HBcAg 1차 항체 및 DAPI를 가진 배양은 형광 반전 현미경에 10X 목표로 관찰될 때 뚜렷한 염색귀 귀착되었습니다.핵국소화는 DAPI와 NTCP-EGFP 발현을 녹색 형광으로 확인함으로써 확인되었다. HBcAg의 발현 부위는 적색 형광으로 위치했다. DAPI, NTCP 및 HBcAg가 동일한 세포에 있을 때, 세포는 HBV에 성공적으로 감염되었습니다.사이클로스포린 A 군은 HBV가 NTCP를 차단하여 세포에 유입되는 것을 막기 때문에 음의 대조군이었다. HBcAg는 293T-NE-3NR에서 검출되었지만 NTCP를 나타내는 293T-NE 세포에서는 293T에서 HBV 감염에 필수적인 유일한 요인이 아닙니다. 좋은 세포 상태와 효율적인 작동은 성공적인 감염 결과를 얻기위한 핵심 포인트입니다.감염 후 부드러운 세척 및 미디어 변경도 중요합니다. 대안으로, HEpG2-NE 세포를 이용한 HBV 감염 방법은 이 방법보다 감염 효율이 높고 안티 HBV 약물 스크리닝에 적합하다. 293T-NE-3NR에서 HBV 감염을 모델링하는 것은 이들 세포의 비간 배경으로 인한 전통적인 세포 모델에 유용한 칭찬이다.이 방법은 비 간 세포에 있는 HBV 감염의 연구 결과 뿐만 아니라 HBV의 간을 위해 요구되는 호스트 요인의 연구를 촉진할 것입니다.
