Analysis and Spectrum for Continuous, analyze the power spectrum for the LFP from the selected channel. Next, click on Analysis, then Coherence for Continuous to analyze the coherence for two LFPs from the left and right sides of the MC.Then, click on Analysis, followed by Correlation with Continuous Variables to analyze the correlation between two LFPs from the left and right sides of the MC.Once done, click on Results, then Numerical Results to save the Power Spectrum Density, Coherence, and Correlation with a xls file name extension. To analyze correlations between the spike and LFP, click on File, then Import Data and BlackRock File to open the continuous signal data and spike data.Then, click on Analysis and Coherence Analysis to analyze the coherence between the spikes and LFP from the selected channel. Finally, click on Results, then Numerical Results to save the results of the spike field coherence with a xls file name extension. The valley width and waveform duration of the units in the MC of the mouse showed that both the valley width and waveform duration of the MC putative pyramidal neurons in mice are higher than those of the putative interneurons.The crosscorrelogram between putative pyramidal neurons and interneurons indicated that the putative pyramidal neurons spiking occurs before the putative interneurons with a window of 18 milliseconds. In the LFP analysis, the LFPs of the left and right MC in normal mice were similar in the power spectrum, suggesting synchronized activities between the left and right MC.Further, the coherence and correlation between the left and right MC were calculated. The curve of spike field coherence in the MC of a normal mouse showed a stronger low gamma coherence for the putative interneurons compared to the pyramidal neurons.
