an orthotopic bladder tumor model and the evaluation of intravesical sarna treatment

نحن نقدم طريقة جديدة لعلاج سرطان المثانة مع تسليم intravesically RNA تفعيل صغيرة (سارنا) في الفأر طعم أجنبي orthotopic نموذج ورم المثانة. يتم تأسيس نموذج الفأر بواسطة قسطرة الإحليل تحت التخدير العام بالاستنشاق. ومن ثم قدم حرق كيميائي على الغشاء المخاطي المثانة داخل المثانة باستخدام محلول نترات الفضة لعرقلة طبقة جلايكان المثانة وتمكن خلايا لنعلق. بعد عدة يغسل بماء معقم، الإنسان سرطان المثانة KU-7-luc2-GFP الخلايا تغرس من خلال قسطرة في المثانة ليسكن لمدة 2 ساعة. ويؤكد نمو لاحق من أورام المثانة ورصدها بواسطة الموجات فوق الصوتية في الجسم الحي المثانة والتصوير bioluminescent. ثم يتم التعامل مع الأورام intravesically مع سارنا صيغت في جزيئات الدهون (LNPs). ويتم رصد نمو الورم مع الموجات فوق الصوتية والتألق الحيوي. وأظهرت جميع خطوات من هذا الإجراء في شريط الفيديو المرافق له.
