an orthotopic bladder tumor model and the evaluation of intravesical sarna treatment

We presenteren een nieuwe methode voor de behandeling van blaaskanker met intravesicaal afgeleverd kleine activerende RNA (Särna) in een orthotope xenograft muis blaas tumor model. De muis model wordt vastgesteld door urethrale catheterisatie onder inhalatie-narcose. Chemische brandwonden wordt vervolgens naar de blaas slijmvlies met behulp van intravesicale zilvernitraatoplossing om de blaas te glycosaminoglycaan laag verstoren en maakt het mogelijk cellen te hechten aan. Na enkele wasbeurten met steriel water, menselijke blaaskanker KU-7-luc2-GFP cellen worden bijgebracht door de katheter in de blaas te wonen gedurende 2 uur. Na de groei van blaastumoren wordt bevestigd en gecontroleerd door in vivo blaas echografie en bioluminescente beeldvorming. De tumoren worden dan intravesicaal behandeld met Särna geformuleerd in lipide nanodeeltjes (LNPs). Groei van de tumor wordt gecontroleerd met echografie en bioluminescentie. Alle stappen van deze procedure worden aangetoond in de bijgevoegde video.
