在某些情况下，特别是腹肌整形，由于手术领域的局限性，不可能获得真正的无菌样品。如果无法使用无菌容器将组织从手术室运送到层流罩，则去除组织外部 1 厘米通常足以防止微生物污染。如果腹肌成形性样品附着在皮肤上，可以使用70%的乙醇擦拭皮肤，在切碎脂肪组织之前对表面进行消毒。
在切碎过程中，将组织切成小块，切成细碎，这一点至关重要。如果外植量过大，ASC 的表面面积将不足以从组织迁移到板中。此外，至关重要的是，组织不要留在板中任何时间超过必要的，以免组织成为坏死。
