Stimulus-triggered averaging (StTA) of electromyographic (EMG) activity from 24 simultaneously recorded forelimb muscles was used to investigate properties of primary motor cortex (M1) output in the macaque monkey. Two monkeys were trained to perform a reach-to-grasp task requiring multijoint coordination of the forelimb. EMG activity was recorded from 24 forelimb muscles including 5 shoulder, 7 elbow, 5 wrist, 5 digit, and 2 intrinsic hand muscles. Microstimulation (15 microA at 15 Hz) was delivered throughout the movement task. From 297 stimulation sites in M1, a total of 2,079 poststimulus effects (PStE) were obtained including 1,398 poststimulus facilitation (PStF) effects and 681 poststimulus suppression (PStS) effects. Of the PStF effects, 60% were in distal and 40% in proximal muscles; 43% were of extensors and 47% flexors. For PStS, the corresponding numbers were 55 and 45% and 36 and 55%, respectively. M1 output effects showed extensive cofacilitation of proximal and distal muscles (96 sites, 42%) including 47 sites that facilitated at least one shoulder, elbow, and distal muscle, 45 sites that facilitated an elbow muscle and a distal muscle, and 22 sites that facilitated at least one muscle at all joints. The muscle synergies represented by outputs from these sites may serve an important role in the production of coordinated, multijoint movements. M1 output effects showed many similarities with red nucleus output although red nucleus effects were generally weaker and showed a strong bias toward facilitation of extensor muscles and a greater tendency to facilitate synergies involving muscles at noncontiguous joints.

The goal of this study was to assess the motor output capabilities of the forelimb representation of the supplementary motor area (SMA) in terms of the sign, latency and strength of effects on electromyographic (EMG) activity. Stimulus triggered averages of EMG activity from 24 muscles of the forelimb were computed in SMA during a reach-to-grasp task. Poststimulus facilitation (PStF) from SMA had two distinct peaks (15.2 and 55.2 ms) and one poststimulus suppression (PStS) peak (32.4 ms). The short onset latency PStF and PStS of SMA were 5.5 and 16.8 ms longer than those of the primary motor cortex (M1). The average magnitudes (peak increase or decrease above baseline) of the short and long latency PStF and PStS from SMA at 60 microA were 13.8, 11.3 and -11.9% respectively. In comparison, M1 PStF and PStS magnitudes at 15 microA were 50.2 and -23.8%. Extrapolating M1 PStF magnitude to 60 microA yields a mean effect that is nearly 15 times greater than the mean PStF from SMA. Moreover, unlike M1, the facilitation of distal muscles from SMA was not significantly greater than the facilitation of proximal muscles. We conclude that the output from SMA to motoneurons is markedly weaker compared with M1 raising doubts about the role of SMA corticospinal neurons in the direct control of muscle activity.

El objetivo de este estudio fue evaluar las capacidades de salida del motor de la representaci&oacute;n del miembro anterior de la &aacute;rea de motor suplementaria (SMA) en t&eacute;rminos de la muestra, la latencia y la fuerza de los efectos sobre la actividad electromiogr&aacute;fica (EMG). Promedios de est&iacute;mulo activada de la actividad EMG de 24 m&uacute;sculos de la extremidad delantera se calcularon en SMA durante una tarea de alcance de entender. Facilitaci&oacute;n poststimulus (PStF) de SMA tuvo dos picos distintos (15,2 y 55.2 ms) y una supresi&oacute;n poststimulus (PST) pico (32,4 ms). La latencia de inicio corto PStF y PST de SMA fueron ms 5.5 y 16,8 m&aacute;s largas que las de la corteza primaria del motor (M1). Las magnitudes promedio (m&aacute;ximo aumento o disminuci&oacute;n por encima de la l&iacute;nea de base) de la latencia corto y largo PStF y PST de SMA en 60 microA fueron 13,8, 11.3 y-11.9% respectivamente. En comparaci&oacute;n, magnitudes M1 PStF y PST en 15 microA fueron 50.2 y-23.8%. Extrapolando la magnitud M1 PStF a 60 microA produce un efecto medio casi 15 veces mayor que la media PStF de SMA. Adem&aacute;s, a diferencia de M1, la facilitaci&oacute;n de m&uacute;sculos distales de SMA no era significativamente mayor que la facilitaci&oacute;n de los m&uacute;sculos proximales. Stimulus-triggered averaging (StTA) of electromyographic (EMG) activity is a form of intracortical microstimulation that enables documentation in awake animals of the sign, magnitude, latency, and distribution of output effects from cortical and brainstem areas to motoneurons of different muscles. In this study, we show that the properties of effects in StTAs are stable and mostly independent of task conditions. StTAs of EMG activity from 24 forelimb muscles were collected from two male rhesus monkeys while they performed three tasks: (1) an isometric step tracking wrist task, (2) an isometric whole-arm push-pull task, and (3) a reach-to-grasp task. Layer V sites in primary motor cortex were identified and microstimuli were applied (15 muA) at a low rate (15 Hz). Our results show that the sign of effects (facilitation or suppression) in StTAs of EMG activity are remarkably stable in the presence of joint angle position changes (96% stable), whole-arm posture changes (97% stable), and across fundamentally different types of tasks such as arm push-pull versus reach-to-grasp (81% stable). Furthermore, comparing effects across different phases of a task also yielded remarkable stability (range, 84-96%). At different shoulder, elbow, and wrist angles, the magnitudes of effects in individual muscles were highly correlated. Our results demonstrate that M1 output effects obtained with StTA of EMG activity are highly stable across widely varying joint angles and motor tasks. This study further validates the use of StTA for mapping and other studies of cortical motor output.

Stability of output effects from motor cortex to forelimb muscles in primates.

Stimulation d&eacute;clench&eacute;e en moyenne (StTA) de l&#39;activit&eacute; &eacute;lectromyographique (EMG) est une forme de microstimulation intracortical qui permet la documentation chez des animaux &eacute;veill&eacute;s du signe, grandeur, latence et r&eacute;partition des effets de la sortie de la corticale et les r&eacute;gions du tronc c&eacute;r&eacute;bral pour les motoneurones des muscles diff&eacute;rents. Dans cette &eacute;tude, nous montrons que les propri&eacute;t&eacute;s des effets en StTAs sont stables et surtout ind&eacute;pendante des conditions de travail. L&#39;activit&eacute; StTAs de l&#39;EMG de 24 muscles des membres ant&eacute;rieurs ont &eacute;t&eacute; pr&eacute;lev&eacute;s dans deux des singes rh&eacute;sus m&acirc;les alors qu&#39;ils ont ex&eacute;cut&eacute; trois t&acirc;ches: (1) une &eacute;tape isom&eacute;trique suivi de t&acirc;che de poignet, (2) une t&acirc;che de bras entiers isom&eacute;trique push pull et (3) une t&acirc;che port&eacute;e &agrave; comprendre. Sites de la couche V dans le cortex moteur primaire ont &eacute;t&eacute; identifi&eacute;s et microstimuli ont &eacute;t&eacute; appliqu&eacute;s (15 muA) &agrave; un taux faible (15 Hz). Reiz ausgel&ouml;ste durchschnittlich (StTA) elektromyographische (EMG)-Aktivit&auml;t ist eine Form der intrakortikale Microstimulation, die Dokumentation in wach Tiere des Zeichens, Helligkeit, Latenz und Verteilung der Ausgabe Effekte aus kortikalen erm&ouml;glicht und Hirnstamm Bereiche Motoneurons der verschiedenen Muskeln. In dieser Studie zeigen wir, dass die Eigenschaften der Effekte in StTAs stabil und vor allem unabh&auml;ngig von Aufgabenbedingungen. StTAs der EMG-Aktivit&auml;t von 24 Vorderbein Muskeln wurden von zwei m&auml;nnlichen Rhesusaffen gesammelt, w&auml;hrend sie drei Aufgaben: (1) eine isometrische Schritt verfolgen Handgelenk Aufgabe, (2) eine isometrische Push-Pull-ganz-Arm-Aufgabe und (3) eine verst&auml;ndliche Reichweite-Aufgabe. Bei verschiedenen Schulter, Ellenbogen und Handgelenk Winkel wurden die Gr&ouml;&szlig;enordnungen von Effekten in einzelnen Muskeln hoch korreliert. Sono stati individuati siti strato V nella corteccia motoria primaria e microstimuli sono stati applicati (15 muA) a bassa quota (15 Hz). I nostri risultati mostrano che il segno di effetti (facilitazione o soppressione) in attivit&agrave; StTAs di EMG &egrave; notevolmente stabili in presenza di cambiamenti di posizione congiunta angolo (96% stabile), cambiamenti di postura intero braccio (97% stabile) e attraverso fondamentalmente diversi tipi di attivit&agrave; come braccio push-pull versus portata da afferrare (81% stabile). Inoltre, confrontando gli effetti attraverso diverse fasi di un compito reso anche notevole stabilit&agrave; (gamma, 84-96%). Presso diverso spalla, gomito e polso angoli, la grandezza degli effetti dei singoli muscoli era altamente correlati. I nostri risultati dimostrano che M1 uscita effetti ottenuti con attivit&agrave; StTA di EMG sono altamente stabili attraverso ampiamente variando angoli congiunti e compiti motori. Questo studio ulteriore convalida l&#39;uso di StTA per mappatura e altri studi di uscita motore corticale.

Although the arrangement of the corticospinal projection in primates is consistent with a more prominent role of the ipsilateral motor cortex on proximal muscles, rather than on distal muscles involved in manual dexterity, the role played by the primary motor cortex on the control of manual dexterity for the ipsilateral hand remains a matter a debate, either in the normal function or after a lesion. We, therefore, tested the impact of permanent unilateral motor cortex lesion on the manual dexterity of the ipsilateral hand in 11 macaque monkeys, within a time window of 60Â days post-lesion. For comparison, unilateral reversible pharmacological inactivation of the motor cortex was produced in an additional monkey. Manual dexterity was assessed quantitatively based on three motor parameters derived from two reach and grasp manual tasks. In contrast to the expected dramatic, complete deficit of manual dexterity of the contralesional hand that persists for several weeks, the impact on the manual dexterity of the ipsilesional hand was generally moderate (but statistically significant) and, when present, lasted less than 20Â days. Out of the 11 monkeys, only 3 showed a deficit of the ipsilesional hand for 2 of the 3 motor parameters, and 4 animals had a deficit for only one motor parameter. Four monkeys did not show any deficit. The reversible inactivation experiment yielded results consistent with the permanent lesion data. In conclusion, the primary motor cortex exerts a modest role on ipsilateral manual dexterity, most likely in the form of indirect hand postural control.

High-frequency repetitive microstimulation has been widely used as a method of investigating the properties of cortical motor output. Despite its widespread use, few studies have investigated how activity evoked by high-frequency stimulation may interact with the existing activity of cortical cells resulting from natural synaptic inputs. A reasonable assumption might be that the stimulus-evoked activity sums with the existing natural activity. However, another possibility is that the stimulus-evoked firing of cortical neurons might block and replace the natural activity. We refer to this latter possibility as "neural hijacking." Evidence from analysis of EMG activity evoked by repetitive microstimulation (200 Hz, 500 ms) of primary motor cortex in two rhesus monkeys during performance of a reach-to-grasp task strongly supports the neural hijacking hypothesis.

The nonhuman primate model is suitable to study mechanisms of functional recovery following lesion of the cerebral cortex (motor cortex), on which therapeutic strategies can be tested. To interpret behavioral data (time course and extent of functional recovery), it is crucial to monitor the properties of the experimental cortical lesion, induced by infusion of the excitotoxin ibotenic acid. In two adult macaque monkeys, ibotenic acid infusions produced a restricted, permanent lesion of the motor cortex. In one monkey, the lesion was monitored over 3.5 weeks, combining laser speckle imaging (LSI) as metabolic readout (cerebral blood flow) and anatomical assessment with magnetic resonance imaging (T2-weighted MRI). The cerebral blood flow, measured online during subsequent injections of the ibotenic acid in the motor cortex, exhibited a dramatic increase, still present after one week, in parallel to a MRI hypersignal. After 3.5 weeks, the cerebral blood flow was strongly reduced (below reference level) and the hypersignal disappeared from the MRI scan, although the lesion was permanent as histologically assessed post-mortem. The MRI data were similar in the second monkey. Our experiments suggest that LSI and MRI, although they reflect different features, vary in parallel during a few weeks following an excitotoxic cortical lesion.

El modelo de primates no humanos es adecuado para estudiar los mecanismos de recuperaci&#xF3;n funcional tras la lesi&#xF3;n de la corteza cerebral (corteza motora), en el que las estrategias terap&#xE9;uticas se pueden probar. Para interpretar los datos de comportamiento (evoluci&#xF3;n en el tiempo y el grado de recuperaci&#xF3;n funcional), es crucial para controlar las propiedades de la lesi&#xF3;n cortical experimental, inducida por la infusi&#xF3;n de &#xE1;cido ibot&#xE9;nico excitotoxina. En dos monos macacos adultos, las infusiones de &#xE1;cido ibot&#xE9;nico produce una lesi&#xF3;n restringida y permanente de la corteza motora. En uno de los monos, la lesi&#xF3;n fue supervisado m&#xE1;s de 3,5 semanas, la combinaci&#xF3;n de l&#xE1;ser de speckle (LSI) como lectura metab&#xF3;lica (el flujo sangu&#xED;neo cerebral) y la evaluaci&#xF3;n anat&#xF3;mica con resonancia magn&#xE9;tica (RM ponderada en T2). El flujo sangu&#xED;neo cerebral, medida en l&#xED;nea durante las siguientes inyecciones de &#xE1;cido ibot&#xE9;nico en la corteza motora, mostraron un aumento dram&#xE1;tico, todav&#xED;a est&#xE1; presente despu&#xE9;s de una semana, en paralelo a una hiperse&#xF1;al de resonancia magn&#xE9;tica. Despu&#xE9;s de 3,5 semanas, el flujo sangu&#xED;neo cerebral se redujo considerablemente (por debajo del nivel de referencia) y la hiperse&#xF1;al desaparecido de la resonancia magn&#xE9;tica, aunque la lesi&#xF3;n fue evaluada histol&#xF3;gicamente permanente como post-mortem. Los datos de resonancia magn&#xE9;tica fueron similares en el segundo mono.
